X-box-binding protein 1-modified neural stem cells for treatment of Parkinson's disease

X-box-binding protein 1-transfected neural stem cells were transplanted into the right lateral ventricles of rats with rotenone-induced Parkinson＇s disease. The survival capacities and differentiation rates of cells expressing the dopaminergic marker tyrosine hydroxylase were higher in X-box-binding protein 1-transfected neural stem cells compared to non-transfected cells. Moreover, dopamine and 3,4-dihydroxyphenylacetic acid levels in the substantia nigra were significantly increased, α-synuclein expression was decreased, and neurological behaviors were significantly ameliorated in rats following transplantation of X-box-binding protein 1-transfected neural stem cells. These results indicate that transplantation of X-box-binding protein 1-transfected neural stem cells can promote stem cell survival and differentiation into dopaminergic neurons, increase dopamine and 3,4-dihydroxyphenylacetic acid levels, reduce α-synuclein aggregation in the substantia nigra, and improve the symptoms of Parkinson＇s disease in rats.

Based on the absorption property of a diazine that can be formed by reaction of glyoxal and 3-methyl-2-benzothiazolinone hydrazone(MBTH)in the Ultravioletvisible(UV-vis)spectral region,a HPLC method was developed for the determination of glyoxal in acetaldehyde *** was derivatised from MBTH and the derivatives(diazine)were analyzed by HPLC for identification and *** determination was performed on a ZORBAX Eclipse XDB-C18 column(4.6250 mm,5 mm)at 35℃ with an injection volume of 10 mL,using a mixture of acetonitrile-water solvent(99∶5,v∶v)as a mobile phase with a flow rate of 0.8 mL·min^(–1).The proper derivative reaction conditions were the temperature of 70℃,MBTH to carbonyl molar ratio of 12,and reaction time of 110 *** glyoxal diazine was a yellow dye with a maximum molar absorptivity at 401 nm and its retention time was 5.2 min under optimal HPLC *** standard curve for glyoxal had a strong linear relationship with a regression coefficient(R2=0.999)in the range of 0.002–0.020 g·L^(–1).The analysis of glyoxal in an oxidising solution gave accurate results with a relative standard deviation(RSD)value of 0.55%.The average relative recovery was 102%.This efficient HPLC technique is also proposed for detecting other dicarbonyl compounds besides glyoxal.

Mechanochemical synthesis of MAPbBr_(3)/carbon sphere composites for boosting carrier-involved superoxide species

Lead halide perovskites MAPbX_(3)(MA = CH_(3)NH_(3) or Cs;X = I, Br, Cl) are well considered to be potential candidates for photocatalytic reaction due to its excellent photoelectrical properties, but they still suffer from the low charge separation efficiency and slow catalytic reaction dynamics. To tackle the drawbacks, herein, MAPbBr_(3)/carbon sphere(CS) composite photocatalysts using glucose as the carbon source were elaborately designed and fabricated via a dry mechanochemical grinding process. The interfacial interaction Pb-O-C chemical bonds were constructed between MAPbBr_(3) and the carbon sphere surface containing organic functional groups. By optimizing the content of CSs, the enhanced photocatalytic degradation kinetic rate of Malachite Green(MG) pollutants(92% within 20 min) for MAPbBr_(3)/CS x( x = 17 wt.%) is about 3.6-fold of that for pristine MAPbBr_(3), which is attributed to the corporative adsorption and enhanced carrier transportation and separation of MAPbBr_(3)/CS x. Furthermore, the possible degradation mechanism was proposed on basis of the electrochemical, mass spectrometry and optical characterization results. Owing to the robust interfacial interaction, effective electron extraction rate( k et = 4.6 × 10^(7) sec^(-1)) from MAPbBr_(3) to CS can be established, which driven oxygen activation where superoxide radicals(·O^(2-)) played an important role in MG degradation. It is expected that mechanochemistry strategy may provide a new route to design efficient lead halide perovskite-carbon or metal oxide or sulfide composite photocatalysts.

目的:评价根据术前外周血系统性免疫炎症指数(systemic immune inflammation index,SII)、中性粒细胞与淋巴细胞比值(neutrophil to lymphocyte ratio,NLR)构建的列线图模型对胃癌根治术后患者预后的预测价值。方法:323例胃癌术后患者以7∶3比例随机分为训练组和验证组。使用X-Tile软件计算SII和NLR的最佳临界值,据此分为高、低两组,采用单因素和多因素回归分析来评估SII和NLR的预后价值,并以此构建预测患者总生存期(overall survival,OS)和无进展生存期(progression-free survival,PFS)的列线图。一致性指数(consistency index,C-index)、绘制受试者工作特征曲线(receiver operating characteristic curve,ROC)和校准曲线与传统TNM分期比较,及决策曲线(decision curve analysis,DCA)用以评价模型。结果:筛选出包括SII和PLR在内的6个独立危险因素(P<0.05);预测总生存期(OS)的列线图C-index为训练集(0.804,95%CI:0.786~0.822)和验证集(0.780,95%CI:0.746~0.814),预测无进展生存期(PFS)列线图的C-index分别为训练集(0.806,95%CI:0.787~0.823)和验证集(0.778,95%CI:0.746~0.810),均明显高于TNM分期。ROC和校准曲线结果显示列线图模型在OS和PFS方面均具有较高的预测准确性;DCA显示该模型具有良好的临床净收益。结论:SII和NLR是影响胃癌根治术后患者预后独立影响因素。SII和NLR构建的列线图模型对胃癌根治术后患者预后具有较高的预测价值。

The characteristic curve of methane adsorbed on coal and its role in the coalbed methane storage research

Based on the adsorption potential theory, this paper processes and analyzes the isothermal adsorption data at different temperatures of four coal samples with different ranks. The research results indicate that the adsorption characteristic curve of methane adsorbed on the coal sample is a single curve, and the mathematical expression of the methane adsorption capacity, temperature and pressure is achieved from the characteristic curve. According to the calculating routine and method presented, the adsorption capacity at other equilibrium conditions can be calculated from the adsorption data of one temperature. The results of this paper are a significant contribution to the research on the storage mechanism of the coalbed methane.

Identification of QTL for Popping Characteristics Using a BC_2F_2 Population and Comparison with Its F_(2:3) Population in Popcorn

Normal corn germplasm can be used to improve popcorn （Zea mays L.） through 1-2 backcrosses with popcorn as recurrent parents. Popping characteristics of derived lines from popcorn × dent or flint corn crosses play a determinant role in popcorn breeding. Advanced backcross QTL methods can effectively combine QTL identification and plant breeding. 220 selected BC2F2 families developed from a cross between Dan 232, a dent corn inbred line, and N04, an elite popcorn inbred line, were evaluated for three popping characteristics, popping volume, flake size, and popping rate, under two environmental conditions. Using composite interval mapping, a total of 10 significant QTLs were detected, and of these, 2 to 4 QTLs were identified for each trait. Six QTLs had favorable alleles contributed by Dan 232. Comparison with the 15 QTLs detected in the F2：3 families showed that 3 QTLs were the same in both populations. The QTLs should be redetected in generations developed through severe selection. Improved N04 and near isogenic lines could be developed from this BC2F2 population through selfing or another 1 to 2 backcrosses with N04.

Paul-Baker systems with 4° flat field and 5° fiat field are studied. Their light obstructions under different f/ratios of the primary mirror are analyzed. Due to the strong f/ratio of the system, a focal length extender is designed in order to match the following fiber instrumentation, and two kinds of dispersion prism correctors are designed for correcting the atmospheric dispersion. We compare the designed Paul-Baker system with LAMOST, the national major scientific project now under construction.
