An effective pharmacological hydrogel induces optic nerve repair and improves visual function

Irreversible eye lesions, such as glaucoma and traumatic optic neuropathy, can cause blindness;however, no effective treatments exist. The optic nerve, in particular, lacks the capacity to spontaneously regenerate, requiring the development of an effective approach for optic nerve repair, which has proven challenging. Here, we demonstrate that a combination of the small molecules 3BDO and trichostatin A(TSA)—which regulate mTOR and HDAC, respectively—packaged in thermosensitive hydrogel for 4-week-sustained release after intravitreal injection, effectively induced optic nerve regeneration in a mouse model of optic nerve crush injury. Moreover, this combination of 3BDO and TSA also protected axon projections and improved visual responses in an old mouse model(11 months old) of glaucoma. Taken together, our data provide a new, local small molecule-based treatment for the effective induction of optic nerve repair, which may represent a foundation for the development of pharmacological methods to treat irreversible eye diseases.

The emergence of the metaverse has ledto the rapidly increasing demand for the generation ofextensive 3D worlds. We consider that an engagingworld is built upon a rational layout of multiple landuse areas (e.g., forest, meadow, and farmland). Tothis end, we propose a generative model of landuse distribution that learns from geographic *** model is based on a transformer architecturethat generates a 2D map of the land-use layout,which can be conditioned on spatial and semanticcontrols, depending on whether either one or bothare provided. This model enables diverse layoutgeneration with user control and layout expansion byextending borders with partial inputs. To generatehigh-quality and satisfactory layouts, we devise ageometric objective function that supervises the modelto perceive layout shapes and regularize generationsusing geometric priors. Additionally, we devise aplanning objective function that supervises the modelto perceive progressive composition demands andsuppress generations deviating from controls. Toevaluate the spatial distribution of the generations, wetrain an autoencoder to embed land-use layouts intovectors to enable comparison between the real andgenerated data using the Wasserstein metric, which isinspired by the Fr´echet inception distance.

The yield ratios of neutron-proton(R(n/p))and^(3)H-^(3)He(R(^(3)H∕^(3)He))with reduced rapidity from 0 to 0.5 were simulated at 50 MeV/u even-even ^(36−56)Ca+^(40)Ca,even-even ^(48−78)Ni+^(58)Ni,and ^(100−139)Sn(every third isotopes)+112 Sn for full reduced impact parameters using the isospin-dependent quantum molecular dynamics(IQMD)*** neutron and proton density distributions and root-mean-square radii of the reaction systems were obtained using the Skyrme-Hartree-Fock model,which was used for the phase space initialization of the projectile and target in *** defined the unified neutron skin thickness asΔRnp=^(1∕2) n−^(1∕2)p,which was negative for neutron-deficient *** unifiedΔRnp values for nuclei with the same relative neutron excess from different isotopic chains were nearly equal,except for extreme neutron-rich isotopes,which is a type of scaling *** yield ratios of the three isotopic chain-induced reactions,which depended on the reduced impact parameter and unified neutron skin thickness,were *** results showed that both R(n/p)and R(^(3)H∕^(3)He)decreased with a reduced impact parameter for extreme neutron-deficient isotopes;however,they increased with reduced impact parameters for extreme neutron-rich isotopes,and increased with theΔRnp of the projectiles for all reduced impact *** addition,a scaling phenomenon was observed betweenΔR np and the yield ratios in peripheral colli-sions from different isotopic chain projectiles(except for extreme neutron-rich isotopes).Thus,R(n/p)and R(^(3)H∕^(3)He)from peripheral collisions were suggested as experimental probes for extracting the neutron or proton skin thicknesses of non-extreme neutron-rich nuclei from different isotopic chains.

Background Endometrial carcinoma is one of the most common female tract genital malignant tumors. Nifedipine, an L-type calcium channel antagonist can inhibit cell proliferation of carcinomas. Recent studies indicated that a rise in the free cytosolic calcium （[Ca2±]c） was a potent inducer of autophagy. Here, we investigated the relationship between nifedipine and autophagy in Hec-IA cells. Methods Cells were cultured with nifedipine （10 μmol/L） and harvested at different times for counting cell number. MTT assay was applied to evaluate the cell viability and transwell assay to reveal cell migration. Apoptotic cells were detected with annexin V/PI assay. Then cells were treated with 3-methyladenine （3-MA）（2.5 mmol/L） for 0, 5, 15, 30, 60, and 120 minutes and the expression of the L-type calcium channel alphalD （Cavl.3） protein was detected. At last, cells were cultured and assigned to four groups with different treatment： untreated （control group）, 10 μmol/L nifedipine （N group）, 2.5 mmol/L 3-MA （3-MA group）, and 10 μmol/L nifedipine plus 2.5 mmol/L 3-MA （N±3MA group）. Autophagy was detected with GFP-LC3 modulation by fluorescent microscopy, and expression of the autophagy-associated proteins （LC3, Beclinl and P70s6K） by Western blotting and monodansylcadaverine （MDC） labeled visualization. Results Proliferation of Hec-lA cells was obviously suppressed by nifedipine compared with that of the untreated cells for 24, 48, and 96 hours （P=0.000 for each day）. The suppression of migration ability of the nifedipine-treated cells （94.0±8.2） was significantly different from that of the untreated cells （160.00±9.50, P=0.021 ）. The level of early period cell apoptosis induced by nifedipine was （2.21_±0.19）%, which was （2.90±0.13）% in control group （P=-0.052）, whereas the late period apoptosis level reached （10.38_±0.96）% and （4.40_±0.60）% （P=0.020）, respectively. The 3-MA group induced a slight increase in the Cavl.3 levels within 15 minutes, but significantly attenuated the

In this study,macro-and meso-mechanical properties of frozen sand molds were discussed based on the Hertz-Mindlin with Bonding(HMB)***-burman,steepest ascent,and central composite designs were utilized to propose a parameter calibration *** effects of mesoscopic parameter variations on the compressive strength and average gradient of stress-strain were investigated through response surface method *** showed that the relative error between the simulated and measured repose angle is 3.1%under calibrated intrinsic contact *** compressive strength and average stress-strain gradient primarily depend on the normal and shear stiffness per unit area,as well as the particle size and porosity of the silica ***,taking load-displacement curves of three frozen sand molds with different geometric characteristics as the target value,the reliability and effectiveness of the frozen sand mold HMB model were verified through uniaxial compression tests and discrete element simulations.

Relative positioning is recognized as an important issue for vehicles in urban ***-vehicle Cooperative Positioning(CP)techniques which fuse the Global Navigation Satellite System(GNSS)and inter-vehicle ranging have attracted attention in improving the performance of baseline estimation between ***,current CP methods estimate the baselines separately and ignore the interactions among the positioning information of different *** interactions are called’information coupling’.In this work,we propose a new multivehicle precise CP framework using the coupled information in the network based on the Carrier Differential GNSS(CDGNSS)and inter-vehicle *** demonstrate the benefit of the coupled information by deriving the Cramer-Rao Lower Bound(CRLB)of the float estimation in *** fully use this coupled information,we propose a Whole-Net CP(WN-CP)method which consists of the Whole-Net Extended Kalman Filter(WN-EKF)as the float estimation filter,and the Partial Baseline Fixing(PBF)as the ambiguity resolution *** WN-EKF fuses the measurements of all baselines simultaneously to improve the performance of float estimation,and the PBF strategy fixes the ambiguities of the one baseline to be estimated,instead of full ambiguity resolution,to reduce the computation load of ambiguity *** tests involving four vehicles were conducted in urban *** results show that the proposed WN-CP method can achieve better performance and meanwhile maintain a low computation load compared to the existing methods.

BACKGROUND Duodenal varices are a lesser-known complication with non-cirrhotic portal hypertension. We report a circuitous route from missed diagnosis of duodenal varices to correction. An extremely rare case of duodenal variceal bleeding secondary to idiopathic portal hypertension(IPH) is expounded in this study, which was controlled by transjugular intra-hepatic porto-systemic shunt(TIPS) plus embolization. CASE SUMMARY A 46-year-old woman with anemia for two years was frequently admitted to the local hospital. Upon examination, anemia was attributed to gastrointestinal tract bleeding, which resulted from duodenal variceal bleeding detected by repeated esophagogastroduodenoscopy. At the end of a complete workup, IPH leadingto duodenal varices was diagnosed. Portal venography revealed that the remarked duodenal varices originated from the proximal superior mesenteric vein. TIPS plus embolization with coils and Histoacryl was performed to obliterate the rupture of duodenal varices. The anemia resolved, and the duodenal varices completely vanished by 2 mo after the initial operation. CONCLUSION TIPS plus embolization may be more appropriate to treat the bleeding of large duodenal varices.
