Di-and tri-methylation of lysine 36 on histone h3(h3K36me2/3)is catalysed by histone methyltransferase Set2,which plays an essential role in transcriptional *** there is a single h3K36 methyltransferase in yeast and higher eukaryotes,two h3K36 methyltransferases,Ash1 and Set2,were present in many filamentous ***,their roles in h3K36 methylation and transcriptional regulation remained *** with methods of RNA-seq and ChIP-seq,we revealed that both Ash1 and Set2 are redundantly required for the full h3K36me2/3 activity in Magnaporthe oryzae,which causes the devastating worldwide rice blast ***1 and Set2 distinguish genomic h3K36me2/3-marked regions and are differentially associated with repressed and activated transcription,***,Ash1-catalysed h3K36me2 was co-localized with h3K27me3 at the chromatin,and Ash1 was required for the enrichment and transcriptional silencing of h3K27me3-occupied *** the different roles of Ash1 and Set2,in h3K36me2/3 enrichment and transcriptional regulation on the stress-responsive genes,they differentially respond to various stresses in ***,we reveal a novel mechanism by which two h3K36 methyltransferases catalyze h3K36me2/3 that differentially associate with transcriptional activities and contribute to enrichment of facultative heterochromatin in eukaryotes.

The change of fluorescence emission manipulated by spin state transition attracts considerable attention owing to its potential applications in magneto-optical switching ***,we report two two-dimensional(2D)hofmann-type spin crossover(SCO)metal-organic frameworks(MOFs)[Fe^(Ⅱ)(PNI)_(2){Ag^(Ⅰ)(CN)_(2)}_(2)]·ChCl_(3)(3Ag·ChCl_(3))and[FeⅡ(PNI)_(2){AuⅠ(CN)_(2)}_(2)]·ChCl_(3)(3Au·ChCl_(3))based on the fluorescent ligand N-(4-pyridylmethyl)-1,8-naphthalimide(PNI).both complexes exhibit interesting SCO behaviors switched by guest solvent molecules,namely three-step transitions for the solvated complexes and complete onestep hysteretic SCO for the desolvated ones,verified by temperature-dependent magnetic susceptibility measurements,Mossbauer spectra,structural analyses,and differential scanning calorimetry ***,temperature-dependent fluorescence spectra exhibit double peaks(monomer and excimer emission)with both emission peaks change consistent with the change in SCO properties during the solvent molecule *** this study,we integrated guest-responsive SCO behavior into MOFs to manipulate the multistability of spin state and fluorescence switching,providing a rational strategy for the development of stimuli-responsive multifunctional materials.

Radiotherapy(RT) can potentially induce systemic immune responses by initiating immunogenic cell death(ICD) of tumor ***,RT-induced antitumor immunologic responses are sporadic and insufficient against cancer ***,we construct multifunctional self-sufficient nanoparticles(MARS) with dual-enzyme activity(GOx and peroxidase-like) to trigger radical storms and activate the cascade-amplified systemic immune responses to suppress both local tumors and metastatic *** addition to limiting the Warburg effect to actualize starvation therapy,MARS catalyzes glucose to produce hydrogen peroxide(h_(2)O_(2)),which is then used in the Cu^(+)-mediated Fenton-like reaction and RT *** and chemodynamic therapy produce reactive oxygen species in the form of radical storms,which have a robust ICD impact on mobilizing the immune ***,when MARS is combined with RT,potent systemic antitumor immunity can be generated by activating antigen-presenting cells,promoting dendritic cells maturation,increasing the infiltration of cytotoxic T lymphocytes,and reprogramming the immuno suppre ssive tumor ***,the synergistic therapy of RT and MARS effectively suppresses local tumor growth,increases mouse longevity,and results in a 90% reduction in lung metastasis and postoperative ***,we provide a viable approach to treating cancer by inducing radical storms and activating cascade-amplified systemic immunity.

Anti-ganglioside antibodies are associated with delayed/poor clinical recovery in Guillain-barrèsyndrome,mostly related to halted axon ***-linking of cell surface gangliosides by anti-ganglioside antibodies triggers inhibition of nerve repair in in vitro and in vivo paradigms of axon *** effects involve the activation of the small GTPase Rho A/ROCK signaling pathways,which negatively modulate growth cone cytoskeleton,similarly to well stablished inhibitors of axon regeneration described so *** aim of this work was to perform a proof of concept study to demonstrate the effectiveness of Y-27632,a selective pharmacological inhibitor of ROCK,in a mouse model of axon regeneration of peripheral nerves,where the passive immunization with a monoclonal antibody targeting gangliosides GD1a and GT1b was previously reported to exert a potent inhibitory effect on regeneration of both myelinated and unmyelinated *** results demonstrate a differential sensitivity of myelinated and unmyelinated axons to the pro-regenerative effect of *** with a total dosage of 9 mg/kg of Y-27632 resulted in a complete prevention of anti-GD1a/GT1b monoclonal antibody-mediated inhibition of axon regeneration of unmyelinated fibers to skin and the functional recovery of mechanical cutaneous *** contrast,the same dose showed toxic effects on the regeneration of myelinated ***,scale down of the dosage of Y-27632 to 5 mg/kg resulted in a significant although not complete recovery of regenerated myelinated axons exposed to anti-GD1a/GT1b monoclonal antibody in the absence of toxicity in animals exposed to only ***,these findings confirm the in vivo participation of Rho A/ROCK signaling pathways in the molecular mechanisms associated with the inhibition of axon regeneration induced by anti-GD1a/GT1b monoclonal *** findings open the possibility of therapeutic pharmacological intervention targeting Rho A/R

Vertebrate life begins with fertilization,and then the zygote genome is activated after transient silencing,a process termed zygotic genome activation(ZGA).Despite its fundamental role in totipotency and the initiation of life,the precise mechanism underlying ZGA initiation remains *** existence of minor ZGA implies the possible critical role of noncoding RNAs in the initiation of ***,we delineate the expression profile of long noncoding RNAs(lncRNAs)in early mouse embryonic development and elucidate their critical role in minor *** with protein-coding genes(PCGs),lncRNAs exhibit a stronger correlation with minor *** h3K9me3 profiles can be observed between lncRNA genes and PCGs,and the enrichment of h3K9me3 before ZGA might explain the suspended expression of major ZGA-related PCGs despite possessing PolII ***,we identified the presence of PolII-enriched MuERV-L around the transcriptional start site of minor ZGA-related lncRNAs,and these repeats are responsible for the activation of minor ZGA-related lncRNAs and subsequent embryo *** study suggests that MuERV-L mediates minor ZGA lncRNA activation as a critical driver between epigenetic reprogramming triggered by fertilization and the embryo developmental program,thus providing clues for understanding the regulatory mechanism of totipotency and establishing bona fide totipotent stem cells.

bACKGROUND hepatocellular carcinoma(hCC),a major contributor to cancer-related deaths,is particularly prevalent in Asia,largely due to hepatitis b virus *** prognosis is generally *** case report contributes to the medical literature by detailing a unique approach in treating a large hCC through multidisciplinary collaboration,particularly in patients with massive hCC complicated by ruptured bleeding,a scenario not extensively documented *** SUMMARY The patient presented with large hCC complicated by intratumoral *** involved a multidisciplinary approach,providing individualized *** strategy included drug-eluting bead transarterial chemoembolization,sorafenib-targeted therapy,laparoscopic partial hepatectomy,and standardized sintilimab monoclonal antibody *** months after treatment,the patient achieved complete radiological remission,with significant symptom *** studies showed no lesions or recurrence,and clinical assessments confirmed complete *** report is notable as possibly the first docu-mented case of successfully treating such complex hCC conditions through integrated multidisciplinary efforts,offering new insights and a reference for future similar *** This study demonstrated effective multidisciplinary treatment for massive hCC with intratumoral bleeding,providing insights for future similar cases.

The coherent muon-to-electron transition(COMET)experiment is a leading experiment for the coherent conversion of μ^(-)N→e^(-)N using a high-intensity pulsed muon beamline,produced using innovative slow-extraction ***,it is critical to measure the muon beam *** set up a muon beam monitor(MbM),where scintillating fibers woven in a cross shape were coupled to silicon photomultipliers to measure the spatial profile and timing structure of the extracted muon beam for the *** MbM detector was tested successfully with a proton beamline at the China Spallation Neutron Source and took data with good performance in the commissioning *** development of the MbM,including its mechanical structure,electronic readout,and beam measurement results,are discussed.

Monitoring temporal changes in sea level is important in assessing coastal *** level anomalies at a tide gauge station,if kinematically conceived,include systematic variations such as trend,acceleration,periodic oscillations,and random *** them,the non-stationary nature of the random sea level variations of known or unknown origin at coastal regions has been long recognized by the sea level *** study proposes the analyses of subgroups of random residual statistics of a rigorously formulated kinematic model solution of tide gauge variations using X-bar and S control *** approach is demonstrated using Key West,Florida tide gauge *** mean and standard errors of 5-year-long subgroups of the residuals revealed that sea level changes at this location have been progressively intensifying from 1913 to the *** oscillations in sea level at this locality may be attributed partly to the thermal expansion of seawater with increasing temperatures causing larger buoyancy-related sea level fluctuations as well as the intensification of atmospheric events including wind patterns and the impact of changes in inverted barometer effects that will alter coastal risk assessments for the future.

The envisaged design and developme nt of individual mate rials to excavate a broader spectrum of desired properties and offer multiple applications are highly *** this in mind,a series of forsterite-structured Mg2GeO4:Eu3+（1 mol%-9 mol%） nanophosphors was synthesized via a solution combustion method using oxalyl dihydrazide as a *** X-ray diffraction patterns confirm the orthorhombic crystal system with a Puma space *** results clearly show irregularly shaped cluster-like structures with aggregation of the *** diffuse reflectance spectra,the optical energy band gap of the Mg2GeO4:Eu3+（1 mol%-9 mol%） nanophosphors was estimated and obtained to be～4.12-4.32 *** photoluminescence emission spectra exhibit intense peaks at ～579,589,610,661,and 707 nm,which are due to characteristic5D0→7F0,5D0→7F1,5Do→7F2,5D0→7F3and5D0→7F4transitions of Eu3+ions,*** Commission International de L'Eclairage color coordinates are gradually tuned from pale red（0.5980,0.4012） to a pure red（0.6385,0.3611） *** glow peaks showcase excellent super-linear response at low doses of γ-irradiations,indicating that the prepared phosphor can be used in thermoluminescent personal dosimetry which is a biological tissue *** aforementioned results demonstrate that the prepared Mg2GeO4:Eu3+（1 mol%-9 mol%） nanophosphors are considered an excellent candidate for dual applications,i.e.,red component in future generation white-light-emitting diodes（WLEDs） and personal dosimetric applications.
