目的探讨环磷腺苷效应元件结合蛋白1(cAMP-response element binding protein 1,CREB1)基因沉默对乳腺癌细胞MCF-7和MDA-MB-231细胞增殖、凋亡、迁移和侵袭的影响。方法针对人CREB1的基因序列设计并构建2条短发夹RNA(short hairpin RNA,shRNA),采用慢病毒转染shRNA至人乳腺癌MCF-7和MDA-MB-231细胞系抑制其CREB1的表达。将实验组分为shCREB1#1组和shCREB1#2组,同时将shSCR空载质粒转染至上述细胞系作为阴性对照组。采用实时定量PCR和Western印迹法检测转染效率;CCK-8法检测细胞增殖能力;集落形成实验检测细胞集落形成能力;流式细胞术检测细胞周期和凋亡率;细胞划痕实验和Transwell实验检测细胞迁移和侵袭能力;Western印迹法检测细胞周期及细胞凋亡相关蛋白的表达。结果相较于阴性对照组,shCREB1#1和shCREB1#2组MCF-7和MDA-MB-231细胞CREB1基因的mRNA和蛋白表达水平均下降(P<0.001)。沉默CREB1后,MCF-7和MDA-MB-231细胞的增殖能力、集落形成能力、迁移和侵袭能力减弱,且凋亡率升高(P<0.05)。沉默CREB1使细胞周期蛋白CDK2、CDK4、CDK6、Cyclin D1以及抗凋亡蛋白Bcl-2、Survivin的表达下调,而使促凋亡蛋白Caspase 3和Bax的表达上调(P<0.05)。结论沉默CREB1可抑制乳腺癌细胞的增殖、迁移和侵袭能力,并诱导其凋亡。

Solid-state reactions are an essential part of the synthesis of various cathode materials for lithium-ion batteries(LIBs).Despite the simplicity and effectiveness for mass production,a subtle variation in synthesis conditions can often give rise to unexpectedly different physical properties,significantly affecting the electrochemical performance of electrode materials.However,this aspect has long been overlooked in the LIB community,which should focus on advancement in understanding the influence of synthesis conditions.As solid-state reactions occur only at the interface of precursor materials,maximizing the interfacial contact area between mixed precursor powders is crucial.Mechanical milling and/or mixing are common practices that have been performed in both academia and industry for this purpose.Unlike the common belief that this pre-treatment before calcination would be of great benefit for the preparation of high-performance LIB materials,we revealed in this work that this practice is not always successful for this goal.In our case study of the preparation of LiCoO_(2),we demonstrated that mechanical mixing-a routinely implemented process for homogeneous mixing of precursors-can be harmful if it is performed without assuring optimal working conditions for mixing.The underlying reasons for this surprising result are elucidated in this work,and we hope that this new insight can help avoid the potential pitfall of routine implementations performed for LIB materials.

Let x:M→S^(n+p)(1)be an n-dimensional submanifold immersed in an(n+p)-dimensional unit sphere S^(n+p)(1).In this paper,we study n-dimensional submanifolds immersed in S^(n+p)(1)which are critical points of the functional S(x)=∫_(M)S^(n/2)dv,where S is the squared length of the second fundamental form of the immersion x.When x:M→S^(2+p)(1)is a surface in S^(2+p)(1),the functional S(x)=∫_(M)S^(n/2)dv represents double volume of image of Gaussian map.For the critical surface of S(x),we get a relationship between the integral of an extrinsic quantity of the surface and its Euler characteristic.Furthermore,we establish a rigidity theorem for the critical surface of S(x).

Conductive cementitious composites are innovated materials that have improved electrical conductivity compared to general types of cement,and are expected to be used in a variety of future infrastructures with unique functionalities such as self-heating,electromagnetic shielding,and piezoelectricity.In the present study,machine learning methods that have been recently applied in various fields were proposed for the prediction of piezoelectric characteristics of carbon nanotubes(CNTs)-incorporated cement composites.Data on the resistivity change of CNTs/cement composites according to various water/binder ratios,loading types,and CNT content were considered as training values.These data were applied to numerous machine learning techniques including linear regression,decision tree,support vector machine,deep belief network,Gaussian process regression,genetic algorithm,bagging ensemble,random forest ensemble,boosting ensemble,long short-term memory,and gated recurrent units to estimate the time-independent and-dependent electrical properties of conductive cementitious composites.By comparing and analyzing the computed results of the proposed methods,an optimal algorithm suitable for application to CNTs-embedded cementitious composites was derived.

Yunnan is exceedingly rich in higher fungi(Ascomycota and Basidiomycota). Given that the number of fungi(including lichens) occurring in a given area is, as Hawksworth suggested, roughly six times that of local vascular plants, a total of approximately 104,000 fungal species would be expected in Yunnan.However, to date only about 6000 fungal species, including roughly 3000 species of higher fungi, have been reported from the province. Although studies on Yunnan's fungi started in the late nineteenth century, significant progress has been made only in the last forty-five years. Over the first twenty-five years of this period, studies on fungal diversity in this area have largely been about taxonomy based on morphological characters and partially on geographical distribution. Over the past twenty years, the combination of both morphological and molecular phylogenetic approaches has become the preferred method to help understand the diversity and evolution of higher fungi. This review focuses on our current knowledge of how geological, geographical, and ecological factors may have contributed to the diversity patterns of higher fungi in Yunnan. Based on this knowledge, three aspects for future studies are suggested.

BACKGROUND Immunotherapy for advanced gastric cancer has attracted widespread attention in recent years.However,the adverse reactions of immunotherapy and its relationship with patient prognosis still need further study.In order to determine the association between adverse reaction factors and prognosis,the aim of this study was to conduct a systematic prognostic analysis.By comprehensively evaluating the clinical data of patients with advanced gastric cancer treated by immunotherapy,a nomogram model will be established to predict the survival status of patients more accurately.AIM To explore the characteristics and predictors of immune-related adverse reactions(irAEs)in advanced gastric cancer patients receiving immunotherapy with programmed death protein-1(PD-1)inhibitors and to analyze the correlation between irAEs and patient prognosis.METHODS A total of 140 patients with advanced gastric cancer who were treated with PD-1 inhibitors in our hospital from June 2021 to October 2023 were selected.Patients were divided into the irAEs group and the non-irAEs group according to whether or not irAEs occurred.Clinical features,manifestations,and prognosis of irAEs in the two groups were collected and analyzed.A multivariate logistic regression model was used to analyze the related factors affecting the occurrence of irAEs,and the prediction model of irAEs was established.The receiver operating characteristic(ROC)curve was used to evaluate the ability of different indicators to predict irAEs.A Kaplan-Meier survival curve was used to analyze the correlation between irAEs and prognosis.The Cox proportional risk model was used to analyze the related factors affecting the prognosis of patients.RESULTS A total of 132 patients were followed up,of whom 63(47.7%)developed irAEs.We looked at the two groups’clinical features and found that the two groups were statistically different in age≥65 years,Ki-67 index,white blood cell count,neutrophil count,and regulatory T cell(Treg)count(all P<0.05).Multivariate logistic regression an

Orbital period and multi-color light curves’investigation of OW Leo are presented for the first time.The orbital period of OW Leo is corrected from P=0.325545 days to P=0.32554052 days in our work,and the observational data from the All-Sky Automated Survey for Super Novae(ASAS-SN)are used to test the newly determined orbital period.Then,the phased light curves are calculated with the new period and the Wilson-Devinney program is applied to model the light curves,which reveal that OW Leo is a W-subtype shallow contact binary system(q=3.05,f=12.8%).The absolute physical parameters of the two component stars are estimated to be M_(1)=0.31(1)M_(⊙),M_(2)=0.95(3)M_(⊙),R_(1)=0.63(1)R⊙,R_(2)=1.04(1)R_(⊙),L_(1)=0.43(1)L_(⊙)and L_(2)=1.01(2)L_(⊙).The evolutionary status shows that the more massive star is less evolved than the less massive star.OW Leo has very low metal abundance,which means its formation and evolution are hardly influenced by any additional component.It is formed from an initially detached binary system through nuclear evolution and angular momentum loss via magnetic braking,and has passed a very long time of main sequence evolution.

Shear failure of slender reinforced concrete beams without stirrups has surely been a complicated occurrence that has proven challenging to adequately understand.The primary purpose of this work is to develop machine learning models capable of reliably predicting the shear strength of non-shear-reinforced slender beams(SB).A database encompassing 1118 experimental findings from the relevant literature was compiled,containing eight distinct factors.Gradient Boosting(GB)technique was developed and evaluated in combination with three different optimization algorithms,namely Particle Swarm Optimization(PSO),Random Annealing Optimization(RA),and Simulated Annealing Optimization(SA).The findings suggested that GB-SA could deliver strong prediction results and effectively generalizes the connection between the input and output variables.Shap values and two-dimensional PDP analysis were then carried out.Engineers may use the findings in this work to define beam’s geometrical components and material used to achieve the desired shear strength of SB without reinforcement.
