AIM: To investigate the anti-tumor effects of nuclear factor-κB (NF-κB) inhibitor SN50 and related mechanisms of SGC7901 human gastric carcinoma cells. METHODS: MTT assay was used to determine the cytotoxic effects of SN50 in gastric cancer cell line SGC7901. Hoechst 33258 staining was used to detect apoptosis morphological changes after SN50 treatment. Activation of autophagy was monitored with monodansylcadaverine (MDC) staining after SN50 *** staining was used to detect the expression of light chain 3 (LC3). Mitochondrial membrane potential was measured using the fluorescent probe JC-1. Western blotting analysis were used to determine the expression of proteins involved in apoptosis and autophagy including p53, p53 upregulated modulator of apoptosis (PUMA), damage-regulated autophagy modulator (DRAM), LC3 and Beclin 1. We detected the effects of p53-mediated autophagy activation on the apoptosis of SGC7901 cells with the p53 inhibitor pifithrin-α. RESULTS: The viability of SGC7901 cells was inhibited after SN50 treatment. Inductions in the expression of apoptotic protein p53 and PUMA as well as autophagic protein DRAM, LC3 and Beclin 1 were detected with Western blotting analysis. SN50-treated cells exhibited punctuate microtubule-associated protein 1 LC3 in immunoreactivity and MDC-labeled vesicles increased after treatment of SN50 by MDC staining. Collapse of mitochondrial membrane potential Δψ were detected for 6 to 24 h after SN50 treatment. SN50-induced increases in PUMA, DRAM, LC3 and Beclin 1 and cell death were blocked by the p53 specific inhibitor pifithrin-α. CONCLUSION: The anti-tumor activity of NF-κB inhibitors is associated with p53-mediated activation of autophagy.

AIM: To investigate the effects of class phosphatidy-linositol 3-kinase (PI3K) inhibitor LY294002 on the invasiveness and related mechanisms of implanted tumors of SGC7901 human gastric carcinoma cells in nude mice. METHODS: Nude mice were randomly divided into model control groups and LY294002 treatment groups. On days 5, 10 and 15 after treatment, the inhibitory rate of tumor growth, pathological changes in tumor specimens, expression levels of matrix metalloproteinase (MMP)-2, MMP-9, CD34 [representing microvessel density (MVD)] and vascular endothelial growth factor (VEGF), as well as apoptosis indexes in tumor samples were ***: In this study, we showed that treatingthe tumors with LY294002 could significantly inhibit carcinoma growth by 11.3%, 29.4% and 36.7%, after 5, 10 and 15 d, respectively, compared to the control group. Hematoxylin & eosin staining indicated that the rate of inhibition increased progressively (23.51% ± 3.11%, 43.20% ± 3.27% and 63.28% ± 2.10% at 5, 10 and 15 d, respectively) along with apoptosis. The expression of MMP-2 was also downregulated (from 71.4% ± 1.6% to 47.9% ± 0.7%, 31.9% ± 0.9% and 7.9% ± 0.7%). The same effects were observed in MMP-9 protein expression (from 49.4% ± 1.5% to 36.9% ± 0.4%, 23.5% ± 0.9% and 7.7% ± 0.6%), the mean MVD (from 51.2% ± 3.1% to 41.9% ± 1.5%, 30.9% ± 1.7% and 14.9% ± 0.8%), and the expression of VEGF (from 47.2% ± 3.1% to 25.9% ± 0.5%, 18.6% ± 1.2% and 5.1% ± 0.9%) by immunohistochemical ***: The classPI3K inhibitor LY294002could inhibit the invasiveness of gastric cancer cells by downregulating the expression of MMP-2, MMP-9, and VEGF, and reducing MVD.

We systematically studied the evaporation residue cross sections of ^(48)Ca-induced reactions on lanthanide and actinide target nuclei under the Dinuclear System(DNS)model framework to check the reliability and applicability of the *** produce new proton-rich Fl and Lv isotopes through hot fusion reactions in the superheavy element region with Z≥104,we utilized the reactions ^(48)Ca+^(236,238,239) Pu and ^(48)Ca+^(242,243,244,250) ***,owing to the detection limit of available equipment(0.1 pb),only 283Fl and 287−289Lv,which have the maximum evaporation residue cross section values of 0.149,0.130,9.522,and 0.309 pb,respectively,can be ***,to produce neutron-deficient isotopes of actinides near the proton drip line with Z=93−100,we attempted to generate the new isotopes(224−227Pu,228−232,237Cm)using the reactions ^(48)Ca+180,182,183 W and ^(48)Ca+^(184,186,187,192) *** maximum evaporation residue cross section values are 0.07,0.06,0.26,and 0.30 nb for the former set of reactions,and 1.96 pb,5.73 pb,12.16 pb,19.39 pb,54.79 pb,and 6.45 nb for the latter,*** results are expected to provide new information for the future synthesis of unknown neutron-deficient isotopes.

Strengthening interface bonding between boron nitride nanosheets(BNNS) and copper matrix is an essential prerequisite for exploiting a new generation of copper matrix composites(CMCs) with high strength and wear resistance. Herein, BNNS/Cu composites were fabricated by the powder metallurgy route, matrix-alloying(adding 1.0 wt% Ti) strategy was adopted to improve the interfacial wettability and strengthen interface adhesion. A typical "sandwich"-like multiply interface structure involving TiN transition layers, BNNS and Cu matrix had been well constructed through the rational heat treatment(900℃for 120 min). Additionally, nano-sized TiB whisker was in situ formed in the vicinity of the interface, it had linked the BNNS-Cu-TiN multiply interface, which played a role of "threading the needle" and significantly strengthened the multi-interfaces bonding. This specific interface structure was finely characterized, and the formation mechanism of solid-state interfacial reaction feature was proposed. The results demonstrated that the ultimate tensile strength(UTS) of BNNS/Cu-(Ti)-900℃ increased from 248 to 530 MPa(increased by 114%), and the coefficient of friction(COF) decreased from 0.51 to 0.28 than pure Cu. This work highlights the importance of interface configuration design, which contributes to the development of CMCs with prominent comprehensive properties.

AIM: To investigate the effects of small interfering RNA (siRNA)-mediated inhibition of Class?I?phosphoinositide 3-kinase (Class?I?PI3K) signal transduction on the proliferation, apoptosis, and autophagy of gastric cancer SGC7901 and MGC803 cells.METHODS: We constructed the recombinant replication adenovirus PI3K(I)-RNA interference (RNAi)-green fluorescent protein (GFP) and control adenovirus NC-RNAi-GFP, and infected it into human gastric cancer cells. MTT assay was used to determine the growth rate of the gastric cancer cells. Activation of autophagy was monitored with monodansylcadaverine (MDC) staining after adenovirus PI3K(I)-RNAi-GFP and control adenovirus NC-RNAi-GFP treatment. Immunofluorescence staining was used to detect the expression of microtubule-associated protein 1 light chain 3 (LC3). Mitochondrial membrane potential was measured using the fluorescent probe JC-1. The expression of autophagy was monitored with MDC, LC3 staining, and transmission electron microscopy. Western blotting was used to detect p53, Beclin-1, Bcl-2, and LC3 protein expression in the culture supernatant.RESULTS: The viability of gastric cancer cells was inhibited after siRNA targeting to the Class?I?PI3K blocked Class?I?PI3K signal pathway. MTT assays revealed that, after SGC7901 cancer cells were treated with adenovirus PI3K(I)-RNAi-GFP, the rate of inhibition reached 27.48% ± 2.71% at 24 h, 41.92% ± 2.02% at 48 h, and 50.85% ± 0.91% at 72 h. After MGC803 cancer cells were treated with adenovirus PI3K(I)-RNAi-GFP, the rate of inhibition reached 24.39% ± 0.93% at 24 h, 47.00% ± 0.87% at 48 h, and 70.30% ± 0.86% at 72 h (P < 0.05 compared to control group). It was determined that when 50 MOI, the transfection efficiency was 95% ± 2.4%. Adenovirus PI3K(I)-RNAi-GFP (50 MOI) induced mitochondrial dysfunction and activated cell apoptosis in SGC7901 cells, and the results described here prove that RNAi of Class?I?PI3K induced apoptosis in SGC7901 cells. In recent years,deep learning methods have gradually been applied to prediction tasks related to Arctic sea ice concentration,but relatively little research has been conducted for larger spatial and temporal scales,mainly due to the limited time coverage of observations and reanalysis ***,deep learning predictions of sea ice thickness(SIT)have yet to receive ample *** this study,two data-driven deep learning(DL)models are built based on the ConvLSTM and fully convolutional U-net(FC-Unet)algorithms and trained using CMIP6 historical simulations for transfer learning and fine-tuned using reanalysis/*** models enable monthly predictions of Arctic SIT without considering the complex physical processes *** comprehensive assessments of prediction skills by season and region,the results suggest that using a broader set of CMIP6 data for transfer learning,as well as incorporating multiple climate variables as predictors,contribute to better prediction results,although both DL models can effectively predict the spatiotemporal features of SIT *** the predicted SIT anomalies of the FC-Unet model,the spatial correlations with reanalysis reach an average level of 89%over all months,while the temporal anomaly correlation coefficients are close to unity in most *** models also demonstrate robust performances in predicting SIT and SIE during extreme *** effectiveness and reliability of the proposed deep transfer learning models in predicting Arctic SIT can facilitate more accurate pan-Arctic predictions,aiding climate change research and real-time business applications.

To solve the divergence problem and overcome the difficulty in guaranteeing filtering accuracy during estimation of the process noise covariance or the measurement noise covariance with traditional new information-based nonlinear filtering methods,we design a new method for estimating noise statistical characteristics of nonlinear systems based on the credibility Kalman Filter(KF)theory considering noise *** method first extends credibility to the Unscented Kalman Filter(UKF)and Extended Kalman Filter(EKF)based on the credibility ***,an optimization model for nonlinear credibility under noise related conditions is established considering noise correlation.A combination of filtering smoothing and credibility iteration formula is used to improve the real-time performance of the nonlinear adaptive credibility KF algorithm,further expanding its application scenarios,and the derivation process of the formula theory is ***,the performance of the nonlinear credibility filtering algorithm is simulated and analyzed from multiple perspectives,and a comparative analysis conducted on specific experimental *** simulation and experimental results show that the proposed credibility EKF and credibility UKF algorithms can estimate the noise covariance more accurately and effectively with lower average estimation time than traditional methods,indicating that the proposed algorithm has stable estimation performance and good real-time performance.

The design of universal segments and deviation control of segment assembly are essential for robust and low-risk tunnel construction.A building information modeling(BIM)-based framework was proposed for parametric modeling,automatic assembly,and deviation control of universal ***,segment models of different levels of detail(LoDs)were built based on BIM visual programming language(VPL)for different project life ***,the geometric constraints,requirements,and procedures for parametric segment assembly were distilled to develop a program that combines a novel typesetting algorithm with a 3D path replanning *** is implemented by introducing a point indication matrix,characterizing segments by sides,and manipulating geometries in a ***,3D path replanning,with non-uniform rational B-splines(NURBS)and arcs as basic shapes,was used to resolve unacceptable deviation situations after ***,the proposed framework was validated on a water diversion line and was found to be more effective and accurate than the previous method.
