Silvestrol alleviates glioblastoma progression through ERK pathway modulation and MANBA and NRG-1 expression

Background:Glioblastoma,a notably malignant tumor within the central nervous system,is distinguished by its aggressive ***,a robust inhibitor of the RNA helicase eukaryotic initiation factor 4A(eIF4A),has shown significant potential as an anticancer ***,the impact of silvestrol on glioblastoma,especially its molecular mechanisms,has not been fully ***:This investigation employed a variety of in vitro assays,such as cell counting kit-8(CCK-8),clonogenic,5-ethynyl-2′-deoxyuridine(EDU),wound healing,and flow cytometry,to evaluate cell cycle progression,apoptosis,cell viability,and *** blot analysis was also performed to study the apoptosis and extracellular regulated kinase(ERK)*** the ERK pathway was inhibited,differentially expressed genes(DEGs)in U87 cells were identified,followed by an analysis of target genes using the gene expression profiling interactive analysis(GEPIA)***:Silvestrol significantly suppressed the proliferation,migration,and colony formation of glioma *** caused cell cycle arrest and enhanced apoptosis in these ***,silvestrol stimulated the ERK pathway,with these effects being reversible by an ERK phosphorylation *** combined with GEPIA,GSCA,UALCAN,TIMER database screened 4 potential drug targets of silvestrol:chromosome 1 open reading frame 226(C1ORF226),mannosidase beta A(MANBA),IQ motif and Sec7 domain 2(IQSEC2),neuregulin 1(NRG-1).Among them,C1ORF226 was lower risk gene while MANBA,IQSEC2,and NRG-1 were high-risk ***,silvestrol notably reduced MANBA mRNA levels,which could be reversed by inhibiting ERK ***,silvestrol markedly decreased NRG-1 protein levels,with an additional reduction observed when the ERK pathway was ***:Silvestrol’s anti-glioma effects are primarily due to the suppression of MANBA expression via the ERK pathway and possibly by hindering the translation of N

Bcklund Transformation and Conservation Laws for the Variable-coefficient N-Coupled Nonlinear Schrdinger Equations with Symbolic Computation

Considering the integrable properties for the coupled equations, the variable-coefficient N- coupled nonlinear Schrodinger equations are under investigation analytically in this paper. Based on the Lax pair with the nonisospectral parameter, a Backlund transformation for such a coupled system denoting in the F functions is constructed with the one-solitonic solution given as the application sample. Furthermore, an infinite number of conservation laws are obtained using symbolic computation.

Exact Periodic and Solitary-Wave Solutions of Multi-component Modified Korteweg-de Vries Equations

The present work extends the search of Jacobi elliptic function solutions for the multi-component modified Korteweg-de Vries equations. When the modulum m →1, those periodic solutions degenerate as the corresponding solitary wave and shock wave ones. Especially, exact solutions for the three-component system are presented in detail and graphically.

Direct Approach to Construct the Periodic Wave Solutions for Two Nonlinear Evolution Equations

With symbolic computation, the Hirota method and Riemann theta function are employed to directly construct the periodic wave solutions for the Hirota-Satsuma equation for shallow water waves and boiti-Leon-Manna- Pempinelli equation. Then, the corresponding figures of the periodic wave solutions are given. Fhrthermore, it is shown that the known soliton solutions can be reduced from the periodic wave solutions.

Effect of Monomer Feeding Mode on the Preparation of Hollow Latexes with High MAA Content in the Core Latex Preparation

In order to prepare hollow latex particles with optimum morphology based on osmotic swelling principle, three- layer core/shell latex particles with 40 wt% MAA in the core were first prepared via multistep seeded emulsion copolymerization, in which monomers were added by a semi-continuous process with monomer addition under two different forms： pure monomers＇ mixture （monomer addition）, and pre-emulsified monomers （pre-emulsion addition）. Then, the hollow latex particles with different morphologies were obtained after alkali post-treatment. Influences of the monomer feeding mode on the emulsion polymerization and the particle morphology were investigated. Results showed that the pre- emulsion addition could significantly improve the polymerization stability in each step, and greatly enhance the uniformity of shell encapsulation. The sizes of the core and core/shell latex particles obtained by the pre-emulsion addition were smaller and more uniform than those synthesized by the monomer addition, and the hollow latex particles with intact morphology were generated by alkali post-treating of the core/shell latexes prepared from the pre-emulsion addition. As the core size increased, the morphology of the post-treated particles underwent evolution from hollow to collapse. Moreover, the mechanism of the particle morphological evolution was proposed.

Effect of the Shell Crosslinking Level and Core/Shell Ratio on the Morphology of Latex Particles in the Preparation of Hollow Latexes

Three-layer core/shell latex particles with various shell crosslinking level and shell thickness were prepared by multistep emulsion polymerization, and the hollow latex particles with different morphologies were then obtained after alkali post-treatment. Influences of divinyl benzene （DVB） content and the core/shell mass ratio on emulsion polymerization and particle morphology were investigated. Results showed that with the increase of DVB content, the percentage of total amount of --COOH on the particle surface and free in aqueous phase （PSFa） decreased, and the morphology of the post-treated particles underwent evolution from cracked, intact hollow to deficient swelling structure. Decreasing the core/shell mass ratio could not only make more carboxyl groups encapsulated by the shell, but also increase the shell resistance to the swelling of the core. The uniform hollow latex particles with intact morphology were obtained when the DVB content was 3.54 wt% and the core/shell mass ratio was 1/6.

Spurious symmetry-broken phase in a bidirectional two-lane ASEP with narrow entrances

As one of the paradigmatic models of non-equilibrium systems, the asymmetric simple exclusion process（ASEP） has been widely used to study many physical, chemical, and biological systems. The ASEP shows a range of nontrivial macroscopic phenomena, among which, the spontaneous symmetry breaking has gained a great deal of attention. Nevertheless,as a basic problem, it has been controversial whether there exist one or two symmetry-broken phases in the ASEP. Based on the mean field analysis and current minimization principle, this paper demonstrates that one of the broken-symmetry phases does not exist in a bidirectional two-lane ASEP with narrow entrances. Moreover, an exponential decay feature is observed,which has been used to predict the phase boundary in the thermodynamic limit. Our findings might be generalized to other ASEP models and thus deepen the understanding of the spontaneous symmetry breaking in non-equilibrium systems.

Bilinear Forms and Soliton Solutions for the Reduced Maxwell-Bloch Equations with Variable Coefficients in Nonlinear Optics

Investigation in this paper is given to the reduced Maxwell-Bloeh equations with variable coetcients, describing the propagation of the intense ultra-short optical pulses through an inhomogeneous two-level dielectric medium. We apply the Hirota method and symbolic computation to study such equations. With the help of the dependent variable transformations, we present the variable-coetteient-dependent bilinear forms. Then, we construct the one-, two- and N- soliton solutions in analytic forms for them.

Dynamics-based optimization of rolling schedule aiming at dual goals of chatter suppression and speed increase for a 5-stand cold tandem rolling mill

In the process of cold tandem rolling,chatter instability leads to serious impacts on enhancing rolling speed,improving product quality,reducing production cost and realizing *** occurs with the rolling speed up to a certain threshold value,but the critical speed is determined by both product specifications and rolling schedules.A 5-stand cold tandem rolling mill whose first three stands and subsequent two stands,respectively,have four and six rolls was investigated by formulating its dynamic equations with the corresponding structure-process *** applying the stability-based calculation model about the critical rolling speed in each stand,the system dynamic responses around the critical rolling speed were simulated,and the system eigenvalues which represent instability and characteristic frequencies were figured ***,via combining the critical rolling speeds with the system dynamic behavior,a dynamics-based optimization model of rolling schedule for the 5-stand cold tandem system was proposed for the purposes of both the chatter suppression and rolling speed *** the optimization model,eight rolling technique parameters(four strip thicknesses and four tensions between the upstream and downstream stands)were taken as design variables,and the constraint conditions were set as no chatter instability in all five stands,and the optimization goal was to maximize the outlet speed of the final *** pattern search method was introduced to solve the optimization *** applying such a dynamics-based optimization model for the 5-stand cold tandem rolling process,the chatter instability was suppressed effectively and the rolling efficiency was improved considerably;therefore,such an optimization model is expected to be valuable for intelligent manufacturing of rolling process.

Nervonic acid(NA)is a very-long-chain monounsaturated fatty acid with pharmaceutical and nutraceutical functions that plays an important role in treating several neurological *** major source of NA is plant seed *** we report fatty acid profiles of seeds and germplasm diversity of six plant species,including three woody plants with high amounts of NA-enriched seed oil,Malania oleifera,Macaranga adenantha,and *** had the largest seed(average 7.40 g single seed),highest oil content(58.71%),and highest NA level(42.22%).The germplasm diversity of *** is associated with its habitat but not *** of *** contained higher NA levels(28.41%)than ***(21.77%),but *** contained a significantly higher oil content(29.22%)and seed *** germplasm varied among populations,with one population having seeds with high oil content(22.63%)and NA level(37.78%).Although *** grow naturally at a range of elevations,no significant differences were detected between *** *** results suggest that *** and *** have greater potential as a source of NA,which will contribute to constructing a germplasm resource nursery and establishing a selection and breeding program to improve the development of NA-enriched plants.
