Current landscape of preoperative neoadjuvant therapies for initial resectable colorectal cancer liver metastasis

Colorectal cancer liver metastasis(CRLM)presents a clinical challenge,and optimizing treatment strategies is crucial for improving patient outcomes.Surgical resection,a key element in achieving prolonged survival,is often linked to a heightened risk of recurrence.Acknowledging the potential benefits of preoperative neoadjuvant chemotherapy in managing resectable liver metastases,this approach has gained attention for its role in tumor downsizing,assessing biological behavior,and reducing the risk of postoperative recurrence.However,the use of neoadjuvant chemotherapy in initially resectable CRLM sparks ongoing debates.The balance between tumor reduction and the risk of hepatic injury,coupled with concerns about delaying surgery,necessitates a nuanced approach.This article explores recent research insights and draws upon the practical experiences at our center to address critical issues regarding considerations for initially resectable cases.Examining the criteria for patient selection and the judicious choice of neoadjuvant regimens are pivotal areas of discussion.Striking the right balance between maximizing treatment efficacy and minimizing adverse effects is imperative.The dynamic landscape of precision medicine is also reflected in the evolving role of gene testing,such as RAS/BRAF and PIK3CA,in tailoring neoadjuvant regimens.Furthermore,the review emphasizes the need for a multidisciplinary approach to navigate the comp-lexities of CRLM.Integrating technical expertise and biological insights is crucial in refining neoadjuvant strategies.The management of progression following neoadjuvant chemotherapy requires a tailored approach,acknowledging the diverse biological behaviors that may emerge.In conclusion,this review aims to provide a comprehensive perspective on the considerations,challenges,and advancements in the use of neoadjuvant chemotherapy for initially resectable CRLM.By combining evidencebased insights with practical experiences,we aspire to contribute to the ongoing discourse on refining treatment

Solving Multi-Objective Vehicle Routing Problems with Time Windows: A Decomposition-Based Multiform Optimization Approach

In solving multi-objective vehicle routing problems with time windows (MOVRPTW),most existing algorithms focus on the optimization of a single problem formulation. However,little effort has been devoted to exploiting valuable knowledge from the alternate formulations of MOVRPTW for better optimization performance. Aiming at this insufficiency,this study proposes a decomposition-based multi-objective multiform evolutionary algorithm (MMFEA/D),which performs the evolutionary search on multiple alternate formulations of MOVRPTW simultaneously to complement each other. In particular,the main characteristics of MMFEA/D are three folds. First,a multiform construction (MFC) strategy is adopted to construct multiple alternate formulations,each of which is formulated by grouping several adjacent subproblems based on the decomposition of MOVRPTW. Second,a transfer reproduction (TFR) mechanism is designed to generate offspring for each formulation via transferring promising solutions from other formulations,making that the useful traits captured from different formulations can be shared and leveraged to guide the evolutionary search. Third,an adaptive local search (ALS) strategy is developed to invest search effort on different alternate formulations as per their usefulness for MOVRPTW,thus facilitating the efficient allocation of computational resources. Experimental studies have demonstrated the superior performance of MMFEA/D on the classical Solomon instances and the real-world instances.

Role of CYP311A1 in wing development of Drosophila melanogaster

Lipid homeostasis is crucial for growth and development of organisms.Several cytochrome P450 monooxygenases(CYPs)are involved in lipid metabolism.The function of Cyp3llal in the anterior midgut as a regulator of phosphatidylethanolamine(PE)metabolism in Drosophila melanogaster has been demonstrated,as depletion of Cyp31lal caused larval growth arrest that was partially rescued by supplying PE.In this study,we investigated the role of CYP311A1 in wing morphogenesis in Drosophila.Using the GAL4-UAS system,Cyp31lal was selectively knocked down in the wing disc.A deformed wing phenotype was observed in flies with reduced Cyp31lal transcripts.BODIPY and oil red O staining revealed a reduction of neutral lipids in the wing disc after the depletion of Cyp31lal.In addition,we observed an enhanced sensitivity to Eosin Y penetration in the wings of Cyp31lal knocked-down flies.Moreover,the reduction of CYP311A1 function in developing wings does not affect cell proliferation and apoptosis,but entails disordered Phalloidin or Cadherin distribution,suggesting an abnormal cell morphology and cell cortex structure in wing epithelial cells.Taken together,our results suggest that Cyp3llal is needed for wing morphogenesis by participating in lipid assembly and cell homeostasis.

Controllable shaping of high-index dielectric nanoparticles by exploiting the giant optical force of femtosecond laser pulses

Nanoparticles made of different materials usually support optical resonances in the visible to near infrared spectral range,such as the localized surface plasmons observed in metallic nanoparticles and the Mie resonances observed in dielectric ones.Such optical resonances,which are important for practical applications,depend strongly on the morphologies of nanoparticles.Laser irradiation is a simple but effective way to modify such optical resonances through the change in the morphology of a nanoparticle.Although laser-induced shaping of metallic nanoparticles has been successfully demonstrated,it remains a big challenge for dielectric nanoparticles due to their larger Young’s modulus and smaller thermal conductivities.Here,we proposed and demonstrated a strategy for realizing controllable shaping of high-index dielectric nanoparticles by exploiting the giant optical force induced by femtosecond laser pulses.It was found that both Si and Ge nanoparticles can be lit up by resonantly exciting the optical resonances with femtosecond laser pulses,leading to the luminescence burst when the laser power exceeds a threshold.In addition,the morphologies of Si and Ge nanoparticles can be modified by utilizing the giant absorption force exerted on them and the reduced Young’s modulus at high temperatures.The shape transformation from sphere to ellipsoid can be realized by laser irradiation,leading to the blueshifts of the optical resonances.It was found that Si and Ge nanoparticles were generally elongated along the direction parallel to the polarization of the laser light.Controllable shaping of Si and Ge can be achieved by deliberately adjusting the excitation wavelength and the laser power.Our findings are helpful for understanding the giant absorption force of femtosecond laser light and are useful for designing nanoscale photonic devices based on shaped highindex nanoparticles.

The 2023 Turkey earthquake doublet: Earthquake relocation, seismic tomography, and stress field inversion

On February 6,2023,two earthquakes with magnitudes of M_(W) 7.8 and M_(W) 7.5 struck southeastern Turkey,causing significant casualties and economic losses.These seismic events occurred along the East Anatolian Fault Zone,a convergent boundary between the Arabian Plate and the Anatolian Subplate.In this study,we analyze the M_(W) 7.8 and M_(W) 7.5 earthquakes by comparing their aftershock relocations,tomographic images,and stress field inversions.The earthquakes were localized in the upper crust and exhibited steep dip angles.Furthermore,the aftershocks occurred either close to the boundaries of low and high P-wave velocity anomaly zones or within the low P-wave velocity anomaly zones.The East Anatolia Fault,associated with the M_(W) 7.8 earthquake,and the SürgüFault,related to the M_(W) 7.5 earthquake,predominantly experienced shear stress.However,their western sections experienced a combination of strike-slip and tensile stresses in addition to shear stress.The ruptures of the M_(W) 7.8 and M_(W) 7.5 earthquakes appear to have bridged a seismic gap that had seen sparse seismicity over the past 200 years prior to the 2023 Turkey earthquake sequence.

Effects of zero-valent iron added in the flooding or drainage process on cadmium immobilization in an acid paddy soil

Zero-valent iron(ZVI)is a promising material for the remediation of Cd-contaminated paddy soils.However,the effects of ZVI added during flooding or drainage processes on cadmium(Cd)retention remain unclear.Herein,Cd-contaminated paddy soil was incubated for 40days of flooding and then for 15 days of drainage,and the underlying mechanisms of Cd immobilization coupled with Fe/S/N redox processes were investigated.The addition of ZVI to the flooding process was more conducive to Cd immobilization.Less potential available Cd was detected by adding ZVI before flooding,which may be due to the increase in paddy soil pH and newly formed secondary Fe minerals.Moreover,the reductive dissolution of Fe minerals promoted the release of soil colloids,thereby increasing significantly the surface sites and causing Cd immobilization.Additionally,the addition of ZVI before flooding played a vital role in Cd retention after soil drainage.In contrast,the addition of ZVI in the drainage phase was not conducive to Cd retention,which might be due to the rapid decrease in soil pH that inhibited Cd adsorption and further immobilization on soil surfaces.The findings of this study demonstrated that Cd availability in paddy soil was largely reduced by adding ZVI during the flooding period and provide a novel insight into the mechanisms of ZVI remediation in Cd-contaminated paddy soils.

Investigation of wake-induced vibration interference between two staggered wave-cone cylinders at subcritical Reynolds number

The wake induced vibration(WIV)of a one-and two-degree-of-freedom(1DOF,2DOF)downstream wave-cone cylinder(WCC)behind a stationary equal-size upstream wave-cone cylinder in the staggered arrangement is numerically investigated at subcritical Reynolds number of 3900 by using shear stressed transfer(SST)k-ωturbulence model.The streamwise pitch ratios(P/Dm)vary from 4 to 6 with a fixed incident angleα=8°.Experimental measurements were also performed for the validation of the present numerical models.It is found that the largest vibration amplitude in crossflow direction occurred at P/Dm=4,Ur=8 with small difference of streamwise vibration at P/Dm=4,6.Different from single wavy-cone cylinder(SWCC),the downstream flexible one of a pair staggered WCCs got larger vibration amplitude during phase switching stage instead of in-phase stage.The upstream wake will suppress the triple frequency of main frequency in the power spectra density(PSD)functions of Cl but stimulate the double one of that.An intriguing vibration mechanism happened in all 2DOF cases where the trajectory of downstream WCC is a significant ellipse rather than a figure of 8.The transformation of phase switching and the variation of the main frequency of drag coefficient(Cd)can be explained by the vortex-shedding modes of downstream WCC.

Interface Design Strategy for GNS/AZ91 Composites with Semi-Coherent Structure

The interfacial structure plays an important role in the mechanical properties of magnesium matrix composite(MMCs)reinforced with graphene nanosheet(GNS)due to their poor wettability with the Mg matrix.An interface design strategy was proposed to form the semi-coherent interfacial structure with superior bonding strength.The lattice mismatch and interfacial bonding strength between Mg/rare earth oxide/carbon were utilized as key characteristics to evaluate the interfacial structure.Lanthanum oxide(La2O3)was selected as the intermediate candidate due to its low lattice mismatch and high interfacial bonding strength.To identify the interfacial structure of Mg/La2O3/graphene,first-principles calculations were conducted to calculate the ideal work of separation and electronic structure of the interfaces.Results demonstrated the presence of strong ionic and covalent interactions at the interface,which theoretically verified the strong interfacial bonding strength among Mg/La2O3/graphene interfaces.To experimentally validate the interface strength,MMCs with the interface structure of Mg/La2O3/GNS were developed.The formation of in-situ La2O3 led to the successful attainment of semi-coherent structures between Mg/La2O3 and La2O3/GNS,resulting in high strength and good ductility of the composite.Overall,this work proposes a new approach to interface design in MMCs with an enhancement of mechanical properties.

Barley Protein LFBEP-C1 from Lactiplantibacillus plantarum dy-1 Fermented Barley Extracts by Inhibiting Lipid Accumulation in a Caenorhabditis elegans Model

Objective This study aimed to investigate the lipid-lowering activity of LFBEP-C1 in high glucose-fed Caenorhabditis elegans(C.elegans).Methods In this study,the fermented barley protein LFBEP-C1 was prepared and tested for its potential anti-obesity effects on C.elegans.The worms were fed Escherichia coli OP50(E.coli OP50),glucose,and different concentrations of LFBEP-C1.Body size,lifespan,movement,triglyceride content,and gene expression were analyzed.The results were analyzed using ANOVA and Tukey's multiple comparison test.Results Compared with the model group,the head-swing frequency of C.elegans in the group of LFBEP-C1 at 20μg/mL increased by 33.88%,and the body-bending frequency increased by 27.09%.This indicated that LFBEP-C1 improved the locomotive ability of C.elegans.The average lifespan of C.elegans reached 13.55 days,and the body length and width of the C.elegans decreased after LFBEP-C1 intake.Additionally,LFBEP-C1 reduced the content of lipid accumulation and triglyceride levels.The expression levels of sbp-1,daf-2,and mdt-15 significantly decreased,while those of daf-16,tph-1,mod-1,and ser-4 significantly increased after LFBEP-C1 intake.Changes in these genes explain the signaling pathways that regulate lipid metabolism.Conclusion LFBEP-C1 significantly reduced lipid deposition in C.elegans fed a high-glucose diet and alleviated the adverse effects of a high-glucose diet on the development,lifespan,and exercise behavior of C.elegans.In addition,LFBEP-C1 regulated lipid metabolism mainly by mediating the expression of genes in the sterol regulatory element-binding protein,insulin,and 5-hydroxytryptamine signaling pathways.

Elevated CO_(2) increases soil redox potential by promoting root radial oxygen loss in paddy field

Soil redox potential(Eh)plays an important role in the biogeochemical cycling of soil nutrients.Whereas its effect soil process and nutrients'availability under elevated atmospheric CO_(2) concentration and warming has seldom been investigated.Thus,in this study,a field experiment was used to elucidate the effect of elevated CO_(2) concentration and warming on soil Eh,redox-sensitive elements and root radial oxygen loss(ROL).We hypothesized elevated CO_(2) and warming could alter soil Eh by promoting or inhibiting ROL.We found that soil Eh in the rhizosphere was significantly higher than that of non-rhizosphere.Elevated CO_(2) enhanced soil Eh by 11.5%,which corresponded to a significant decrease in soil Fe^(2+)and Mn^(2+)concentration.Under elevated CO_(2),the concentration of Fe^(2+)and Mn^(2+)decreased by 14.7%and 13.7%,respectively.We also found that elevated CO_(2) altered rice root aerenchyma structure and promoted rice root ROL.Under elevated CO_(2),rice root ROL increased by 79.5%and 112.2%for Yangdao 6 and Changyou 5,respectively.Warming had no effect on soil Eh and rice root ROL.While warming increased the concentration of Mn^(2+)and SO_(4)^(2-)by 4.9%and 19.3%,respectively.There was a significant interaction between elevated CO_(2) and warming on Fe^(2+)and Mn^(2+).Under elevated CO_(2),warming had no effect on the concentration of Fe^(2+)but decreased Mn^(2+)concentration significantly.Our study demonstrated that elevated atmospheric CO_(2) in the future could increase soil Eh by promoting rice root ROL,which will alter some soil nutrients'availability,such as Fe^(2+)and Mn^(2+).
