Constitutive expression of feedback-insensitive cystathionine γ-synthase increases methionine levels in soybean leaves and seeds

Soybean （Glycine max （L.） Merr.） is a major crop that provides plant-origin protein and oil for humans and livestock. Al- though the soybean vegetative tissues and seeds provide a major source of high-quality protein, they suffer from low con- centration of an essential sulfur-containing amino acid, methionine, which significantly limits their nutritional quality. The level of methionine is mainly controlled by the first unique enzyme of methionine synthesis, cystathione y-synthase （CGS）. Aiming to elevate methionine level in vegetative tissues and seeds, we constitutively over-expressed a feedback-insensitive Arabidopsis CGS （AtD-CGS） in soybean cultivars, Zigongdongdou （ZD） and Jilinxiaoli 1 （JX）. The levels of soluble methionine increased remarkably in leaves of transgenic soybeans compared to wild-type plants （6.6- and 7.3-fold in two transgenic ZD lines, and 3.7-fold in one transgenic JX line）. Furthermore, the total methionine contents were significantly increased in seeds of the transgenic ZD lines （1.5- to 4.8-fold increase） and the transgenic JX lines （1.3- to 2.3-fold increase） than in the wild type. The protein contents of the transgenic soybean seeds were significantly elevated compared to the wild type, suggesting that the scarcity of methionine in soybeans may limit protein accumulation in soybean seeds. The increased protein content did not alter the profile of major storage proteins in the seeds. Generally, this study provides a promising strategy to increase the levels of methionine and protein in soybean through the breeding programs.

Effect of surface self-nanocrystallization and Si infiltration on Si diffusion behavior, hardness and magnetic properties of pure Fe

Surface nanocrystallization of pure Fe was performed using an improved surface treatment process. The phase transformation and Si infiltration depth of the pure Fe before and after surface mechanical attrition treatment （SMAT） were compared by X-ray diffraction, scanning electron microscopy and energy-dispersive X-ray spectroscopy. The results indicated that nanocrystallization of Fe surface was achieved using SMAT, which resulted in deeper penetration of Si. Prolonging time of SMAT and Si infiltration also resulted in increasing microhardness, with the hardness first increasing with increasing distance from the surface and then decreasing. Furthermore, longer Si infiltration time, nanocrystallization of Si and longer SMAT time resulted in higher saturation magnetization （MS）. The greatest Si penetration depth （150 μm）, maximum hardness （280 HV）, and maximum MS （1.849 × 10^6 A/m） were achieved after SMAT for 45 min and Si infiltration for 9 h. The interaction between adjacent grains after surface nanocrystallization leads to a region of the magnetic domain wall structure located at the grain boundary, which causes the remanence enhancement effect.

Sialon ceramics were prepared by the nitridation of Si,Al and Al2O3 powders with the transition metals(Fe Co,Ni)*** phase composition,microstructure and mechanical properties of specimens were studied Compared with the catalyst-free specimen,the addition of Fe,Co and Ni greatly promoted the nitridation process of Sialon ceramics and improved the weight gain rate and mechanical properties of *** the increased content of catalysts,the apparent porosity of the products decreased first and then increased,contrary to flexural strength and compressive *** specimen with 2.5 wt%Fe had the highest flexural strength 145.8 MPa and it exhibited the highest compressive strength of 594.6 MPa when Co content was 2.5 wt%,whereas they were 71.9 and 78.7 MPa,respectively,when without catalyst.

In order to find leading compounds with an excellent fungicidal activity, the title compound 2-（ 1,3-dithiolan-2- yl-idene）-1-phenyl-2-（ 1,2,4-tfiazol-1-yl） ethanone was synthesized according to the biological isosterism and its structure was confirmed by means of IR, MS, ^1 H NMR and elemental analysis. The single crystal structure of the title compound was determined by X-ray diffraction. The preliminary biological test shows that the synthesized compound exhibits some biological activities.

In this study,nano-sized SnO_(2) decorated on carbon cloth(SnO_(2)/CC)is prepared through a simple and facile solid *** nano-sized SnO_(2) is uniformly distributed on the surface of carbon fibers in carbon cloth,providing sufficient free space to relieve volume expansion and reduce electrode pulverization during *** as-prepared SnO_(2)/CC as a flexible,self-supporting and additive-free anode electrode for sodium-ion/lithium-ion batteries(SIBs/LIBs)can demonstrate outstanding electrochemical ***_(2)/CC after annealing at 350℃(SC-350)as an anode for SIBs can deliver a reversible capacity of 0.587 mA h cm^(-2)at the current density of 0.3 mA cm^(-2)after 100 *** addition,when cycling at 1.5 mA cm^(-2),SC-350 can maintain 1.69 mA h cm^(-2)after 500 cycles when used as LIB *** results illustrate that the as-prepared SnO_(2)/CC can be a promising flexible anode material for flexible SIBs/LIBs and provide a simple and practical method for designing new flexible electrode materials.

P2-type layered oxides are receiving significant interest due to their superior structure and intrinsic *** are strenuous attempts to balance the structure stability,phase transition as well as desirable electrochemical performances by inducing anion/cation ions,changing morphology,adjusting valence,*** this work,several same-period elements of Sc,Ti,V,Cr,Fe,Cu and Zn are doped into Na_(0.50)Li_(0.08)Mn_(0.60)Co_(0.16)Ni_(0.16)O_(2)cathodes,which are manipulated by ions radii and valence state,further studied by operando X-ray powder diffraction patterns(XRD).As a result,the Cu^(2+)doped cathode performed higher rate capacities(as high as 86 mAh/g even at 10 C)and more stable structures(capacity retention of~89.4%for 100 cycles),which owing to the synergistic effect among the tightened TMO_(2)layer,enlarged d-spacing,reduce O-O electrostatic repulsion,ameliorate lattice distortion as well as mitigate ordering of Na^(+)/vacancy.

Analysis of the independent-and interactive-photo-thermal effects on soybean flowering

Soybean（Glycine max（L.） Merr.） is a typical short-day and warm season plant, and the interval between emergence and flowering has long been known to be regulated by environmental factors, primarily photoperiod and temperature. While the effects of photoperiod and temperature on soybean flowering have been extensively studied, a dissection of the component photo-thermal effects has not been documented for Chinese germplasm. Our objective of the current study was to evaluate the independent- and interactive-photo-thermal responses of 71 cultivars from 6 ecotypes spanning the soybean production regions in China. These cultivars were subjected in pot experiments to different temperature regimes by planting in spring（low temperature（LT）） and summer（high temperature（HT））, and integrating with short day（SD, 12 h）, natural day（ND, variable day-length）, and long day（LD, 16 h） treatments over two years. The duration of the vegetative phase from emergence to first bloom（R1） was recorded, and the photo-thermal response was calculated. The outcome of this characterization led to the following conclusions：（1） There were significant differences in photo-thermal response among the different ecotypes. High-latitude ecotypes were less sensitive to the independent- and interactive-photo-thermal effects than low-latitude ecotypes; and（2） there was an interaction between photoperiod and temperature, with the effect of photoperiod on thermal sensitivity being greater under the LD than the SD condition, and with the effect of temperature on photoperiodic sensitivity being greater under the LT than the HT condition. The strengths and limitations of this study are discussed in terms of implications for current knowledge and future research directions. The study provides better understanding of photo-thermal effects on flowering in soybean genotypes from different ecotypes throughout China and of the implications for their adaptation more broadly.

Prototype of the readout electronics for the RICH PID detector in the STCF

The ring imaging Cherenkov(RICH) detector for particle identification(PID) is being evaluated for the future super tau-charm facility(STCF) complex. In this work, the prototype readout electronics for the RICH PID detector is designed. The prototype RICH PID detector is based on a thick gas electron multiplier combined with a micromegas detector for Cherenkov light detection. Considering that there will be a large number(~ 690,000) of detector channels in future RICH detector, the readout electronics faces many challenges to precisely measuring time and charge information, such as reducing the noise,increasing density, and improving precision. The requirements of the readout electronics are explored, the downselection of the ASICs is made and thus a prototype readout electronics is designed and implemented. Tests are also conducted to evaluate the performance of the prototype readout electronics, and the results indicate that the time resolution is better than ~ 1 ns(RMS) when the input charge is greater than ~ 12 fC based on the APV25chip, while the time resolution is better than ~ 1 ns(RMS) at an input charge of over ~ 48 fC based on the AGET and STCF ASIC chips, and the equivalent noise charge is better than ~ 0.5 fC(RMS) @ 20 pF based on the three ASICs. The test results indicate that the prototype readout electronics design meets the requirement of the future RICH PID detector and thus provides a reference for future engineering.

Global gene expression analysis in liver of db/db mice treated with catalpol

Catalpol,a major bioactive component from Rehmannia glutinosa,which has been used to treat *** present study was designed to elucidate the anti-diabetic effect and mechanism of action for catalpol in db/db *** db/db mice were randomly divided into six groups(10/group) according to their blood glucose levels:db/db control,metformin(positive control),and four dose levels of catalpol treatment(25,50,100,and 200 mg·kg^(-1)),and 10 db/m mice were used as the normal *** the groups were administered orally for 8 *** levels of fasting blood glucose(FBG),random blood glucose(RBG),glucose tolerance,insulin tolerance,and glycated serum protein(GSP) and the globe gene expression in liver tissues were *** results showed that catalpol treatment obviously reduced water intake and food intake in a dose-dependent *** treatment also remarkably reduce fasting blood glucose(FBG) and random blood glucose(RBG) in a dose-dependent *** RBG-lowering effect of catalpol was better than that of ***,catalpol significantly improved glucose tolerance and insulin tolerance via increasing insulin *** treatment significantly decreased GSP *** comparisons of gene expression in liver tissues among normal control mice,db/db mice and catalpol treated mice(200 and 100 mg·kg^(-1)) indicated that there were significant increases in the expressions of 287 genes,whichwere mainly involved in lipid metabolism,response to stress,energy metabolism,and cellular processes,and significant decreases in the expressions of 520 genes,which were mainly involved in cell growth,death,immune system,and response to *** genes expressed differentially were linked to glucose metabolism or insulin signaling pathways,including Irs1(insulin receptor substrate 1),Idh2(isocitrate dehydrogenase 2(NADP+),mitochondrial),G6pd2(glucose-6-phosphate dehydrogenase 2),and SOCS3(suppressor of cytokine signaling 3).In conclusion,catalpol e
