Mechanism of acupuncture for limb spasm of post-stroke spasticity based on GABA and BDNF/Trkb-KCC2 signaling pathway

Background:To investigate the effects of acupuncture on post-stroke limb spasm model rats and the underlying mechanism.Methods:A total of 50 Sprague-Dawley rats were randomly divided into three groups,Control group(10),Model group(20)and Zhishen Tiaoxing(ZSTX)acupuncture group(20).Middle cerebral artery occlusion was conducted in SD rats to establish post-stroke limb spasm rats,which were treated with ZSTX acupuncture.Behavioral assays were determined by the Narrow ally test,the limb muscle tension was detected by the BL-420S test system,and infarct volume was assessed after the cerebral infarction by 2,3,5-triphenyltetrazolium chloride staining.Heterogeneous neurotransmitterγ-aminobutyric acid(GABA)and its receptors GABAA and GABAB in the cerebral cortex of the infarct area were determined by immunofluorescence assay.The release of Trkb and K-Cl cotransporter isoform 2 was detected by an immunofluorescence double labeling study.Western Blot was utilized to measure the expression of BDNF and Trkb.Results:The results showed that the behavioral assays in post-stroke limb spasm rats were significantly improved by the treatment of ZSTX acupuncture.14 days of ZSTX acupuncture can effectively inhibit muscle tone and decrease Infarct volume,which was measured with BL-420S biological function experiment system and triphenyltetrazolium chloride.Meanwhile,the results of Double-Label Immunofluorescence Assays showed that ZSTX acupuncture improved the expression of GABA,GABAA,GABAB,BDNF,and K-Cl cotransporter isoform 2.Double-Label Immunofluorescence Assays and WB results showed that 14 days ZSTX acupuncture declined the expression of Trkb.Conclusions:Our results suggest that 14 days of ZSTX acupuncture can significantly improve post-stroke limb spasm.Meanwhile,the pathogenesis of post-stroke limb spasm and the efficacy of ZSTX acupuncture involve metabolic pathways of neurotransmitters,and electro-acupuncture can treat post-stroke limb spasm by regulating BDNF/Trkb-KCC2 signaling pathway.

Dopamine D2 receptors in the dorsomedial prefrontal cortex modulate social hierarchy in male mice

Social hierarchy greatly influences behavior and health.Both human and animal studies have signaled the medial prefrontal cortex(mPFC)as specifically related to social hierarchy.Dopamine D1 receptors(D1Rs)and D2 receptors(D2Rs)are abundantly expressed in the mPFC,modulat-ing its functions.However,it is unclear how DR-expressing neurons in the mPFC regulate social hierarchy.Here,using a confrontation tube test,we found that most adult C57BL/6J male mice could establish a linear social rank after 1 week of cohabitation.Lower rank individuals showed social anxiety together with decreased serum testosterone levels.D2R expression was significantly downregulated in the dorsal part of mPFC(dmPFC)in lower rank individuals,whereas D1R expression showed no significant difference among the rank groups in the whole mPFC.Virus knockdown of D2Rs in the dmPFC led to mice being particularly prone to lose the contests in the confrontation tube test.Finally,simultaneous D2R activation in the subordinates and D2R inhibition in the dominants in a pair switched their dominant-subordinate relationship.The above results indicate that D2Rs in the dmPFC play an important role in social dominance.Our findings provide novel insights into the divergent func-tions of prefrontal D1Rs and D2Rs in social dominance,which may contribute to ameliorating social dysfunctions along with abnormal social hierarchy.

High-lattice-adapted surface modifying Na_(4)MnV(PO_(4))_(3) for better sodium storage

Sodium-ion batteries(SIBs)are required to possess long cycle life when used for large-scale energy storage.The polyanionic Na_(4)MnV(PO_(4))_(3)(NMVP)reveals good cyclic stability due to its unique three-dimensional(3D)frame structure,but it still faces the challenge of interfacial degradation in practical applications.In this work,NASICON-type Na_(1.3)Al_(0.7)Ti_(1.3)(PO_(4))_(3)(NATP)was deposited on the surface of NMVP to promote interface stability by surface modification and gradient doping.As a result,the optimized NMVP@2%NATP released a capacity retention of 44.8%after 1000 cycles at 5 C,much higher than that of the initial NMVP(28.9%).The enhanced electrochemical performance was mainly attributed to NATP coating acting as a fast ion transport carrier and physical barrier,significantly facilitating the Na^(+)diffusion and isolating side reaction at the electrode/electrolyte interface.On the other hand,Ti_(4)^(+)and Al^(3+)cations from the NATP were partially doped inside the NMVP surface to boost the transport of Na^(+),and the perfect lattice matching of NVMP and NATP improved the interface and structural stability accompanying long cycling.This work demonstrated the effectiveness of surface modification with high lattice match material and provided new perspectives for high energy density solid-state SIBs.

Multicolor emission from lanthanide ions doped lead-free Cs_(3)Sb_(2)Cl_(9) perovskite nanocrystals

The toxicity of lead ions has become the severe challenge for the all-inorganic lead halide p erovskite materials,although some works have rep orted the lead-free perovskite nanocrystals(NCs),the photoluminescence quantum yield(PLQY)of these materials is still unsatisfactory.Meanwhile,because the halogen ions can be easily exchanged,the controllable multicolor emission in perovskite NCs is difficult to realize in current reports.In this work,we introduced lanthanide ions into lead-free Cs_(3)Sb_(2)Cl_(9) perovskite NCs.Benefitting from the energy transfer between Cs_(3)Sb_(2)Cl_(9) perovskite NC host and lanthanide ions,the multicolor emission was realized.Based on controlling the doping concentration of Tb^(3+)and Eu^(3+)ions,the white light emission under UV excitation would be turned easily in the Tb^(3+)/Eu^(3+)codoped NCs.In addition,efficient energy transfer from perovskite NCs to Tb^(3+)or Eu^(3+)ions is beneficial to improving the optical properties of lead-free perovskite NCs,resulting in maximum PLQYs of red,green and white light emission of 22.6%,19.7%and 28.5%,respectively.Finally,a white light emitting device(WLED)was fabricated with a power efficiency of 18.5 lm/W,which presents the Commission Internationale de l'Eclairage(CIE)of(0.33,0.35).

Effect of operational and geometric parameters on the hydrodynamics of a Wurster coater:A CFD-DEM study

A coupled CFD-DEM method was used to study the hydrodynamics of a Wurster coater.Firstly,the CFD part of the model was validated by the accurate prediction of the pressure drops over a pseudo-2D fluidized bed under various gas velocities.The effect of gas velocity,gap height,tube length and batch volume of the particles on the cycle time and the residence time of the particles was thoroughly investigated.The central jet gas velocity ui was found to speed up the particle cycle but undermine the coating efficiency.The gas velocity at the horizontal transport zone u2 was able to promote the horizontal transport of the particles but should not be too high,otherwise,it would obstruct the normal falling back of the particles in the downward zone.Big gap heights would decrease the coating efficiency but tube length had little impact on that.The increment of batch volume would commonly abase the cycle time and the working efficiency under a given u1.The de-fluidization problem arose when the batch volume increased to 550 mL.However,this problem could be swept out by the optimization of u1 and u2.In a mixture of different sizes,the coarse particles enjoyed higher coating efficiency and could travel closer to the nozzles.This may shield the fine particles from getting enough coating liquids,and thus coarse particles and fine particles were not recommended to get coated in the same batch.

Background:Warm acupuncture,or warm-needle acupuncture,is a traditional external treatment widely used in China for thousands of years.However,the effects of different treatment times on limb spasticity post-stroke have not been evaluated.Methods:We searched PubMed,Embase,the Cochrane Library,the Web of Science,the China National Knowledge Infrastructure,the Chinese Scientific and Journal Database,the Wanfang database,and the Chinese Biomedical Literature Database from inception to May 21st,2020,to seek information regarding randomized controlled trials studying warm-needle acupuncture for limb spasticity post-stroke.Two authors selected 24 studies fitting the eligibility criteria and developed this systematic review and meta-analysis.Results:Warm-needle acupuncture is shown to be more effective than other treatments used in the control groups(odds ratio(OR)=3.61,95%confidence intervals(CI)(2.51,5.19),Plinic Spasticity Index(MD=1.06,95%CI(0.51,1.60),P=0.0002),improved sensorimotor impairments according to the Fugl-Meyer Assessment(MD=9.51,95%CI(6.99,12.03),P<0.001),and improved activities of daily living according to the Barthel Index(MD=9.70,95%CI(6.27,13.12),P<0.001).Subgroup analysis based on the length of the treatment using the Modified Ashworth Scale,Fugl-Meyer Assessment,and Barthel Index showed better outcomes for patients who received more than 20 days of treatments(P<0.001).Conclusion:Warm-needle acupuncture is an effective treatment for limb spasticity post-stroke.A treatment frequency of more than 20 days appears to have better effectiveness than that of less than 20 days.

Tetrabromobisphenol A exerts thyroid disrupting effects but has little overt impact on postnatal brain development and neurobehaviors in mice

Tetrabromobisphenol A(TBBPA)is a widely used brominated flame retardant.There is evidence showing that TBBPA can exert thyroid disrupting effects in mammals,but different results were also reported,along with inconsistent reports regarding its neurotoxicity.Here,we investigated thyroid disrupting effects and neurotoxicity of TBBPA(5,50,500μg/(kg·day))to male mice following maternal and direct exposure through drinking water,with the antithyroid drug propylthiouracil(PTU)as the positive control.On postnatal day(PND)15,we expectedly observed severe thyroid compensatory hyperplasia and cerebellar developmental retardation in PTU-treated pups.The highest dose of TBBPA also caused thyroid histological alteration but had no effects on cerebellar development in terms of Purkinje cell morphology and the thickness of the internal granular layer and the molecular layer of the cerebellum.During puberty and adulthood,the thyroid morphological alterations became more pronounced in the TBBPA-treated animals,accompanied by decreased serum thyroid hormone levels.Furthermore,the 50 and 500μg/(kg·day)TBBPA groups showed a significant decrease in the serum level of serotonin,a neurotransmitter associated with anxiety behaviors.Correspondingly,the highest dose group displayed anxiety-like behaviors in the elevated plus-maze test on PND 35,but this neurobehavioral alteration disappeared on PND 56.Moreover,no changes in neurobehavioral parameters tested were found in TBBPAtreated animals at puberty and adulthood.Altogether,all observations show that TBBPA can exert thyroid disrupting effects but has little overt impact on brain development and neurobehaviors in mice,suggesting that thyroid disruption does not necessarily cause overtly adverse neurodevelopmental outcomes.

A New Method for the Aging Evaluation of Oil-Paper Insulation Using η-Butanol and Methanol

To improve the accuracy and efficiency of the aging life prediction and assessment of transformer oil-paper insulation,and to make up for the deficiencies of traditional characterizers,such as 2-furfural,carbon monoxide,and carbon dioxide,a method for the simultaneous determination of methanol,ethanol,n−propanol,and η-butanol in oil with a single injection is established by headspace-gas chromatography-mass spectrometry.The measured results show that the determination limits of the four alcohol characterizers can be controlled to 10μg/kg level.Based on this method,the change patterns of the above four alcohols with thermal aging time and degree of polymerization are obtained through thermal aging experimental research.Ethanol,η-propanol,and η-butanol in oil indicate nearly linear correlations with thermal aging time and degree of polymerization,similar to that of methanol.By analyzing 52 sets of measured data of 500 kV EHV transformers in operation,η-butanol is found to have excellent performance,and a new method to evaluate the aging state of oil-paper insulation employing η-butanol and methanol is proposed along with the aging attention value model.The measured data of 500 kV EHV transformers in operation indicate that the combination of η-butanol and methanol as the preferred characterizers can effectively compensate for the shortcoming of traditional characterizers in the early stages of aging,and the feasibility of the method is verified.Two possible pathways for the generation of η-butanol by cellulose cleavage during the aging of oil-paper are proposed from the chemical structure of cellulose.

Reactive oxygen species(ROS)are produced as undesirable by-products of metabolism in various cellular compartments,especially in response to unfavorable environmental conditions,throughout the life cycle of plants.Stressinduced ROS production disrupts normal cellular function and leads to oxidative damage.To cope with excessive ROS,plants are equipped with a sophisticated antioxidative defense system consisting of enzymatic and non-enzymatic components that scavenge ROS or inhibit their harmful effects on biomolecules.Nonetheless,when maintained at relatively low levels,ROS act as signaling molecules that regulate plant growth,development,and adaptation to adverse conditions.Here,we provide an overview of current approaches for detecting ROS.We also discuss recent advances in understanding ROS signaling,ROS metabolism,and the roles of ROS in plant growth and responses to various abiotic stresses.
