The role of Melanocortin-1 Receptor(MC1R) in driving pigmentation is widely recognised. Recent studies however,have identified its involvement even in the control of cell proliferation, inflammation and free radicals ***1R could be envisaged as a key element of control, in which different photo-protective strategies are *** latter strategies include,in association with melanins,the antioxidants,anti-inflammatory agents,and DNA repair *** studies have shown the ability of activated MC1R to promote immediate defence responses to ultraviolet radiation-induced oxidative stress,and to exert anti-inflammatory effects by inhibiting NF-kB activity and by counteracting free radical species,generated in response to oxidative ***,an alteration of MC1R,for what concerns its capacity to control these latter aspects,could justify the predisposition to develop melanoma,in the individuals carrying MC1R loss of function variants,not exclusively on the basis of an altered quality and content of *** the possible transcription factors downstream lipid mediated pathways,we will focus on Peroxisome Proliferator-Activated Receptor-γ(PPARγ), known to induce differentiation in several cells,when stimulated by specific *** demonstrate thatα-MSH-mediated PPAR-γactivation is independent from cAMP/PKA pathway, which is responsible for most of melanogenic *** also show that PPAR-γis activated in response to the phospholypase C(PLC)-mediated way,which was never studied before under *** study shows,for the first time,that PPAR-γacts as a transcription factor and physiological effector of melanocyte differentiation downstream of MC1R *** it sets MC1R in a new perspective,as a modulator of lipid involving pathways only partially explored under this G-protein coupled receptor.
