In order to investigate the reaction mechanism of intramolecular C-H insertion by Rh(II)-mediated carbenoids with trans-(2-phenylcyclopropyl) carbinal radical as the mechanistic probe, diazo compounds 2-(2-phenylcyclopropyl)ethyl diazoacetoacetate 8a and methyl 2-diazo-3oxo-6-(2-phenylcyclopropyl) hexanoate 8b have been synthesized. Preliminary investigation of the intramolecular C-H insertion with Rh2(OAc)4 as catalyst supports a concerted insertion process.

AIM: To analyze the 5-year disease-free survival(DFS) of primary orbital lymphoma(POL) by clinical characteristics and imaging ***: A total of 72 patients, 43 males and 29 females, with histologically confirmed POL, were retrospectively recruited between January 2012 and May 2017. The information on clinical characteristics, imaging features, and 5-year DFS was obtained. Univariate and multivariate forward logistic regression analyses were used to identify the variables significantly associated with 5-year DFS. Kaplan-Meier was applied for survival analysis. RESULTS: Univariate analysis revealed that uni-or bilateral orbital involvement, single or multiple lesions, treatment methods, and contrast enhancement pattern on images were significant for 5-year DFS(P=0.022, 0.042, <0.001, and 0.028, respectively), while in multivariate logistic regression analysis, only uni-or bilateral orbital involvement, treatment methods and contrast enhancement pattern on images were significant(r=0.453, 0.897, and 0.556, P=0.038, <0.001 and 0.022, respectively). The survival curves for DFS were obtained. CONCLUSION: The majority of POL are B-cell lymphomas. Unilateral orbital involvement, homogeneous contrast enhancement on images, and the appropriate treatment schemes result to be significant factors for a good prognosis for POL.

Huang-Qin Decoction(HQD)is a classic prescription for diarrhea in Chinese medicine *** studies have demonstrated that HQD and its modified formulation PHY906 could ameliorate irinotecan(CPT-11)induced gastrointestinal(GI)toxicity and enhance its anticancer therapeutic ***,which constituents in HQD are effective is still unclear so *** study aims to screen out the key bioactive components combination from HQD that could enhance the anticancer effect of ***,the potential bioactive constituents were obtained through system pharmacology *** the bioactivity of each constituent was investigated synthetically from the aspects of NCM460 cell migration,TNF-αrelease of THP-1-derived macrophage and MTT assay in HCT116 *** contribution of each constituent in HQD was evaluated using the bioactive index Ei,which taken the content and bioactivity into comprehensive *** then,the most contributing constituents were selected out to form a keycomponent *** last,the bioefficacy of the key-component combination was validated in vitro and in *** a result,a key-component combination(HB4)consisting of four compounds baicalin,baicalein,glycyrrhizic acid and wogonin was screened *** vitro assessment indicated that HB4 could enhance the effect of CPT-11 on inhibiting cell proliferation and inducing apoptosis in ***,the in vivo study confirmed that HB4 and HQD have similar pharmacological activity and could both enhance the antitumor effect of CPT-11 in HCT116 xenograft ***,HB4 could also reduce the CPT-11 induced GI toxicity.

Cerium oxalate hydrate particles were synthesized by precipitation technique under external magnetic field with respective intensities of 0,0.1,0.3,and 0.6 *** particle morphologies were investigated by scanning electron microscopy(SEM)and transmission electron microscope(TEM).Results show that external magnetic field has a great influence on the growth behavior of cerium oxalate hydrate ***-like and plate-like cerium oxalate hydrate particles with smaller size are formed without the applying of external magnetic ***,the extremely big rod-like particles with the average size of 12.47μm were prepared under external magnetic field of 0.6 *** increasing magnetic field intensity makes the grain grow *** analysis supports the presence of 8.5 H_(2)O molecules associated with cerium ***,the growth mechanism of cerium oxalate hydrate under external magnetic field was briefly discussed.

3D-QSAR and Docking Studies of 1,3,4-Thiazolidinone Derivatives Using R-Group Search and Surflex-dock

In this paper, a three-dimensional quantitative structure-activity relationships(3 D-QSAR) study for 20 HIV-1 reverse transcriptase(RT) inhibitors was established using Topomer Co MFA. The models were built based on different fragment cutting models, with the most effective model of the multiple correlation coefficients of fitting(r^2) to be 0.920, cross-validation(q^2) of 0.575, and external validation(Q_(ext)~2) being 0.701. The results indicated that the model obtained has both favorable estimation stability and good prediction capability. Topomer Search was used to search R-group from ZINC database. As a result, a series of R-groups with relatively high activity contribution was obtained. By No. 6 molecule filtering, 3 R_1 and 15 R_2 groups were selected, and employed to alternately substitute for the R_1 and R_2 of sample 6. Finally, 45 new compounds were designed, and the Topomer CoMFA model was used to predicate the biological activity, so the Topomer Search is effective in screening and can guide the design of new HIV/AIDS drugs. The molecular docking method was also used to study the interactions of these drugs by docking the ligands into HIV-1 RT active site, which revealed the likely bioactive conformations. This study showed that there are extensive interactions between the 1,3,4-thiazolidinone revertase inhibitors and His84, Asp145, Lys33 and Leu83 residues in the active site of HIV-1 RT. These results provide useful insights for the design of potent new inhibitors of RT.

A 3D-QSAR Study of HIV-1 Integrase Inhibitors Using RASMS and Topomer CoMFA

Acquired Immunodeficiency Syndrome(AIDS) is a significant human health threat around the world. Therefore, the study of anti-human immunodeficiency virus(HIV) drug design has become an important task for today’s society. In this paper, a three-dimensional quantitative structure-activity relationships study(3 D-QSAR) was conducted on 53 HIV-1 integrase inhibitors(IN) using random sampling analysis on molecular surface(RASMS) and Topomer comparative molecular field analysis(Topomer CoMFA). The multiple correlation coefficients of fitting, cross-validation, and external validation of two models were 0.926, 0.815 and 0.908 and 0.930, 0.726 and 0.855, respectively. The results indicated that two models obtained had both favorable estimation stability and good prediction capability. Topomer Search was used to search appropriate R groups from ZINC database, and 28 new compounds were designed thereby. The Topomer CoMFA model was subsequently used to predict the biological activity of these compounds, showing that 24 of the new compounds were more active than the template molecule. Ligands of the template molecule and new designed compounds were used for molecular docking to study the interaction of these compounds with the protein receptor. The results show that the ligands would form hydrogen-bonding interactions with the residues LEU58, THR83, GLN62, MET155, LYS119 and ALA154 of the protein receptor generally, thereby providing additional insights for the design of even more effective HIV/AIDS drugs.
