Peroxisome proliferator-activated receptorγ(PPARγ)is a transcriptional coactivator that binds to a diverse range of transcription ***γcoactivator 1(PGC-1)coactivators possess an extensive range of biological effects in different tissues,and play a key part in the regulation of the oxidative metabolism,consequently modulating the production of reactive oxygen species,autophagy,and mitochondrial *** to these findings,a large body of studies,aiming to establish the role of PGC-1 in the neuromuscular system,has shown that PGC-1 could be a promising target for therapies targeting neuromuscular *** these,some evidence has shown that various signaling pathways linked to PGC-1αare deregulated in muscular dystrophy,leading to a reduced capacity for mitochondrial oxidative phosphorylation and increased reactive oxygen species(ROS)*** the light of these results,any intervention aimed at activating PGC-1 could contribute towards ameliorating the progression of muscular ***-1αis influenced by different patho-physiological/pharmacological *** products have been reported to display modulatory effects on PPARγactivation with fewer side effects in comparison to synthetic *** together,this review summarizes the current knowledge on Duchenne muscular dystrophy,focusing on the potential effects of natural compounds,acting as regulators of PGC-1α.

Enhanced axonal regeneration and functional recovery of the injured sciatic nerve in a rat model by lithium-loaded electrospun nanofibrous scaffolds

Increasing evidence indicates that engineered nerve grafts have great potential for the regeneration of peripheral nerve injuries(PNIs).While most studies have focused only on the topographical features of the grafts,we have considered both the biophysical and biochemical manipulations in our applied *** achieve this,we fabricated an electrospun nanofibrous scaffold(ENS)containing polylactide nanofibers loaded with lithium(Li)ions,a Wnt/β-catenin signaling *** addition,we seeded human adipose-derived mesenchymal stem cells(hADMSCs)onto this engineered scaffold to examine if their differentiation toward Schwann-like cells was *** further examined the efficacy of the scaffolds for nerve regeneration in vivo via grafting in a PNI rat *** results showed that Li-loaded ENSs gradually released Li within 11 d,at concentrations ranging from 0.02 to(3.64±0.10)mmol/L,and upregulated the expression of Wnt/β-catenin target genes(cyclinD1 and c-Myc)as well as those of Schwann cell markers(growth-associated protein 43(GAP43),S100 calcium binding protein B(S100B),glial fibrillary acidic protein(GFAP),and SRY-box transcription factor 10(SOX10))in differentiated *** the PNI rat model,implantation of Li-loaded ENSs with/without cells improved behavioral features such as sensory and motor functions as well as the electrophysiological characteristics of the injured *** improved function was further validated by histological analysis of sciatic nerves grafted with Li-loaded ENSs,which showed no fibrous connective tissue but enhanced organized myelinated *** potential of Li-loaded ENSs in promoting Schwann cell differentiation of hADMSCs and axonal regeneration of injured sciatic nerves suggests their potential for application in peripheral nerve tissue engineering.

Predictable forward performance processes(PFPPs)are stochastic optimal control frameworks for an agent who controls a randomly evolving system but can only prescribe the system dynamics for a short period *** is a common scenario in which a controlling agent frequently re-calibrates her *** introduce a new class of PFPPs based on rank-dependent utility,generalizing existing models that are based on expected utility theory(EUT).We establish existence of rank-dependent PFPPs under a conditionally complete market and exogenous probability distortion functions which are updated *** show that their construction reduces to solving an integral equation that generalizes the integral equation obtained under EUT in previous *** then propose a new approach for solving the integral equation via theory of Volterra *** illustrate our result in the special case of conditionally complete Black-Scholes model.

AIM:To investigate the effect of MEK/ERK1/2 pathway on peroxisome proliferator-activated receptors(PPARγ)agonist-induced alterations in Δ6-desaturase(Δ6D)and stearoyl-CoA desaturase 1(SCD1)in hepatocellular carcinoma cell line ***:HepG2 cells cultured in RPMI-1640 were exposed to the commonly used ERK1/2 pathway inhibitor PD98059 and PPARγ agonist,*** RNA was isolated and reverse transcribed from treated *** in gene expression and metabolites ratio,as activity index for Δ6D and SCD1,were then determined using reverse transcriptionpolymerase chain reaction and gas liquid chromatography,***:The expression of both Δ6D(P = 0.03)and SCD1(P = 0.01)increased following PD98059 treatment,with a higher impact on SCD1(24.5%vs 62.5%).Although pioglitazone increased the mRNA level(1.47 ± 0.10 vs 0.88 ± 0.02,P = 0.006)and activity index(1.40 ± 0.07 vs 0.79 ± 0.11,P < 0.001)of Δ6D,no such changes have been observed for SCD1 activity index in pioglitazone-treated ***1 gene expression(+26.4%,P = 0.041)and activity index(+52.8%,P = 0.035)were significantly increased by MEK inhibition in the presence of pioglitazone,as compared with pioglitazone alone and control ***,the response of Δ6D expression and activity index to pioglitazone was unaffected by incubation with ***:PPARγ and ERK1/2 signaling pathway affect differentially and may have inhibitory crosstalk effects on the genes expression of 6D and SCD1,and subsequently on their enzymatic activities.

Hydrate Prevention Strategies and the Associated Cost in the Gulf of Mexico

With the petroleum industry endeavoring to develop promising oil and gas in deeper water, gas hydrates prevention is a serious concern for oil and gas producing companies producing at conditions in the hydrate region. This paper details lessons learned from the successful field deployment of AA LDHI and proper implementation strategies used for 3 different practical fields as case studies in the Gulf of Mexico. From the 3 field experiences, the AA LDHI has been used to replace the conventional thermodynamic hydrate inhibitor due to its numerous benefits during steady state operations and transition operations where AA LDHI is injected prior to extended shut in and restart for fields producing at low water cut. However, the strategy to develop a cost effective chemical management of hydrates for fields producing at high water cut is by pumping methanol or diesel to push down the wellbore fluid below the mud line during planned and unplanned shut-ins to delay water production, it also secures the riser with non hydrate fluids. This illustrates how the AA LDHIs are used in conjunction with more conventional hydrate management approaches to reach an optimal cost effective field hydrate management solution. However, this shows that the key to overall success of hydrate prevention is a full integration of a good front end design, a comprehensive deployment and an effective down hole monitoring system.
