Characterization of flgK gene and FlgK protein required for H pylori Colonization-from cloning to clinical relevance

AIM： To characterize the role of flgK and its protein product in Hpylori colonization. METHODS： The PCR cloning method identified the flgK gene. An isogenic flgK mutant was constructed by gene replacement and confirmed by Southern blot analysis and PCR analysis. The recombinant FlgK protein （r-FlgK） was purified. Electron microscopy （EM） was applied to demonstrate the flagella of H pylori. An in vitro motility test was assessed in semisolid medium. The densities of H pylori colonization with either the wild-type strain or its flgK mutant were compared among BALB/c mice with or without pre-immunization with r-FlgK. The serological responses to r-FlgK were analyzed for 70 clinical patients with different densities of H pylori colonization. RESULTS： From a duodenal ulcer strain, the flgK gene was cloned and it contained 1821 bp, with a 95.7% identity to the published sequences. No flagella were observed under EM for the mutant strain, which had a loss of motility. Hpylori density was lower in the BALB/c mice inoculated by the mutant or with pre-immunization with r-FlgK compared to unimmunized mice or mice inoculated by the wild-type strain （P 〈 0.05）. In the H pylori-infected patients, the serological responses to r-FlgK were uniformly low in titer.CONCLUSION： FlgK encoded by flgK is important for flagella formation and H pylori motility. Deficiency in FlgK or an enhanced serological response to r-FlgK can interfere with Hpylori colonization. FlgK of Hpylori could be a novel target for vaccination.

Lithium ion batteries(LIBs)that can be operated under extended temperature range hold significant application *** in this work,we successfully synthesized Co2V2O7 electrode with rich porosity from a facile hydrothermal and combustion *** applied as anode for LIBs,the electrode displayed excellent stability and rate performance in a wide range of ***,a stable capacity of 206 mAhg 1 was retained after cycling at a high current density of 10 A·g-1 for 6,000 cycles at room temperature(25℃).And even when tested under extreme conditions,i.e.,-20 and 60℃,the battery still maintained its remarkable stability and rate *** example,at-20℃,a capacity of 633 mAh·g 1 was retained after 50 cycles at 0.1 A·g 1;and even after cycling at 60℃ at 10 A·g-1 for 1,000 cycles,a reversible capacity of 885 mAh·g-1 can be *** believe the development of such electrode material will fciliate progress of the next-generation LIBs with wide operating windows.

Synthesis and characterization of Cu-Pt/MWCNTs for use in electrocatalytic applications

Improving catalyst performance by studying the preparation parameters, material structures and analysis methods is important work for catalyst researchers. In this study, Cu-Pt nanocomposites with different metal ratios were synthesized on MWCNTs using a polyol method. The structures and morphologies of the nano hybrid catalysts were analyzed by XRD and TEM. The XRD results indicate that Pt and Cu formed a bimetallic solid solution. The average diameters of the Pt and Cu-Pt nanoparticles on MWCNTs were between 3-10 nm and 5-15 nm, respectively. The electrocatalytic behavior of methanol oxidation reactions was investigated by cyclic voltammery （CV）. Cu（20%）/[Pt/MWCNTs（1：4）], which contained an optimal degree of solid solution, enhanced the methanol oxidation to the greatest degree.

Boundary-effect and scale-heating rate equivalence effect of cracking behavior in the rock models subjected to heating from the central borehole

Thermal cracking is broadly observed in rock engineering.A finite element numerical model which considers the heterogeneity of rock materials and the damage evolution process was used to simulate the thermal cracking behavior of square rock samples heated from the central *** thermal and mechanical behaviors of two cases,i.e.,the case with large size but low heating rate and the case with small size but high heating rate were compared to study the crack initiation location in the models with different model sizes and heating *** simulated stress and temperature fields,as well as the failure pattern,were in good agreement with the experimental *** temperature and thermal stress distribution during the heating process in both cases indicated that high tensile stress was concentrated around the thermal gradient front,which resulted in the cracks initiating at the location with a certain distance away from the *** results show that under the same heating rate,crack initiation location moves outwards with the increment of the model size then remains approximately at one location,which reflects the ***,the results indicate that the relative crack initiation locations in two cases are nearly the same if the ratio between the heating rate in two cases(Tx/Ty)nearly equal to the square of the inverse ratio of corresponding model side lengths((ay/ax)2).This concept is named the scale-heating rate equivalence effect in this *** is beneficial for studying thermal cracking of rock both numerically and experimentally since the model size can be significantly decreased.

Interactive modeling of lofted shapes from a single image

Modeling the complete geometry of general shapes from a single image is an ill-posed *** hints are often incorporated to resolve ambiguities and provide guidance during the modeling process. In this work, we present a novel interactive approach for extracting high-quality freeform shapes from a single image. This is inspired by the popular lofting technique in many CAD systems, and only requires minimal user input. Given an input image, the user only needs to sketch several projected cross sections, provide a"main axis", and specify some geometric relations. Our algorithm then automatically optimizes the common normal to the sections with respect to these constraints,and interpolates between the sections, resulting in a high-quality 3 D model that conforms to both the original image and the user input. The entire modeling session is efficient and intuitive. We demonstrate the effectiveness of our approach based on qualitative tests on a variety of images, and quantitative comparisons with the ground truth using synthetic images.

Electrochemical decolorization of dye wastewater by surface-activated boron-doped nanocrystalline diamond electrode

Complex organics contained in dye wastewater are difficult to degrade and often require electrochemical advanced oxidation processes（EAOPs） to treat it. Surface activation of the electrode used in such treatment is an important factor determining the success of the *** performance of boron-doped nanocrystalline diamond（BD-NCD） film electrode for decolorization of Acid Yellow（ay-36） azo dye with respect to the surface activation by electrochemical polarization was studied. Anodic polarization found to be more suitable as electrode pretreatment compared to cathodic one. After anodic polarization, the originally H-terminated surface of BD-NCD was changed into O-terminated, making it more *** to the oxidation of surface functional groups and some portion of sp2 carbon in the BD-NCD film during anodic polarization, the electrode was successfully being activated showing lower background current, wider potential window and considerably less surface activity compared to the non-polarized one. Consequently, electrooxidation（EO） capability of the anodically-polarized BD-NCD to degrade ay-36 dye was significantly enhanced, capable of nearly total decolorization and chemical oxygen demand（COD） removal even after several times of re-using. The BD-NCD film electrode favored acidic condition for the dye degradation; and the presence of chloride ion in the solution was found to be more advantageous than sulfate active species.
