Genomic characterization of multidrug-resistant extraintestinal pathogenic Escherichia coli isolated from grain culture soils

Multidrug-resistant(MDR)Escherichia coli,mainly extraintestinal pathogenic E.coli(ExPEC),has been widely reported in infections worldwide.In agricultural soils,manure is a hotspot for the dissemination of antimicrobial resistance genes(ARGs)and pathogenic bacteria;however,MDR bacteria have also been reported in soils with no history of manure use.In addition,cross-resistance and co-resistance have been described as responsible for the metal-driven selection of bacteria resistant to antimicrobials.Therefore,the aim of this study was to analyze three MDR E.coli isolates obtained from brazilian grain culture soil samples with no history of manure use by whole-genome sequencing.The MDR E.coli isolates were recovered from soils from corn and coffee fields,and presented resistance toβ-lactams,quinolones,aminoglycosides,tetracyclines,sulphonamides,and dihydrofolate reductase inhibitor.Resistome analysis showed ARGs to several antimicrobials(i.e.,β-lactams,tetracyclines,aminoglycosides,sulphonamides,trimethoprim,phenicols,fosfomycin,and macrolides)as well as several metal resistance genes and antibacterial biocide resistance genes.In addition,known mutations in quinolone-resistance-determining regions of GyrA(Ser83Leu and Asp87Asn),ParE(Ser458Thr),and ParC(Ser80Ile)were also detected.Virulome analysis showed the presence of virulence genes(lpfA,mcmA,gad,mchF,iroN,cma,and iss)associated with ExPEC.Multidrug-resistant ExPEC isolates were assigned to phylogenetic group b1.The presence of MDR b1-ExPEC in soil samples shows the ability of these isolates to survive in soils.This study reports for the first time some sequence types(i.e.,ST345,ST448,and ST1146)of MDR E.coli in brazilian soils.Therefore,these findings contribute to the monitoring of antimicrobial resistance and surveillance studies based on whole-genome sequencing worldwide.

In this paper we study a bilinear optimal control problem for a diffusive Lotka-Volterra competition model with chemo-repulsion in a bounded domain of ℝ^(ℕ),N=2,3.This model describes the competition of two species in which one of them avoid encounters with rivals through a chemo-repulsion mechanism.We prove the existence and uniqueness of weak-strong solutions,and then we analyze the existence of a global optimal solution for a related bilinear optimal control problem,where the control is acting on the chemical signal.Posteriorly,we derive first-order optimality conditions for local optimal solutions using the Lagrange multipliers theory.Finally,we propose a discrete approximation scheme of the optimality system based on the gradient method,which is validated with some computational experiments.

This study presents an investigation of the impact of the quasi-stationary voltage support provided by a voltage source converter(VSC)connected to a single point of a power system.based on the directional derivative concept,an analytical method is developed to quantify the sensitivities of the AC bus voltage with respect to the VSC reactive power control modes.based on a real case study,it is shown that the method applies to VSC units that are part of VSC-HVDC systems,which can operate in a point-to-point or multi-terminal configuration.Time-domain simulations are performed to verify the findings from the application of the analytical method on a reduced size power system.

Metabolic syndrome is a pre-diabetic state characterized by several biochemical and physiological alterations,including insulin resistance,visceral fat accumulation,and dyslipidemias,which increase the risk for developing cardiovascular disease.Metabolic syndrome is associated with augmented sympathetic tone,which could account for the etiology of pre-diabetic cardiomyopathy.This review summarizes the current knowledge of the pathophysiological consequences of enhanced and sustainedβ-adrenergic response in pre-diabetes,focusing on cardiac dysfunction reported in diet-induced experimental models of pre-diabetic cardiomyopathy.The research reviewed indicates that both protein kinase A and Ca^(2+)/calmodulin-dependent protein kinase Ⅱ play important roles in functional responses mediated byβ1-adrenoceptors;therefore,alterations in the expression or function of these kinases can be deleterious.This review also outlines recent information on the role of protein kinase A and Ca^(2+)/calmodulin-dependent protein kinase Ⅱ in abnormal Ca^(2+)handling by cardiomyocytes from diet-induced models of pre-diabetic cardiomyopathy.

background:The usefulness of C-reactive protein(CRP)as a non-specific marker of inflammation during pregnancy and lactation is unclear in impoverished populations where co-existing infections and vitamin deficiencies are common.Methods:This cross-sectional study in Panama recruited 120 pregnant and 99 lactating Ngäbe-buglewomen from 14 communities in rural Panama.Obstetric history,indoor wood smoke exposure,fieldwork,bMI,vitamins A,b_(12),D,and folic acid,and inflammation markers(CRP,neutrophil/lymphocyte ratio(NLR),plateletcrit and cytokines)were measured.Multiple regressions explored both associations of CRP with other inflammatory markers and associations of CRP and elevated CRP based on trimester-specific cut-offs with maternal factors,infections and vitamin deficiencies.Results:CRP was higher in pregnancy(51.4±4.7 nmol/L)than lactation(27.8±3.5 nmol/L)and was elevated above trimester specific cut-offs in 21%of pregnant and 30%of lactating women.Vitamin deficiencies were common(vitamin A 29.6%;vitamin D 68.5%;vitamin b1268%;folic acid 25.5%)and over 50%of women had two or more concurrent deficiencies as well as multiple infections.Multiple regression models highlighted differences in variables associated with CRP between pregnancy and lactation.In pregnancy,CRP was positively associated with greater indoor wood smoke exposure,caries and hookworm and negatively associated with Ascaris and vaginal Lactobacillus and bacteroides/Gardnerella scores.Consistent with this,greater wood smoke exposure,caries as well as higher diplococcal infection score increased the odds of trimester-elevated CRP concentrations whereas longer gestational age lowered the likelihood of a trimester-elevated CRP.During lactation,folic acid deficiency was associated with higher CRP whereas parity,number of eosinophils and Mobiluncus score were associated with lower CRP.Also,a higher bMI and Trichomonas vaginalis score increased the likelihood of an elevated CRP whereas higher parity and number of eosinophils were associated with lower like

Objective: In cesarean section (CS), which, early vs. delayed cord clamping is better for neonatal and maternal hematocrit/hemoglobin level is not yet completely determined. This randomized controlled study attempted to determine this issue. Methods:b> Study population consisted of 64 full-term pregnant women/neonates undergoing planned CS: 32 received early cord clamping (ECC: 15 seconds after birth) and 32 delayed cord clamping (DCC: 90 seconds). We measured and analyzed 1) neonatal hematocrit at the first 24 - 48 hours, and 2) maternal-hemoglobin-change before and after CS.

Objective. The nuclear factor(NF)-κb is one of the pivotal regulators of autoimmunity and inflammation, which has been shown to be activated in the inflamed mucosa of patients with celiac disease(CD). Recently, in the NFKb1 gene promoter region, a common insertion/deletion(-94ins/ delATTG) polymorphism located between two putative key promoter regulatory elements was described. The aim of this study was to investigate the contribution of the -94ins/ delATTG NFKb1 gene promoter functional variant to CD genetic predisposition. Material and methods. A case-control cohort comprising 478 patients with CD and 711 healthy controls as well as a panel of 196 celiac families was genotyped for the 94ins/delATTG NFKb1 polymorphism, using a polymerase chain reaction (PCR) method combined with fluorescence technology. Results. We found no statistically significant differences between CD patients and controls when the -94ins/delATTG genotype and allele distributions were compared. Accordingly, the familial analysis did not reach statistically significant deviation in the transmission of -94ins/delATTG alleles to the affected offspring. Conclusions. From these results, it could be suggested that the -94ins/delATTG NFKb1 polymorphism does not play a major role in CD susceptibility.

Rationale: Acute respiratory failure is an uncommon complication of pregnancy. However, it is the most frequent organ dysfunction associated with obstetric admissions to an intensive care unit. The obstetric population is a different group due to its physiology and the presence of the fetus that lacks evidence in the literature within the subject of ventilatory support. Noninvasive positive pressure ventilation (NIPPV) is often avoided due to the lack of knowledge on the safety and efficacy of this modality. The gravity field and gravity data reduction across the continental area of Nigeria

This research presents the variation of the gravity field and associated gravity field components over the continental area of Nigeria to provide data for geoscience research,geodetic and engineering works,aerodynamic studies and deep crustal inferences.Accurate positions and elevations were observed at 58 of the 59 base stations of the Primary Gravity Network of Nigeria(PGNN),whose absolute gravity values had been accurately determined.The absolute gravity values were plotted against their respective positions to reveal the distribution pattern and strength of the gravity field within the study area.Theoretical gravity values at each base station were generated using the Somigliana's equation.The free-air gravity and free-air anomaly gravity values were generated with respect to the World Geodetic System 1984(WGS84)ellipsoid using GPS-derived elevation data.Then,the perturbing potential,free-air gravity with respect to the geoid,and the indirect effects were evaluated.The average of the indirect effects was used to adjust the WGS84 gravity formula to produce a gravity formula that better approximates the geoid across the continental area of Nigeria,compatible with the heights measured relative to the geoid,which can serve as a reference for establishing a vertical height control.The bouguer gravity and bouguer gravity anomalies across Nigeria revealed a“trans-southern gravity high strip”interpreted to be associated with mantle upwelling.Two new major mega-lineaments related to mantle upwelling were mapped.A batholith province trending NWeSE was delineated,occurring from north central Nigeria to the north western region and containing closures of“bouguer gravity lows”interpreted as batholiths.A separate closure of“bouguer gravity low”was detected at Azare,north eastern Nigeria,which may be due to the presence of intrusive granitic body.It is recommended that the mantle structure beneath“the trans-southern gravity high strip”,“delineated batholith province”and“isolated gravity closures”around the northeast of Nige
