Prevalence and risk factors associated with Japanese encephalitis virus infection in swine population of Assam,India

Objective:To assess the prevalence of Japanese encephalitis virus(JEV)and associated risk factors in the swine population of ***:A total of 432 swine serum and blood samples were collected from barpeta and Sonitpur districts of Assam and were screened for the presence of JEV *** related to risk factors was collected using a self-designed questionnaire from 120 swine-rearing ***-mixed models were used for prevalence *** and multivariate regression models were constructed to evaluate the association of demography,season and management practices with JEV positive ***:Overall,the JEV infection prevalence was 51.6%at farm and 47.1%at slaughter *** analysis of partial sequence of envelope gene of two positive field samples revealed that both isolates belonged to genotypeⅢ*** 1 shared a common clade with human isolates while isolate 2 belonged to the same clade as that of other JEV swine strain isolated from *** final multivariate model showed that two factors including monsoon season(Adjusted Or 5.6;95%CI 2.1-14.9;Pr logging in the area near the pig shelter(Adjusted Or 16.9;95%CI 6.1-47.3;Pre associated with greater odds of swine being infected with ***:High prevalence of JEV in swine population of Assam state indicates a significant risk of virus transmission to humans while risk factor study underlines the urgent need for awareness campaigns in the Assam.

The focus of this study is to investigate the arsenic movement and impacts on the residual concentrations on groundwater pollution *** Gangetic plain area in the ballia,Uttar Pradesh is selected as study area,which is also reported to extreme arsenic pollution in soil-water system.A modelling approach is developed to assess the arsenic flux in partially saturated zone using data of soil texture,soil hydraulic properties and *** type,slope,and land-use cover is considered for estimating the transient flux at the top boundary from daily precipitation and evapotranspiration data of the study *** transport in the subsurface is predicted by the mass transfer equation,which is derived by integrating Darcy's law with the equation of mass *** arsenic profiles of varying hydrogeological conditions associated with different locations in the study area are presented as breakthrough *** results shows that the arsenic transport is dominated by the advective flux and strongly depends on the soilmoisture flow *** may increases the arsenic load to underlaying groundwater *** simulated results suggest that mobility plays a vital role arsenic transport as well as on adsorbed arsenic concentration in ***,the adsorption isotherms show that the high peak curve for bairai and low at Sikarderpur.A higher pollution risk is observed in the belthara road,whereas a lower vulnerability is computed in the north and northeast *** study can help in strategising sustainable groundwater management and protection planning of identified regions of India.

Many plants contain ribosome inactivating proteins (rIPs) with N-glycosidase activity, which depurinate large ribosomal rNA and arrest protein synthesis. rIPs so far tested inhibit replication of mrNA as well as DNA viruses and these proteins, isolated from plants, are found to be effective against a broad range of viruses such as human immunodeficiency virus (HIV), hepatitis b virus (HbV) and herpes simplex virus (HSV). Most of the research work related to rIPs has been focused on antiviral activity against HIV; however, the exact mechanism of antiviral activity is still not clear. The mechanism of antiviral activity was thought to follow inactivation of the host cell ribosome, leading to inhibition of viral protein translation and host cell death. Enzymatic activity of rIPs is not limited to depurination of the large rrNA, in addition they can depurinate viral DNA as well as rNA. recently, Phase I/II clinical trials have demonstrated the potential use of rIPs for treating patients with HIV disease. The aim of this review is to focus on various rIPs from plants associated with anti-HIV activity.

High-dose infliximab for treatment of pediatric ulcerative colitis:A survey of clinical practice

AIM:To assess attitudes and trends regarding the use of high-dose infliximab among pediatric gastroenterologists for treatment of pediatric ulcerative colitis(UC).METHODS:A 19-item survey was distributed to subscribers of the pediatric gastroenterology(PEDSGI) *** were submitted anonymously and results compiled in a secure ***:A total of 113 subscribers(88% based in the United States) responded(101 pediatric gastroenterology attendings and 12 pediatric gastroenterology fellows).There were 46% in academic medical institutions and 39% in hospital-based *** majority(91%) were treating >10 patients with UC;13% were treating >100 patients with UC;91% had prescribed infliximab(IFX) 5 mg/kg for UC;72% had prescribed IFX 10 mg/kg for *** a 5-point Likert scale,factors that influenced the decision not to increase IFX dosing in patients with UC included:"improvement on initial dose of IFX"(mean:3.88) and "decision to move to colectomy"(3.69).Lowest mean Likert scores were:"lack of guidelines or literature regarding increased IFX dosing"(1.96) and "insurance authorization or other insurance issues"(2.34)."Insurance authorization or other insurance issues" was identified by 39% as at least somewhat of a factor(Likert score ≥ 3) in their decision not to increase the IFX *** 10 mg/kg was more commonly used for the treatment of pediatric UC among responders based in the United States(75/100) compared to non-United States responders(6/13,P = 0.047).Induction of remission was reported by 78% of all responders and 81% reported maintenance of remission with IFX 10 mg/*** responder reported one death with IFX 10 mg/***:IFX 10 mg/kg is more commonly used in the United States to treat pediatric *** and safety data are required to avoid insurance barriers for its use.

b-cell Ligand Processing Pathways Detected by Large-scale Comparative Analysis

The initiation of b-cell ligand recognition is a critical step for the generation of an immune response against foreign bodies. We sought to identify the biochemical pathways involved in the b-cell ligand recognition cascade and sets of ligands that trigger similar immunological responses. We utilized several comparative approaches to analyze the gene coexpression networks generated from a set of microarray experiments spanning 33 different ligands. First, we compared the degree distributions of the generated networks. Second, we utilized a pairwise network alignment algorithm, biNA, to align the networks based on the hubs in the networks. Third, we aligned the networks based on a set of KEGG pathways. We summarized our results by constructing a consensus hierarchy of pathways that are involved in b cell ligand recognition. The resulting pathways were further validated through literature for their common physiological responses. Col- lectively, the results based on our comparative analyses of degree distributions, alignment of hubs, and alignment based on KEGG path- ways provide a basis for molecular characterization of the immune response states of b-cells and demonstrate the power of comparative approaches （e.g., gene coexpression network alignment algorithms） in elucidating biochemical pathways involved in complex signaling events in ceils.

Multiplex qPCr for serodetection and serotyping of hepatitis viruses: A brief review

The present review describes the current status of multiplex quantitative real time polymerase chain reaction(q PCr) assays developed and used globally for detection and subtyping of hepatitis viruses in body fluids. Several studies have reported the use of multiplex q PCr for the detection of hepatitis viruses, including hepatitis A virus(HAV), hepatitis b virus(HbV), hepatitis C virus(HCV), hepatitis D virus(HDV), and hepatitis E virus(HEV). In addition, multiplex q PCr has also been developed for genotyping HbV, HCV, and HEV subtypes. Although a single step multiplex q PCr assay for all six hepatitis viruses, i.e., A to G viruses, is not yet reported, it may be available in the near future as the technologies continue to advance. All studies use a conserved region of the viral genome as the basis of amplification and hydrolysis probes as the preferred chemistries for improved detection. based on a standard plot prepared using varying concentrations of template and the observed threshold cycle value, it is possible to determine the linear dynamic range and to calculate an exact copy number of virus in the specimen. Advantages of multiplex q PCr assay over singleplex or other molecular techniques in samples from patients with co-infection include fast results, low cost, and a single step investigation process.

Discovery and preclinical development of ⅢM-160, a bergenia ciliatabased anti-inflammatory and anti-arthritic botanical drug candidate

Objective: bergenia ciliata(Haw.) Sternb. is used in the Indian traditional system of medicine to treat various ailments including rheumatism and to heal wounds. The objective of the present study was to perform a preclinical characterization of the b. ciliata-based botanical extract ***: ⅢM-160 was chemically standardized and analyzed for heavy metal content, aflatoxins,pesticides and microbial load. The in vitro and in vivo efficacies were determined in suitable models of inflammation, arthritis and nociception. An acute oral toxicity study was performed in Swiss albino mice.A suitable oral formulation was developed and ***: bergenin was found to be the major component(9.1% w/w) of ⅢM-160. The botanical lead displayed inhibition of lipopolysaccharide-induced production of proinflammatory cytokines in THP-1 cells, with selectivity toward interleukin-6(IL-6) and had an excellent safety-window. It showed anti-inflammatory, anti-arthritic and antinociceptive activity in animal models and was not toxic at oral doses up to 2 g/kg in Swiss-albino mice. The gastroretentive, sustained-release capsule formulation showed sustained-release of the bergenin over the period of 24 h, resulting in improved plasma-exposure of bergenin in Sprague–Dawley ***: The dual-activity of IL-6 inhibition and antinociception marks the suitability of ⅢM-160 for treating rheumatoid arthritis. This study will serve as the benchmark for further research on this botanical formulation.

Potential aspect of rice husk biomass in Australia for nanocrystalline cellulose production

Nanocrystalline cellulose(NCC) was produced from rice husk biomass(Oryza sativa) by a chemical extraction process to explore the potential aspect of agro-waste biomass in Australia. In this work, the delignified rice husk pulp(D-rHP) was produced by alkaline delignification of raw rice husk biomass(r-rHb) using 4 mol·L^(-1) alkali solutions(Na OH) in a jacketed glass reactor under specific experimental conditions. D-rHP was bleached using 15% sodium hypochlorite, and the bleached rice husk pulp was coded as b-rHP. Finally,raw suspension of NCC was produced by the acid hydrolysis of b-rHP using 4 mol·L^(-1) sulphuric acid. The raw suspension of NCC was neutralized by a buffer solution and analyzed by TAPPI, FT-Ir, XrD, SEM, AFM, and TEM. FT-Ir spectra of NCC are different to r-rHb but similar with b-rHP and D-rHP. From XrD results, the crystallinity of NCC was found to be approximately 65%. In AFM analysis particle thicknesses have been confirmed to be in the range of(25 ± 15.14) nm or(27 ± 15.14) nm which is almost the same. From TEM analysis particle dimensions have been confirmed to be in the range of(50 ± 29.38) nm width and(550 ± 302.75) nm length with the aspect ratio ~ 11:1(length/diameter) at a 500 nm scale bar. On the other hand, at a 200 nm scale bar the particle dimensions have been confirmed to be in the range of(35 ± 17) nm width and(275 ± 151.38)nm length with the aspect ratio ~ 8:1. The aspect ratio of individual crystalline domain was determined in TEM analysis which is 10:1(100/10). Therefore the aspect ratios and dimensions of nanoparticles in NCC suspension are almost the same and in nano-meter scale, as confirmed from both AFM and TEM results. The yield of NCC from b-rHP was found to be approximately 95%, and the recovery of cellulose from r-rHb is about 90%.

A cross-sectional survey of 489 male subjects in the age group 15 to 65 years engageddirectly or indirectly in mango cultivation along with 208 control subjects was carried out to find their socio-economic, environmental and health conditions. The conditions like high illiteracy rate (49.5%), poverty (PCI less than rs 100 per month, 52.2%), poor housing (mud houses, 66.7%) unsafe water supply (78.6%) were prevailing in the surveyed population. The high respiratory morbidity may be attributed to high prevalence of smoking andprolonged inhalation of organic dusts during farming operation associated with illiteracyand poor socio-economic status. Gastrointestinal disorders were related to poor hygienic conditions, smoking and consumption of contaminated water. The symptoms pertaining to CNS, skin and eyes were found to be associated with exposure to pesticides
