Neuroimmunology is emerging as a pivotal field,shedding light on the intricate dialogues between the central nervous system(CNS)and the immune *** exploration is particularly significant in understanding microglia,the CNS’s innate immune cells,beyond the conventional conflation of“neuroinflammation”and“microglial activation.”This conflation has clouded the true complexity of these processes,potentially stalling scientific progress and the development of new *** challenge the long-standing perspectives that have oversimplified these interactions,advocating for a deeper exploration of the dynamic relationship between neuroinflammation and microglial *** dissecting specific molecular pathways,we aim to illuminate their elaborate roles in neuroinflammatory responses,especially in the context of Alzheimer’s disease(AD).Here,neuroinflammation is not merely a passive observer or a direct antagonist but a complex agent in the disease’s *** article calls for a significant paradigm shift towards an integrative,multi-omics approach to *** such a comprehensive framework is crucial for advancing our understanding of neuroinflammatory conditions and paving the way for targeted therapeutic strategies for brain diseases.

We develop first order optimality conditions for constrained vector optimization. The partial orders for the objective and the constraints are induced by closed and convex cones with nonempty interior. After presenting some well known existence results for these problems, based on a scalarization approach, we establish necessity of the optimality conditions under a Slater-like constraint qualification, and then sufficiency for the K-convex case. We present two alternative sets of optimality conditions, with the same properties in connection with necessity and sufficiency, but which are different with respect to the dimension of the spaces to which the dual multipliers belong. We introduce a duality scheme, with a point-to-set dual objective, for which strong duality holds. Some examples and open problems for future research are also presented.

The development of antibiotics is regarded as one of the most significant scientific advances in modern medicine. before the commercialization of antibiotics, infectious diseases were catastrophic throughout history [1]. Penicillin was used to treat serious infections in the 1940s, but unfortunately, with the introduction of antibiotics came the emergence of antibiotic resistance [2,3]. Evolved and adapted methicillin-resistant Staphylococcus aureus (MRSA) strains are presently impacting community and healthcare systems across the globe [4].

Virulence factors of Enterococcus strains isolated from patients with inflammatory bowel disease

AIM: To determine the features of Enterococcus that contribute to the development and maintenance of the inflammatory process in patients with inflammatory bowel disease (IbD). METHODS: Multiplex polymerase chain reaction (PCR) was applied to assess the presence of genes that encode virulence factors [surface aggregating protein (asa1), gelatinase (gelE), cytolysin (cylA), extracellular surface protein (esp) and hyaluronidase (hyl)] in the genomic DNA of 28 strains of Enterococcus isolated from the intestinal tissues of children with IbD (n =16) and of children without IbD (controls; n = 12). Additionally, strains with confirmed presence of the gelE gene were tested by PCR for the presence of quorum sensing genes (fsrA, fsrb, fsrC) that control the gelatinase production. Gelatinase activity was tested on agar plates containing 1.6% gelatin. We also analysed the ability of Enterococcus strains to release and decompose hydrogen peroxide (using Analytical Merckoquant peroxide test strips) and tested their ability to adhere to Caco-2 human gut epithelium cells and form biofilms in vitro. RESULTS: A comparison of the genomes of Enterococcus strains isolated from the inflamed mucosa of patients with IbD with those of the control group showed statistically significant differences in the frequency of theasa1 gene and thegelE gene. Furthermore, the cumulative occurrence of different virulence genes in the genome of a single strain ofEnterococcus isolated from the IbD patient group is greater than in a strain from the control group, although no significant difference was found. Statistically significant differences in the decomposition of hydrogen peroxide and adherence to the Caco-2 epithelial cell line between the strains from the patient group and control group were demonstrated. AIM： To assess the effectiveness and side-effects of lamivudine therapy for children with chronic hepatitis b （CHb） who fail to respond to or have contraindications to interferon-α （IFN-α） therapy. METHODS： Fifty-nine children with CHb were treated with 100 mg lamivudine tablets given orally once daily for 12 too. Alanine aminotransferase （ALT） activity was evaluated monthly during the therapy and every 3 months after its discontinuation. Hbe antigen, antiHbe antibodies, HbV DNA level in serum were evaluated at baseline and every six months during and after the lamivudine therapy. Sustained viral response （SVR） to lamivudine therapy was defined as permanent （not shorter than 6 mo after the end of the therapy）, namely ALl＂ activity normalization, seroconversion of HbeAg to anti-Hbe antibodies, and undetectable viral HbV-DNA in serum （lower than 200 copies per mL）. The analysis of the side-effects of the lamivudine treatment was based upon interviews with the patients and their parents using a questionnaire concerning subjective and objective symptoms, clinical examinations, and laboratory tests performed during clinical visits monthly during the therapy, and every 3 mo after the therapy. RESULTS： ALT normalisation occurred in 47 （79.7%） patients between the first and 11^th mo of treatment （mean 4.4±2.95 mo, median 4.0 mo）, and in 18 （30.5%） of them after 2 mo of the therapy. There was no correlation between the time of ALT normalization and the children＇s age, the age of HbV infection, the duration of HbV infection, inflammation activity score （grading）, staging, ALT activity before treatment, serum HbV DNA level, and lamivudnie dose per kg of body weight. HbeAg/anti Hbe seroconversion was achieved in 27.1% of *** higher rate of seroconversion was connected with lower serum HbV DNA level and longer duration of HbV infection.
