The regulation of spermatogonial proliferation and apoptosis is of great significance for maintaining *** single-cell RNA sequencing(scRNA-seq)analysis of the testis was performed to identify genes upregulated in *** scRNAseq analysis,we identified the spermatogonia upregulated gene origin recognition complex subunit 6(Orc6),which is involved in DNA replication and cell cycle regulation;its protein expression in the human and mouse testis was detected by western blot and *** explore the potential function of Orc6 in spermatogonia,the C18-4 cell line was transfected with control or Orc6 ***,5-ethynyl-2-deoxyuridine(EdU)and terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assays,flow cytometry,and western blot were used to evaluate its effects on proliferation and *** was revealed that ORC6 could promote proliferation and inhibit apoptosis of C18-4 *** RNA sequencing and bioinformatics analysis indicated that Orc6 was involved in the activation of wingless/integrated(Wnt)/β-catenin *** blot revealed that the expression ofβ-catenin protein and its phosphorylation(Ser675)were significantly decreased when silencing the expression of *** findings indicated that Orc6 was upregulated in spermatogonia,whereby it regulated proliferation and apoptosis by activating Wnt/β-catenin signaling.

脂质是橡胶树胶乳的主要非胶组分,与胶乳代谢和天然橡胶质量形成密切相关,但其代谢调控相关研究欠缺。WRINKLED(WRI)是植物AP2转录因子家族成员,在促进碳源从糖流向油脂合成中发挥了重要作用。本研究从橡胶树胶乳中克隆到3个HbWRI基因家族成员,分别为HbWRI1,HbWRI2和HbWRI3,并对其进行生物信息学分析、不同组织部位表达分析以及亚细胞定位分析。结果表明,HbWRI1,HbWRI2和HbWRI3的编码序列全长分别为1128、1122 bp和1050 bp,分别编码了375、373和349个氨基酸。HbWRI1-3蛋白具有2个60~70个氨基酸大小的AP2保守结构域,第1个AP2结构域中包含1个VYL基序和1个14-3-3蛋白结合基序。生信分析预测HbWRI1-3均无跨膜结构域,无信号肽,为疏水性蛋白,且含有多个磷酸化位点。亚细胞定位分析表明,HbWRI1-3蛋白定位于细胞核上。不同组织部位表达分析表明,HbWRI1、HbWRI2和HbWRI3基因均在橡胶树胶乳中表达量最高,且在胶乳中HbWRI3是家族成员中表达量最高的基因,暗示HbWRI1-3可能参与了胶乳脂质合成调控。该研究为进一步探索HbWRI1-3转录因子在橡胶树胶乳代谢及影响天然橡胶质量形成中的作用奠定基础。

脊髓损伤(spinal cord injury,SCI)是一种严重的中枢神经系统疾病,致残率和死亡率较高,具有复杂的病理生理机制。微小RNA(microRNA,miRNA)作为一种非编码RNA,在SCI中发挥重要作用。miRNA参与SCI后的炎症反应、氧化应激、轴突再生、细胞凋亡等的调控,并与长链非编码RNA(long non-coding RNA,lncRNA)、环状RNA(circular RNA,circRNA)相互作用调节SCI的病理生理过程。本文归纳了SCI后miRNA的表达变化,综述了miRNA在SCI中的靶向机制及miRNA靶向药物的研究现状,为SCI的基础和临床研究提供新靶点和新视野。

BACKGROUND Nonalcoholic fatty liver disease(NAFLD) is currently the outstanding cause of chronic liver disease in children and adolescents, especially in overweight and obese groups. Liver biopsy is the reference standard to diagnose NAFLD but invasive, thus it is not the best choice in clinical diagnosis and *** resonance(MR) is widely used in clinical trials to noninvasively quantify liver fat content in adults and children in foreign countries. While currently, it is rarely used in Chinese children and adolescents. We postulated that quantifying hepatic steatosis by MR could be extended to children and adolescents in *** To investigate the accuracy of MR imaging(MRI) in quantifying liver fat with MR spectroscopy(MRS) as a reference. A secondary goal was to assess the prevalence of NAFLD in overweight and obese Chinese children and *** were 86 children and adolescents enrolled in this study, including 65 overweight and obese children and 21 healthy children. The participants underwent MRI and MRS. MRI and MRS were performed using multi-echo Dixon and HISTO sequences, respectively, to calculate hepatic proton density fat fraction(PDFF). Hepatic steatosis was diagnosed using MRS-PDFF > 5% as the threshold. Spearman's analysis was used to evaluate the correlation between MRI and MRS. The agreement between these two methods was assessed by BlandAltman *** The MRI-PDFF in the MRS region of interest and the entire liver was 9.9% ±10.3% with a range of 0.3%-39.9%, and 10.6% ± 9.4% with a range of 1.9%-38.9%,respectively. 为探究生物炭在土壤氮素分布以及对根系氮素吸收利用与产量形成中所产生的影响,开展两年(2019-2020)定点试验。试验以先玉335为供试材料,采用裂区试验设计,以氮肥为主区,300 kg/hm^(2)(N300)、150 kg/hm^(2)(N150)、不施氮肥(N0);以生物炭为副区,不施生物炭(C0)、8 t/hm^(2)(C8)、16 t/hm^(2)(C16)、24 t/hm^(2)(C24)。结果表明,在0~15 cm土层内,C8N0、C16N0、C24N0处理组合的有机碳在2019年和2020年较C0N0处理组合分别增加16.27%~31.52%和7.27%~35.57%。在2019年和2020年,C8N150、C16N150、C24N150处理组合较C0N150处理组合的根长分别提升11.22%~51.18%和6.32%~46.17%,产量分别提升10.70%~23.55%和4.77%~22.68%。综上,生物炭施炭量在24 t/hm^(2)时,施氮肥150 kg/hm^(2)对土壤有机质积累分布和根系的发育状况达到最佳状态。

AIM To prepare hybridoma cell lines which secrete anti HCV recombinant NS3 and NS5 proteins′ monoclonal antibodies, and to evaluate their usage in the study of the distribution of HCV NS3 and NS5 antigen in liver tissues. METHODS The hybridoma cell lines were raised using the spleen cells of BALB/C mouse immunized with recombinant NS3 and NS5 proteins according to the conventional protocols. The antibody secreting cells were screened using solid phase ELISA and cloned by limited dilution method. In order to determine the specificity of these hybridoma cell lines, the culture supernatant of these cells was western blot assayed with expression and nonexpresion E. coli and ELISA with other antigens, including HCV core and NS3 and HBsAg. Immunohistochemistry of 51 cases paraffin embedded liver tissues was performed to determine the distribution of HCV NS5 antigen as well as NS3 antigen in liver tissues. RESULTS Eight hybridoma cell lines secreting monoclonal antibodies against HCV NS3 and NS5 proteins were raised. They are named 2B6, 2F3, 3D8, 3D9, 8B2, 6F11, 4C6 and 7D9. Among them only 2B6 against NS3 protein can react with the polypipetides of C7 that is another recombinant polypipetides of NS3 gene. Others have no reaction with HCV core and HBsAg of HBV, and there is no cross reaction between NS3Ag and anti NS5Ag McAb and between NS5Ag and anti NS3 McAb. The immunohistochemistry results indicate that no HCV antigen was detected in the specimens of HBV infection in 20 cases. In 31 HCV infected specimens the positive rate of NS3Ag and NS5Ag are 51 6% (16/31) and 54 9% (17/31), respectively. There were six pure HCV infected specimens in these 31 specimens and half of them were HCV NS3Ag and NS5Ag positive. In the co infection of HBV and HCV group the positive rate of NS3Ag and NS5Ag were 52% (13/25) and 56% (14/25), respectively, almost the same with that of pure HCV infected group. A new and innovative detector system based on a silicon strip detector dedicated to the study of the reaction induced by lighter radioactive beams is described *** detector system consists of five sets of three types of telescopes,which are successfully used to measure the angular distributions of both elastic scattering and breakup simultaneously, on the Radioactive Ion Beam Line in Lanzhou at Heavy Ion Research Facility in Lanzhou. This silicon detector array is used to measure the elastic scattering angular distributions of ^(11) Be on a ^(208) Pb target at E_(lab) = 140 and 209 MeV. A comparison of the Monte Carlo simulations with the experimental results shows a reasonable consistency.
