Cleidocranial dysplasia with a rare mutation:Study of a family with review of literature

Introduction: The present study was aimed at advancing the understanding of the pathogenesis of cleidocranial dysplasia (CCD) by presenting a case study based on history, physical examination, typical radiological features, and molecular analysis and a review of the literature. Methods: This study began with a 23-year-old boy (proband) who was referred to the department of oral and maxillofacial radiology with chief complaint of the upper-left first molar tooth and routine dental examination. While evaluating the panoramic radiograph, the patient had approximately 57 teeth in his both of the jaws. Clinical, radiographical and molecular features of the proband, two siblings and their parents were examined and then, DNA analysis was performed. Results: Overall, we present 3 CCD patients with a mutation in the VWRPY motif. The deletion of c. 1754_1757 delTTTG (NM_001024630.2) is determined and it leads to a frame shift mutation and stop codon, p. V585Gfs56X. Conclusions: The present study emphasized the importance of further clinical and molecular investigation when even a single case of CCD is identified within a family. This is the first study performed in Turkey about a family with a mutation in the VWRPY motif. Genotype-phenotype association studies in individuals with CCD are necessary to provide important insights into molecular mechanisms associated with this disease.

目的探讨激酶纤溶酶原激活剂(PLAU)和蛋白激酶B1(AKT1)在口腔鳞癌(OSCC)及正常组织中的表达、预后和两分子在OSCC组织中的相关性。方法采用免疫组化法检测70例OSCC及50例正常组织中PLAU和AKT1的表达水平,分析PLAU和AKT1表达与临床病理特征、预后和两分子在OSCC组织中的相关性,并使用生物信息学数据库进一步验证实验结果。结果PLAU和AKT1在OSCC组织中的表达高于正常组织(P<0.05);Kaplan-Meier分析显示,PLAU和AKT1低表达患者的生存时间长于高表达患者(P<0.05);Spearman秩和相关检验显示,在OSCC组织中,PLAU和AKT1表达有着较强的相关性(r=0.357,P<0.05);GEPIA生物信息学数据库分析结果与实验结果一致。结论PLAU和AKT1在OSCC组织中高表达,且与患者不良预后相关,在OSCC组织中PLAU和AKT1存在相关性。

Cyanocobalamin and 25-Hydroxy Vitamin D Levels in Gout Patients: An Overlooked Issue

Gout is one of the most frequent type of inflammatory arthritis in developed countries. The elevation of serum uric acid levels and the deposition of monosodium urate crystals in joints and/or soft tissues are the mechanisms of pathogenesis. Uric acid is a product of the metabolic cleavage of purine nucleotides and organ meats, beef, pork, and lamb, anchovies, sardines, herring, mackerel, scallops, gravy and beer are known to be very rich in purine. On the other hand, some of these foods are also the main sources of vitamin B12 (cyanocobalamin). As a chronic inflammatory arthritis corticosteroids are frequently prescribed for gout patients, meaning a higher risk for osteoporosis which may be blocked by daily calcium and vitamin D replacement. However, there are no recommendations about screening or replacement of Gout patients for vitamin D and B12. Herein, we evaluated our patients retrospectively to document their vitamin levels and also to find the factors associated with vitamin deficiency. Totally, 90 patients, 71 (79.9%) male and 19 (20.1%) female patients with a median diagnostic age of 55 (19 - 80) were included. Thirty six (40%) patients were newly diagnosed (group 1) but 54 (60%) patients had established diseases (group-2) with median disease duration of 36 (11-240) months. Nearly half (47.2%) of the patients in group-1 and 37% of the patients in group-2 had vitamin B 12 deficiency. Similarly, 38.9% in each group had vitamin D deficiency and 52.8% in group-1 and 44.4% in group-2 had vitamin D insufficiency. In conclusion, we strongly recommend routine screening and replacement of vitamin B12 and D for patients with Gout.

Integrated Assessment of Cardiac PET/MRI: Co-Registered PET and MRI Polar Plots by Mutual MR-Based Segmentation of the Left Ventricular Myocardium

Background: In the present study, we sought to describe a procedure for the creation of co-registered positron emission tomography (PET) and magnetic resonance imaging (MRI) polar plots of cardiac PET/MRI examinations, validate the resulting plots against available standard methods in patients with myocardial infarction and provide examples that demonstrate the advantage of the novel approach over existing standards. Methods: Co-registered LGE and PET short-axis images were transformed into polar maps based on a radial sampling pattern. LGE was automatically detected using an automated thresholding algorithm (ATA). In 20 PET/MRI examinations in patients with acute myocardial infarction, agreement between manual LGE assessment and the ATA classification was calculated. Also agreement between MRI-segmentation based PET polar plots and standard PET polar plots (created with the Corridor4DM software package) was assessed. Results: No statistically significant difference in infarct sizes between manual and ATA segmentation was found (p = 0.12). Both methods were highly correlated (Pearson’s r = 0.96, p Conclusion: A straightforward software approach for the creation of co-registered PET and MRI polar plots was described and successfully demonstrated in PET/MRI studies of myocardial infarction and inflammation.

An Optimum and Effective Drawing Teaching System in Engineering

In this study, the authors shall illustrate the difficulties, pros and cons of "Technical Drawing" courses in engineering curriculums. The authors also show the efficiency, effectiveness of students" grasping the engineering drawing concepts in depth. The authors have conducted a survey on students and their instructors for teaching the course. Whether the course should be taught with manual handling and then computerized or starting with computer aid and finishing the content of the course, the authors shall also go in detail of syllabus how the drawing rooms and aid tools will be. This part of study shall look into the ergonomics, safety and conditions of classrooms. Responses of 300 students will be shared in this presentation. Statistical analysis will detect the effects of different factors involved in the questionnaire.

Inhibition of Biofilms by Non-Thermal Plasma Treated Novel Solutions

Biofilms act as a reservoir of infection, and periodically release cells in vicinity that are capable of developing new biofilm colonies and disseminate infection. Many chronic bacterial infections are serious that are associated with biofilms and have high morbidity and mortality, partly due to their higher resistance to antimicrobial agents, and partly due to lack of strong biocides which can efficiently treat and inhibit biofilm formation. We recently demonstrated that nonequilibrium non-thermal dielectric-barrier discharge plasma (Plasma) can also be applied to control pathogens via applying treated-liquids, and these liquids acquire broad-spectrum antimicrobial properties. In present studies we demonstrated a range of plasma-activated simple chemical solutions which significantly inhibited biofilm formation by multidrug-resistant bacterial pathogens. Plasma-activated methionine solution exhibited strong inhibitory activity against the biofilms of car-bapenem-resistant Acinetobacter baumannii, methicillin-resistant Staphylococcus aureus, metal-lo-β-lactamase (NDM1)-positive Klebsiella pneumoniae, and Enterococcus faecalis, and prevented the formation of biofilms by about 70% as compared to untreated controls in single exposure. In addition to inhibition of biofilm formation, a complete inactivation of biofilm-embedded bacterial cells was observed in less than 30 minute’s exposure to candidate plasma-activated methionine solution. These findings suggest that plasma-activated solutions have a potential to prevent biofilm formation, and as biofilm inhibitor.

目的探讨超声灰度直方图对腮腺多形性腺瘤与腺淋巴瘤的鉴别诊断价值。方法对18例多形性腺瘤(PA)与13例腺淋巴瘤(AL)患者的术前超声图像进行灰度直方图分析,比较PA与AL组患者直方图的均值、方差、偏度、峰度及第1、10、50、90、99百分位数的差异。应用受试者工作特征(ROC)曲线计算各参数的鉴别诊断效能。结果 PA组的方差高于AL组,偏度和峰度值低于AL组,其差异具有统计学意义(P0.05)。ROC曲线分析显示,峰度值最具鉴别诊断效能,曲线下面积(AUC)为0.744 (P=0.022),最佳临界值为1.71,敏感度为76.9%,特异度为61.1%,约登指数为0.380。方差的鉴别诊断效能、最佳临界值、敏感度、特异度及约登指数分别为0.735 (P=0.028)、864.94、72.2%、76.9%及0.491。结论超声灰度直方图对PA与AL具有一定的鉴别诊断价值。

Cerebral magnetic resonance imaging in quiescent Crohn's disease patients with fatigue

AIM To evaluate brain involvement in quiescent Crohn's disease (CD) patients with fatigue using quantitative magnetic resonance imaging(MRI).METHODS Multiple MRI techniques were used to assess cerebral changes in 20 quiescent CD patients with fatigue(defined with at least 6 points out of an 11-point numeric rating scale compared with 17 healthy age and gender matched controls without fatigue. Furthermore, mental status was assessed by cognitive functioning, based on the neuropsychological inventory including the different domains global cognitive functioning, memory and executive functioning and in addition mood and quality of life scores. Cognitive functioning and mood status were correlated with MRI findings in the both study *** Reduced glutamate + glutamine (Glx = Glu + Gln) concentrations (P = 0.02) and ratios to total creatine(P = 0.02) were found in CD patients compared with controls. Significant increased Cerebral Blood Flow(P = 0.05) was found in CD patients (53.08 ± 6.14 mL/100 g/min) compared with controls (47.60 ± 8.62 mL/100 g/min). CD patients encountered significantly more depressive symptoms (P < 0.001). Cognitive functioning scores related to memory (P = 0.007) and executive functioning (P = 0.02) were lower in CD patients and both scores showed correlation with depression and anxiety. No correlation was found subcortical volumes between CD patients and controls in the T1-weighted analysis. In addition, no correlation was found between mental status and MRI *** This work shows evidence for perfusion, neurochemical and mental differences in the brain of CD patients with fatigue compared with healthy controls.
