Effect of glutathione on brain nitric oxide levels in an experimental epilepsy mouse model

BACKGROUND： Oxidative stress plays an important role in the pathophysiology of epilepsy. Glutathione, known as one of the compounds of antioxidant defense, has been shown to inhibit convulsions. Nitric oxide has a proconvulsant effect on a pentylenetetrazole-induced animal model. OBJECTIVE： To evaluate the effects of glutathione administration on nitric oxide levels in brain regions of convulsive and kindling pentylenetetrazole-induced seizure models. DESIGN, TIME, AND SETTING： A randomized, controlled, animal experiment. The study was performed at the Department of Physiology, Gaziantep University and Department of Chemistry-Biochemistry,Kahramamaras Sutcu Imam University in 2006. MATERIALS： Pentylenetetrazole and glutathione were purchased from Sigma, USA. METHODS： A total of 80 mice were assigned to 8 groups （n = 10）： normal control, saline control （1 mL normal saline）, convulsive pentylenetetrazole （single intraperitoneal administration of pentylenetetrazole, 60 mg/kg）, convulsive pentylenetrazole plus glutathione （single administration of 60 mg/kg pentylenetetrazole and 200 mg/kg glutathione）, five-dose glutathione （intraperitoneal injection of 200 mg/kg glutathione respectively at 1, 3, 5, 7, and 10 days）, single-dose glutathione （single administration of 200 mg/kg glutathione）, pentylenetetrazole kindling （intraperitoneal administration of pentylenetetrazole of 40 mg/kg at 1,3, 5, 7, and 10 days）, and pentylenetetrazole kindling plus glutathione group （intraperitoneal injection of 40 mg/kg pentylenetetrazole and 200 mg/kg glutathione respectively at 1, 3, 5, 7, and 10 days）. MAIN OUTCOME MEASURES： All mice were sacrificed 1 hour after the last administration. Brain nitric oxide levels were determined by spectrophotometry. RESULTS： There were no significant differences in nitric oxide levels between the normal control, saline control, five-dose glutathione, and single-dose glutathione groups （P 〉 0.05）. This study was conducted to understand the cellular proliferative effect of Photobiomodulation Therapy(PBMT)on thawed dental pulp stem cells(DPSCs)stored for 2 *** this purpose,cells were exposed to PBMT for short period of time to evaluate the most appropriate PBMT parameter for stimulating cellular proliferation that can be used for future tissue engineering *** characterized DPSCs were seperated into three groups according to the laser energy densities(5 J/cm^(2) or 7 J/cm^(2))applied and a group was served as control in which cells did not receive any laser *** cells in laser-irradiated groups were further divided into two subgroups according to the period of application(24 h and 0 h)and exposed to Gallium-Aluminum-Arsenide diode laser *** viability and the proliferation rate of the cells were analyzed with the 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide(MTT)assay,any PBMT related cellular cytotoxicity were determined by performing a lactate dehydrogenase assay(LDH)and statistical analysis of data were *** percentage of proliferation seemed to increase upon laser therapy in both different doses of irradiation(5 J/cm^(2) and 7 J/cm^(2)).DPSCs showed significantly higher proliferation rate upon 7 J/cm^(2) irradiation in both 0 h and 24 h when compared to control ***,DPSCs irradiated with 5 J/cm^(2) dose induced relatively lower proliferation rate when compared to 7 J/cm^(2) dose of *** to the LDH data,PBMT exposure did not show any significant cytotoxicity at both energy densities in all different time *** at 7 J/cm^(2) should be an effective parameter to stimulate proliferation of long-term cryopreserved DPSCs in a short term time *** therapy may be an upcoming tool for future tissue enngineering and regenerative dentistry applications.

Comparison of a modified anoscope and the purse-string anoscope in stapled haemorrhoidopexy

AIM:To compare the results of the anoscope of the PPH kit and a modified anoscope during stapled haemorrhoidopexy. METHODS:The hospital records of 37 patients who underwent stapled haemorrhoidopexy between 2001 and 2006 were *** purse-string suture anoscope in the PPH kit was used on 15 patients(Group 1), and the modified anoscope was used on 22 patients (Group 2).Demographic characteristics of the patients, operation time,surgeon's performance,analgesic requirement,and complications were compared. RESULTS:Operation time was significantly longer in Group 1(42.0±8.4 min vs 27.7±8.0 min,P=0.039). The surgeons reported their operative performance as significantly better in Group 2(the results of the assessments were poor in ten,medium in four and good in one in Group 1,while good in all patients in Group 2,P<0.001).The need for haemostatic sutures was significantly higher in Group 1(six cases)and was needed in two cases in Group 2(P=0.034). CONCLUSION:Operation time decreased and thesurgeon's satisfaction increased with use of the modified anoscope,and fewer haemostatic sutures were required if the surgeon waited longer before and after firing the stapler.

Evaluation of choroidal thickness using spectral-domain optical coherence tomography in patients with severe obstructive sleep apnea syndrome: a comparative study

AIM: To assess choroidal thickness in patients with severe obstructive sleep apnea syndrome(OSAS) and compare them with healthy controls, using spectral domain optical coherence tomography(OCT).METHODS: In this observational, cross-sectional study,choroidal thicknesses of 23 newly severe OSAS patients and 23 body mass index- age- and sex-matched healthy subjects were measured using a high-speed, high-resolution frequency domain- OCT device( λ = 840 nm,26 000 A-scans/s, 5 μm axial resolution). All patients underwent a complete ophthalmic examination before the measurements. OCT measurements were taken at the same time of day(9:00 a.m.), in order to minimize the effects of diurnal ***: There was a statistically significant difference in median choroidal thickness between the OSAS patients(201 μm; range 145-237 μm) and the controls(324 μm; range 296-383 μm;P <0.001). There were significant differences at all measurement points(P <0.001 for all). The apnea-hypopnea index(AHI)values were more than 30 in all OSAS patients and the mean AHI was 48.57 ±6.54. The interexaminer intraclass correlation coefficient(ICC) for the mean choroidal thickness was 0.938(95% CI, 0.908-0.985) and ICC was greater than 0.90 for all measurement ***: The decreased choroidal thickness of patients with severe OSAS might be related to the the autonomic disregulation associated with this *** studies are needed to evaluate the etiopathologic relationship between choroidal thickness and OSAS.

Tens of thousands of people worldwide became infected with syndrome coronavirus-2(SARS-CoV-2).Death rate in general population is about 1%-6%,but this rate rises up to 15%in those with *** publications showed that the clinical progression of this disease in organ recipients is more destructive,with a fatality rate of up to 14%-25%.We aimed to review the effect of pandemic on various transplantation *** disease 2019(COVID-19)has not only interrupted waiting list patients’lives;it has also impacted transplantation strategies,transplant surgeries and broke donation ***-19 was directly and indirectly accountable for 73%surplus in mortality of this population as compared to wait listed patients in earlier *** impact of chronic immunosuppression on outcomes of COVID-19 remains unclear but understanding the immunological mechanisms related to the virus is critically important for the lifetime of transplantation and immune suppressed *** is hard to endorse changing anti-rejection therapy,as the existing data appraised is not adequate to advise substituting tacrolimus with cyclosporine during severe COVID-19 disease.

Primula vulgaris extract induces cell cycle arrest and apoptosis in human cervix cancer cells

Primula vulgaris belongs to the genus Primula, members of which are frequently used in folk medicine. Various studies have investigated the cytotoxic effect of different Primula species, but there have been limited studies on the cytotoxic effect of P vulgaris. The aim of this study was to investigate the cytotoxic effects, and possible mechanisms involved, of P vulgaris flower extract on human cervical cancer （HeLa） cells. The cytotoxic effect of the extract on HeLa cells was revealed using the MTT assay. Mechanisms involved in the extract＇s cytotoxic effect were then investigated in terms of apoptosis, mitochondrial membrane potential, and the cell cycle, using fluorometric methods. P vulgaris flower extract exhibited selective cytotoxic effects against HeLa cells by arresting their cell cycle at the S phase, and inducing the number of apoptotic cells compared to normal fibroblast cells by reducing mitochondrial membrane potential in a concentration-dependent manner. This is the first study to reveal the antiproliferative effect ofP. vulgaris flower extract. Further studies are now needed to identify the cytotoxic molecules in the extract and their mechanisms.

Environmental control of daily stem radius increment in the montane conifer Cedrus libani

Characteristics of sildenafil erections in healthy young men

Aim： To determine the effect of sildenafil citrate on the nocturnal penile erections （i.e. time to onset, the duration of erection, and the interval between first and second erections） of healthy young men. Methods： Twenty-two potent men, 23-29 years old, were recruited for the study. All subjects completed three sessions over consecutive nights using the RigiScan monitoring device （Dacomed, Minneapolis, USA）. After a first night of adaptation, night 2 records were their baseline values, and on night 3 they received 100 mg of sildenafil citrate. Statistical comparisons were done between the second and third night data. Results： The mean time to onset of the first erection with sildenafil citrate was （34 ± 18） min, whereas it was （74 ± 24） min （P 〈 0.001） without sildenafil citrate. The number of erections observed during the first 5 h after sildenafil citrate medication was 3.6 ± 0.5 in contrast to 2.4 ± 0.5 with no medica- tion （P = 0.001）. The interval between first and second erections was shorter with sildenafil citrate：（52 ± 26） min vs. （85 ± 34） min （P = 0.01）. The duration of the last erection was statistically significantly longer with the sildenafil citrate：（64 ± 33） min vs. （42 ± 28） min （P 〈 0.01）. Conclusion： Healthy young men achieved erection within 34 min after sildenafil citrate administration, which is shorter than the 1 h interval proposed by the manufacturer. The interval between the first and second erections was shorter and the duration of the last nocturnal erection was longer.

FRP筋超高性能海水海砂混凝土梁抗剪性能研究

为研究纤维增强复合材料(Fiber Reinforced Polymer,FRP)筋超高性能海水海砂混凝土(Ultra-high Performance Seawater Sea-sand Concrete,UHPSSC)梁的抗剪性能,采用三点弯曲试验,探究剪跨比、箍筋间距和筋材类型(CFRP、BFRP、HFRP)对FRP-UHPSSC梁抗剪性能的影响规律.试验结果表明:FRP-UHPSSC梁在加载时裂缝发展迅速,破坏时具有明显的脆性特征;跨中荷载-挠度曲线在首条裂缝出现前后均呈现出双线性变化,裂缝出现后梁的荷载-挠度关系曲线斜率变小,挠度增长加快;FRP-UHPSSC梁的抗剪承载力随剪跨比和箍筋间距的增大而减小,剪跨比相同的试验梁,其开裂荷载基本相同;筋材的弹性模量越大,梁的抗剪承载力越高,同时也越能抑制梁的竖向变形;最后采用4部规范中的抗剪公式对试验工况进行分析,规范公式对BFRP-UHPSSC梁和HFRP-UHPSSC梁抗剪承载力计算的最小误差分别超过40%和30%,筋材类型不同的试验工况抗剪承载力计算结果差异较大.

Current and future surgery strategies for spinal cord injuries

Spinal cord trauma is a prominent cause of mortality and morbidity. In developed countries a spinal cord injury(SCI) occurs every 16 min. SCI occurs due to tissue destruction, primarily by mechanical and secondarily ischemic. Primary damage occurs at the time of the injury. It cannot be improved. Following the primary injury, secondary harm mechanisms gradually result in neuronal death. One of the prominent causesof secondary harm is energy deficit, emerging from ischemia, whose main cause in the early stage, is impaired perfusion. Due to the advanced techniques in spinal surgery, SCI is still challenging for surgeons. Spinal cord doesn't have a self-repair property. The main damage occurs at the time of the injury primarily by mechanical factors that cannot be improved. Secondarily mechanisms take part in the following sections. Spinal compression and neurological deficit are two major factors used to decide on surgery. According to advanced imaging techniques the classifications systems for spinal injury has been changed in time. Aim of the surgery is to decompress the spinal channel and to restore the spinal alinement and mobilize the patient as soon as possible. Use of neuroprotective agents as well as methods to achieve cell regeneration in addition to surgery would contribute to the solution.
