Genetic effects conferring heat tolerance in upland cotton(Gossypium hirsutum L.)

Background:Climate change and particularly global warming has emerged as an alarming threat to the crop productivity of field crops and exerted drastic effects on the cropping *** of cotton has been dropped down to one million bales from 1.4 million bales since 2015 in Pakistan due to the increase in temperature at critical growth stages,i.e.,flowering and boll *** in view the importance of cotton in the country,this study was conducted to investigate the genetic effects conferring heat tolerance in six populations(P1,P2,F1,F2,BC1 and BC2)developed from cross-1 and cross-2,i.e.,VH-282×FH-142 and DNH-40×***:The results revealed that cross-1 performed better in heat stress as compared with cross-2 for majority of the traits *** weight and ginning outturn(GO-T)were highly effected under heat stress and had negative correlation with Relative cell injury(RCI).Boll weight,fiber length,fiber strength and fiber fineness were under the control of non-additive gene action,whereas RCI was controlled by additive gene *** values of genetic advance coupled with higher values of broad sense heritability for these traits except RCI confirmed the role of non-additive genetic *** types of epistasis were recorded for fiber strength in cross-1 in normal ***,complementary type of non-allelic interaction was recorded for fiber strength under normal condition,fiber fineness and RCI under heat stressed condition in ***,boll weight,GOT and fiber length in populations derived from cross-2 in normal condition were also under the influence of complementary type of non-allelic *** differences among values of mid parent and better parent heterosis for boll weight in both normal and heat stress condition provided the opportunity to cotton breeders for utilization of this germplasm for improvement of this trait through exploitation of heterosis ***:Cross-1 perfo

INTRODUCTION The past two decades have witnessed one of the most contentious scientific debates in the field of temperature management after cardiac arrest in *** started in 2002,following the publication of two groundbreaking trials showing lower mortality rates and improved neurologic recovery with the use of active cooling.[1,2]International recommendations advocate active cooling in the range of 32–34℃for comatose survivors of out-of-hospital cardiac arrest (OHCA).[3,4]The year2013 marked the tipping point with the release of the TTM 1 trial.

A novel electrochemical sensor was fabricated by electrodeposition of gold nanoparticles on a poly(L-methionine)(PMT)-modified glassy carbon electrode(GCE) to form a nano-Au/PMT composite-modified GCE(nano-Au/PMT/GCE).Scanning electron microscopy and electrochemical techniques were used to characterize the composite *** modified electrode exhibited considerable electrocatalytic activity towards the oxidation of dopamine(DA) and uric acid(UA) in phosphate buffer solution(pH = 7.00).Differential pulse voltammetry revealed that the electrocatalytic oxidation currents of DA and UA were linearly related to concentration over the range of 5.0 ×10–8 to 10–6 mol/L for DA and 7.0 × 10–8 to 10–6 mol/L for *** detection limits were 3.7 × 10–8mol/L for DA and 4.5 × 10–8 mol/L for UA at a signal-to-noise ratio of *** to our experimental results,nano-Au/PMT/GCE can be used as a sensitive and selective sensor for simultaneous determination of DA and UA.

Barley chitinase genes expression revamp resistance against whitefly (Bemisia Tabaci) in transgenic cotton (Gossypium hirsutum L.)

Background Chitinase is an enzyme that hydrolyzes chitin,a major component of the exoskeleton of insects,including plant pests like *** present study aimed to investigate the expression of chemically synthesized barley ch1 and chi2 genes in cotton(Gossypium hirsutum)through Agrobacterium-mediated ***-five putative transgenic cotton plants were obtained,out of which fifteen plants successfully survived and were shifted to the *** gene-specific primers,amplification of 447 bp and 401 bp fragments confirmed the presence of the ch1 and chi2 genes in five transgenic cotton plants of the T0 *** five plants were further evalu-ated for their mRNA expression *** T0 transgenic cotton plants with the highest mRNA expression level and better yield performance in field,were selected to raise their subsequent *** The T1 cotton plants showed the highest mRNA expression levels of 3.5-fold in P10(2)for the ch1 gene and 3.7-fold in P2(1)for the chi2 *** in situ hybridization(FISH)confirmed a single copy number of ch1 and chi2(hemizygous)on chromosome ***,the efficacy of transgenes on whitefly was evaluated through an insect bioassay,where after 96 h of infestation,mortality rates of whitefly were calculated to be 78%–80%in transgenic cotton *** number of eggs on transgenic cotton plants were calculated to be 0.1%–0.12 per plant compared with the non-transgenic plants where egg number was calculated to be 0.90–1.00 per *** Based on these findings,it can be concluded that the chemically synthesized barley chitinase genes(ch1 and chi2)have the potential to be effective against insects with chitin exoskeletons,including *** transgenic cotton plants expressing these genes showed increased resistance to whiteflies,resulting in reduced egg numbers and higher mortality rates.

Three-Dimensional Analysis of Buried Steel Pipes under Moving Loads

The three-dimensional response of buried steel pipes under vehicle loads is investigated using the finite element analysis. The analysis is conducted using the finite element program ABAQUS. The effects of the vehicle parameters, pipeline parameters and soil parameters on the response of the buried pipeline were discussed. The results indicate that the maximum principal stresses in a buried pipe under vehicle loads are significant for burial depths of less than 1 m. The maximum principal stresses of the buried pipeline decrease as the burial depth, vehicle velocity and surrounding soil’s elasticity modulus increase. For small burial depths, the stresses in buried pipes caused by vehicle motion in the direction normal to the pipe axis are more critical. However, the effects of motion direction are insignificant when the burial depth and the surrounding soil’s elasticity modulus increase. As the diameter of a buried pipe decreases, the maximum principal stresses increase.

Mechanical properties of CNT reinforced hybrid functionally graded materials for bioimplants

The hybrid functionally graded materials(FGM) of hydroxyapatite(HA), stainless steel 316L(SS316L) and carbon nanotubes(CNT) were synthesized for biomedical implants. Three different types of FGM were produced by the combination of SS316 L and CNT to reinforce HA in discrete layers of FGM. In the first type of FGM, concentration of SS316 L was varied from 10% to 40%(mass fraction) with an increment of 10% to reinforce micro HA. In the second type of FGM, 0.5%(mass fraction) functionalized CNT was added by maintaining the rest of composition as that of the first type of FGM. In the third type of FGM, mixture of micro and nano HA(mass ratio1:1) was used, keeping rest of composition similar to the second type of FGM. All types of FGM were subjected to uniaxial compaction and sintered by pressureless sintering technique at similar compaction and sintering parameters. The results show that the densification is enhanced with the addition of CNT and nanocrystalline HA in the FGM. Hardness and fracture toughness increase in both FGM reinforced with CNT, but the increase of the hardness and fracture toughness are more pronounced in FGM with micro and nanocrystalline HA.

Etiology of Upper GI Bleeding on Endoscopy

Background: Several circumstances and conditions lead to UGIB. The commonest among them peptic ulcer to an approximation of 50% in cases. The second major cause may be esoghageal varices and third is mallory-weis tears. There are also other causes, which may include erosins, tumors and other malformations like arteriovenous. Methods: This was an observational retrospective study of all patients referred to endoscopy. The study exclusion criteria include all patients who had repeat endoscopies within one month of the previous endoscopy whereas all the patients with UGIB were included in this study. Results: We retrieved data of total 150 patients who had experienced upper gastrointestinal endoscopy during the selected time period covered in the study, March 2015 to February 2016. The mean age of the patients was 43.3 ± 13.80. 80 (53.3%) of the patients were male and 70 (46.7%) were females. Exploring more into UGIB causes the majority of patients were with esophageal varices 68 (45.3%) with male to female share was 2:1. The second most cause was esophagitis (n = 22, (14.67%)), the distribution of the cause was similar in male to female. Conclusion: We may conclude from our findings that the most common cause of UGIB is esophageal varices in our population.

Environmental Assessment and Analysis of Chemical Properties of Drinking Water Using Geo-Spatial Technologies: Examples from Lahore Metropolitan

The contamination of drinking water in Pakistan is a serious threat to its citizens. Urbanization leads to an increased demand of drinking water supply that leads to excessive drawdown that further causes lowering of water table. The current WASA (Water and Sanitation Authority) supply network does not fulfill the demand of a growing metropolitan city of Lahore. The drawdown of Lahore is absolutely reliant on Water and Sanitation Authority (WASA) water supply network that causes numerous problems due to inappropriate management of groundwater capital and increased urbanization. This research investigates groundwater qualities: Alkalinity, Arsenic, Calcium, chloride, Total Hardness, Magnesium, Nitrate, pH and Total Dissolved solids using geographic information system (GIS). Residents of high concentrated towns, like Samnabad Town, Allama Iqbal Town, Gulberg, Data Ganj bakhsh Town, Ravi Town and Wahga town witness cut-down of water supply mainly due to over burden on tube wells. Chemical properties of drinking water show that most of the parameters were within the allowable limits of WHO whereas the pH, and arsenic values are higher than the average. Arsenic is a carcinogenic element which causes cancer and is higher in the drinking water. Finally, this study identifies highly contaminated groundwater zones and makes it convenient to find actual pollutants. Therefore, plans are needed to protect the aquifer.
