Gumiganghwal-tang is a traditional herbal medicine widely used for its anti-inflammatory,analgesic,and antipyretic ***,the safety and efficacy of its active ingredients based on an in vivo pharmacokinetic(PK)study have yet been *** have established a sensitive and accurate UPLC-ESI-MS/MS method and conducted a PK study on 14 constituents of Gumiganghwal-tang through human plasma *** conditions were optimized according to the physicochemical properties of the 14 compounds to facilitate efficient separation and eliminate overlap or interference between ***-C18 and Inertsil-C8 columns were used as UPLC stationary phases,and acetonitrile and aqueous formic acid were used as mobile *** the analytes were quantified with a triple quadrupole mass spectrometer using electrospray ionization in multiple reaction monitoring *** chromatograms of 14 bioactive compounds showed excellent elution and sensitivity,and each peak was selectively separated and quantified without interference with each other or *** established analytical method was based on international guidelines and was successfully used to perform PK studies of 14 herbal ingredients in humans after oral administration with Gumiganghwal-tang *** oral absorption of most active components of Gumiganghwal-tang was relatively rapid and remained considerably long in the body to be quantified in plasma up to 48 h after administration.

The extract of Celtis choseniana Nakai alleviates testosterone-in-duced benign prostatic hyperplasia through inhibiting 5αreductase type 2 and the Akt/NF-κB/AR pathway

Benign prostatic hyperplasia(BPH)is a chronic male disease characterized by the enlarged *** choseniana Nakai(***)is medicinally used to alleviate pain,gastric disease,and lung *** this study,the effect of *** extract on BPH was investigated using testosterone-induced *** Dawley rats were divided into five groups:control,BPH(testosterone 5 mg·kg^(−1)),Fina(finasteride 2 mg·kg^(−1)),and ***(50 and 100 mg·kg^(−1)).After four weeks of TP treatment with finasteride or ***,prostate weights and DHT levels were *** addition,the prostates were histopathologically examined and measured for protein kinase B(Akt)/nuclear factor-κB(NF-κB)/AR signaling,proliferation,apoptosis,and ***-state weight and epithelial thickness were reduced in the *** groups compared with that in the BPH *** extract of *** acted as a 5αreductase inhibitor,reducing DHT levels in the ***,the extract of *** blocked the activation of p-Akt,nuclear NF-κB activation and reduced the expression of AR and PSA compared with ***,the ex-pression of Bax,PARP-1,and p53 increased,while the expression of bcl-2 *** present study demonstrated that ***-ana extract alleviated testosterone-induced BPH by suppressing 5αreductase and Akt/NF-κB activation,reducing AR signaling and in-ducing apoptosis and autophagy in the *** results suggested that *** probably contain potential herbal agents to alleviate BPH.

This review highlights the recent advancements in Mg research in South Korea with a prime focus on high-speed-extrudable Mg–Bi-based alloys for high productivity and strength,innovative techniques utilizing{10–12}twinning for improved mechanical properties,and alloying and processing methods for enhanced corrosion ***-alloyed Mg–Bi-based alloys possess thermally stableα-Mg matrix and secondary phase,which ensures high-speed extrusion of these alloys at elevated temperatures without hot ***,they exhibit outstanding extrudability with a maximum extrusion speed of up to 70 m/***,their high alloying contents offer excellent strength even after high-speed extrusion through strong solid solution hardening and particle hardening effects,making them suitable for high-performance extruded Mg *** pre-twinning process utilizing{10–12}twinning and the combined process of pre-twinning and subsequent annealing have shown promise in controlling microstructure and texture of wrought Mg alloys and thus enhancing their mechanical *** pre-twinning process enhances tensile strength,fatigue properties,and age-hardening rate of Mg ***,the combined processes of pre-twinning and subsequent annealing considerably improve their ductility,stretch formability,bending formability,and damping *** have been made to improve the corrosion resistance of Mg alloys through alloying additions,process treatments,and surface *** elements like Ca,Sc,and Sm alter the microstructural features(such as secondary phases and grain size)that affect the corrosion *** treatments such as multidirectional forging,screw rolling,and pulse electron beam can also improve the corrosion resistance by refining the ***,advanced surface coating technologies can create durable and corrosion-resistant layers for effectively protecting the Mg *** these research activities conducted

Functional Insights of Plant GSK3-like Kinases： Multi-Taskers in Diverse Cellular Signal Transduction Pathways

The physiological importance of GSK3-like kinases in plants emerged when the functional role of plant GSK3-like kinases represented by BIN2 was first elucidated in the brassinosteroid （BR）-regulated signal transduction pathway. While early studies focused more on understanding how GSK3-like kinases regulate BR signaling, recent studies have implicated many novel substrates of GSK3-like kinases that are involved in a variety of cellular processes as well as BR signaling. Plant GSK3-like kinases play diverse roles in physiological and developmental processes such as cell growth, root and stomatal cell development, flower development, xylem differentiation, light response, and stress responses. Here, we review the progress made in recent years in understanding the versatile functions of plant GSK3-like kinases. Based on the relationship between GSK3-like kinases and their newly identified substrates, we discuss the physiological and biochemical relevance of various cellular signaling mediated by GSK3-like kinases in plants.

Ethanol extract of Kalopanax septemlobus leaf inhibits HepG2 human hepatocellular carcinoma cell proliferation via inducing cell cycle arrest at G_1 phase

Objective:To investigate the effects of an ethanol extract of Kalopanax septemlobus(Thunb.)***(EEKS) on cell proliferation in human hepatocellular carcinoma cells and its mechanisms of ***:Cells were treated with EEKS and subsequently analyzed for cell proliferation and flow cytometry *** of cell cycle regulators were determined by reverse transcriptase polymerase chain reaction analysis and Western blotting,and activation of eyclin-associaled kinases studied using kinase ***:The EEKS suppressed cell proliferation in both HepG2 and Hep3 B cells,but showed a more sensitive anli-proliferative activity in HepG2 *** cytometry analysis revealed an association between the growth inhibitory effect of EEKS and with G_1 phase cell cycle arrest in HepG2 cells,along with the dephosphorylation of retinoblastoma protein(pRB) and enhanced binding of pRB with the E2 F transcription factor family *** with EEKS also increased the expression of cyclin-dependent kinase(CDK) inhibitors,such as p21WAF1/CIP1 and *** any noticeable changes in G_1 cyclins and CDKs(except for a slight decrease in CDK4).Treatment of HepG2 cells with EEKS also increased the binding of p21 and p27 with CDK4 and *** was paralleled by a marked decrease in the cyclin D- and cyclin E-associated kinase ***:Overall,our findings suggest that EEKS may be an effective treatment for liver cancer through suppression of cancer cell proliferation via G_1,cell cycle arrest Further studies arc required to identify the active compounds in EEKS.

Effects of ethanol on the tonicity of corporal tissue and the intracellular Ca^2＋ concentration of human corporal smooth muscle cells

Heavy alcohol consumption is associated with an increased risk of erectile dysfunction （ED）; however, the acute effects of ethanol （EtOH） on penile tissue are not fully understood. We sought to investigate the effects of EtOH on corporal tissue tonicity, as well as the intracellular Ca^2＋ concentration （[Ca^2＋]i） and potassium channel activity of corporal smooth muscle. Strips of corpus cavernosum （CC） from rabbits were mounted in organ baths for isometric tension studies. Electrical field stimulation （EFS） was applied to strips precontracted with 10 μmol L^-1 phenylephrine as a control. EtOH was then added to the organ bath and incubated before EFS. The [Ca^2＋]i levels were monitored by the ratio of fura-2 fluorescence intensities using the fura-2 loading method. Single-channel and whole-cell currents were recorded by the conventional patch-clamp technique in short-term cultured smooth muscle cells from human CC tissue. The corpus cavernosal relaxant response of EFS was decreased in proportion to the concentration of EtOH. EtOH induced a sustained increase in [Ca^2＋]i in a dose-dependent manner, Extracellular application of EtOH significantly increased whole-cell K^＋ currents in a concentration-dependent manner （P 〈 0.05）. EtOH also increased the open probability in cell-attached patches; however, in inside-out patches, the application of EtOH to the intracellular aspect of the patches induced slight inhibition of Ca^2＋-activated potassium channel （KCa） activity. EtOH caused a dose-dependent increase in cavemosal tension by alterations to [Ca^2＋]i. Although EtOH did not affect KCa channels directly, it increased the channel activity by increasing [Ca^2＋]i. The increased corpus cavemosal tone caused by EtOH might be one of the mechanisms of ED after heavy drinking.

Optimization of rotor shape for constant torque improvement and radial magnetic force minimization

The design of notch and barrier was optimized in order to improve the characteristics of constant torque while minimizing the cogging torque that occurs as a result of teeth and slot structure. The barrier was installed in order to minimize the cogging torque and torque ripple by finite element method (FEM) with a reduced barrier width toward the center of magnetic pole. The position and width of notch, which can offset cogging torque, can be calculated with energy distribution of air-gap using Fourier series. The optimized model demonstrates a 60% decrease in the cogging torque, a 75.3% decrease in the torque ripple and a 3% increase in the operating torque when compared with the basic model.

Multiplexed DNA-functionalized graphene sensor with artificial intelligence-based discrimination performance for analyzing chemical vapor compositions

This study presents a new technology that can detect and discriminate individual chemical vapors to determine the chemical vapor composition of mixed chemical composition in situ based on a multiplexed DNA-functionalized graphene(MDFG)nanoelectrode without the need to condense the original vapor or target *** the best of our knowledge,our artificial intelligence(Al)-operated arrayed electrodes were capable of identifying the compositions of mixed chemical gases with a mixed ratio in the early *** innovative technology comprised an optimized combination of nanodeposited arrayed electrodes and artificial intelligence techniques with advanced sensing capabilities that could operate within biological limits,resulting in the verification of mixed vapor chemical *** selective sensors that are tolerant to high humidity levels provide a target for"breath chemovapor fingerprinting"for the early diagnosis of *** feature selection analysis achieved recognition rates of 99%and above under low-humidity conditions and 98%and above under humid conditions for mixed chemical *** 1D convolutional neural network analysis performed better,discriminating the compositional state of chemical vapor under low-and high-humidity conditions almost *** study provides a basis for the use of a multiplexed DNA-functionalized graphene gas sensor array and artificial intelligence-based discrimination of chemical vapor compositions in breath analysis applications.

In situ observation of atomic movement in a ferroelectric film under an external electric field and stress

Atomic movement under application of external stimuli （i.e., electric field or mechanical stress） in oxide materials has not been observed due to a lack of experimental methods but has been well known to determine the electric polarization. Here, we investigated atomic movement arising from the ferroelectric response of BiFeO3 thin films under the effect of an electric field and stress in real time using a combination of switching spectroscop）6 time-resolved X-ray microdiffraction, and in situ stress engineering. Under an electric field applied to a BiFeO3 film, the hysteresis loop of the reflected X-ray intensity was found to result from the opposing directions of displaced atoms between the up and down polarization states. An additional shift of atoms arising from the linearly increased dielectric component of the polarization in BiFeO3 was confirmed through gradual reduction of the diffracted X-ray intensity. The electric-field- induced displacement of oxygen atoms was found to be larger than that of Fe atom for both ferroelectric switching and increase of the polarization. The effect of external stress on the BiFeO3 thin film, which was controlled by applying an electric field to the highly piezoelectric substrate, showed smaller atomic shifts than for the case of applying an electric field to the film, despite the similar tetragonality.
