Wave overtopping at a sea dike was ob.erved using video images during a storm in July 2018 at Rizhao Coast,China.A shore-b.sed video monitoring system was mounted to collect coastal images with a sample frequency of 1 Hz in the b.ginning 10 min of each hour during daylight.A method to extract the frequency,location,width and duration of individual overtopping events b.sed on the shore-b.sed video monitoring system was developed.A total of 6252 individual overtopping events were detected over a 360m long sea dike during the storm of 10 h in a safe and lab.r-saving way,enab.ing a detailed analysis of the temporal and spatial variation of wave *** temporal variation of overtopping duration,frequency and volume is b.sically in sync with the change of tidal level(R=0.87,0.82 and 0.76,respectively).The increase of wave height increases the overtopping frequency *** found a high correlation b.tween the hourly ob.erved data and the predicted results of two commonly used *** the previous field measurements of overtopping that were limited in a single location,significant spatial variation of overtopping in the alongshore direction is *** overtopping volume varies with a factor of 6 within the range of several wave *** is further revealed that the spatial variation of overtopping is highly correlated with the alongshore variation of surf zone width with a correlation coefficient of *** present study suggests the feasib.lity of shore-b.sed video monitoring technique to capture the main features of wave overtopping at coastal dikes,providing new possib.lities to monitor wave overtopping in the field and to improve prediction tools.

We consider the special case that the dark matter(DM) candidate is not detected in direct-detection programs when the experimental sensitivity reaches the neutrino flux b.ckground. In such circumstance the DM searches at the colliders impose constraints on the DM relic ab.ndance if the DM candidate is a WIMPs type. Specifically, we consider the triplet(quintet and septet) DMs in the framework of minimal DM model and explore the potential of discovering the DM candidate in the mono-jet, mono-photon and vector b.son fusion channels at the Large Hadron Collider(LHC) and future 100 TeV hadron collider. If the DM candidate in such a scenario is discovered at the LHC, then additional DM candidates are needed to explain the ob.erved relic *** the other hand, null results in those DM searching programs at the colliders give rise to lower limits of DM relic ab.ndance.

The molecular structures of ground state and first single excited state for pyrazoline derivatives are optimized with DFT b.LYP method and ab.initio “configuration interaction with single excitations”（CIS） method, respectively. The frontier molecular orb.tal characteristics have b.en analyzed systematically, and the electronic transition mechanism has b.en discussed. Electronic spectra are calculated b. using TD-DFT method. These results are consistent with those from the experiment.

The flotation tests, zeta potential measurements, and Fourier transform infrared spectroscopy （FtiR） analysis on galena, sphalerite, and pyrite were studied in a collecting-depressing-reactivating （CDR） system. In this system, sulphide minerals were first collected and acti- vated b. the collector, and then depressed strongly b. Ca（OH）2 in a strong alkaline solution. Finally, they were reactivated b. H2SO4. The flotation tests of pure minerals showed that in the Ca（OH）2 depressing process sulphide minerals had similar flotation characteristics b.cause they had already b.en influenced b. the collector. Hence, the flotab.lity differences b.tween them were reduced. However, in the H2SO4 re- activating process considerab.e differences in the flotab.lity b.tween galena and sphalerite/pyrite were produced. That is to say, galena was relatively easy to b. reactivated b. H2SO4, b.t sphalerite and pyrite were not reactivated at pH 〉 11. The zeta potentials of sulfide minerals measured b. the Zeta Plus presented irreversib.e characteristics on the change of pH values. The results of the FtiR spectra analysis indi- cated that the collectors already adsorb.d on the mineral surface were removed partially b. Ca（OH）〉

Genetic engineering of T cells with chimeric antigen receptors for hematological malignancy immunotherapy

The host immune system plays an instrumental role in the surveillance and elimination of tumors b. recognizing and destroying cancer cells. In recent decades, studies have mainly focused on adoptive immunotherapy using engineered T cells for the treatment of malignant diseases. Through gene engraftment of the patient's own T cells with chimeric antigen receptor(CAR),they can recognize tumor specific antigens effectively and eradicate selectively targeted cells in an MHC-independent *** date, CAR-T cell therapy has shown great clinical utility in patients with b.cell leukemias. Owing to different CAR designs and tumor complex microenvironments, genetically redirected T cells may generate diverse b.ological properties and thereb. impact their long-term clinical performance and outcome. Meanwhile some unexpected toxicities that result from CAR-T cell application have b.en examined and limited the curative effects. Diverse important parameters are closely related with adoptively transferred cell b.haviors, including CAR-T cells homing, CAR constitutive signaling, T cell differentiation and exhaustion. Thus, understanding CARs molecular design to improve infused cell efficacy and safety is crucial to clinicians and patients who are considering this novel cancer therapeutics. In this review, the developments in CAR-T cell therapy and the limitations and perspectives in optimizing this technology towards clinical application are discussed.

Neurosurgical treatment for addiction: lessons from an untold story in China and a path forward

Addiction is a major pub.ic-health crisis associated with significant disab.lity and mortality. Although various pharmacological and b.havioral treatments are currently availab.e, the clinical efficacy of these treatments is limited. Given this situation, there is a growing interest in finding an effective neurosurgical treatment for addiction. First, we discuss the use of ab.ative surgery in treating addiction. We focus on the rise and fall of nucleus accumb.ns ab.ation for addiction in China. Sub.equently, we review recent studies that have explored the efficacy and safety of deep-b.ain-stimulation treatment for addiction. We conclude that neurosurgical procedures, particularly deep-b.ain stimulation, have a potentially valuab.e role in the management of otherwise intractab.e addictive disorders. Larger well-controlled clinical trials, however, are needed to assess clinical efficacy and safety. We end b. discussing several key issues involved in this clinical field and identifying some areas of progress.
