Transgenic Date Palm Containing Endotoxin Cry3Aa Gene

Date palm, like all other crops, is very sensitive to the injury by many insect pests, which may lead to the death of the affected plant and causes decrease in yield. In the present study, an efficient Agrobacterium for genetic transformation was successfully achieved for well known date palm （Phoenix dactylifera L.） cv. Medjool and Khalas using callus as explant. Embryogenic callus were recorded 100% mortality when cultured on MS medium containing 100 mg/L kanamycin with different cultivars, thus it was chosen for the selection of transformed explants. Embryogenic callus of Medjool and Khalas were incubated with Agrobacterium tumefaciens strain LBA 4404 for 0.5, 1, 2, 4 and 24 h on LB medium. After the incubation periods, embryogenic callus was transferred to MS medium with 0.1 mg/L NAA, 0.05 mg/L BA, 250 mg/L carbenicillin and 100 mg/L kanamycin for detection of transgenic embryogenic callus. Polymerase chain reaction （PCR） was used for the rapid screening of Cry3Aa gene. For screening, total genomic DNA was isolated from transformants. Using primer specific to Cry3Aa gene （forward and reverse）, a PCR product with a size of about 2,000 bp was amplified when all nucleic acid from the transformants were utilized as templates. PCR analysis confirmed the appearance of the transgene of 2,000 bp in one individual plantlet. Presence and integration of foreign Cry3Aa gene in regenerated kanamycin resistant embryogenic callus was also confirmed by Southern blot hybridization. It was found that one transgenic embryogenic callus for both Medjool and Khalas showed a single copy of gene integration. These results signify the successful transfer of Cry3Aa gene into date palm plant.

Effect of Titania Additive on Structural and Mechanical Properties of Alumina–Fluorapatite Composites

Mechanical properties of alumina-fluorapatite composites with different titania additive amounts （0, 0.5, 1, 1.4, 2, 3, 4 and 5 wt%） have been investigated between 1200 and 1600℃. The optimum values of densification and mechanical properties of composites have been reached with 1.4 wt% of titania after the sintering process at 1500℃ for 1 h. Thus, the rupture strength of alumina-26.52 wt% Fap-1.4 wt% TiO2 reaches 75 MPa. At higher temperature and beyond 1.4 wt% TiO2 ,the densification and mechanical properties were hindered by the formation of both intergranular porosity and secondary phase. X-ray diffraction （XRD） analysis of alumina-Fap-TiO2 composites shows the formation of aluminium titanate （Al2O3-TiO2：Al2TiO5 ）. The 27Al magic angle scanning nuclear magnetic resonance analysis of Al2O3-Fap-TiO2 composites reveals the presence of octahedral and pentahedral aluminium and novel environment relative to tetrahedral aluminium sites.

Aim： To investigate the effects of crude garlic on adult male rat reproductive functions. Methods： Thirty male rats were divided into five groups： group 1 （untreated） and groups 2, 3, 4 and 5 were fed for 30 days with 5%, 10%, 15% and 30% crude garlic, respectively. Testes and accessory organs were weighed and some markers were assessed. Light and electron microscopy observations were also performed. Results： A significant decrease was observed in the body weight of groups 4 （14%; P 〈 0.01） and 5 （20%; P 〈 0.01）; of the prostate weight in group 5 （29.1%; P 〈 0.05） and of seminal vesicle weight in groups 3 （14.4%; P 〈 0.01）, 4 （18.3%; P 〈 0.01） and 5 （27.3%; P 〈 0.01）. In contrast, testis and epididymis weights were unchanged. In epididymis tissue, the alpha glucosidase activity and the spermatozoa density were unchanged. The treatment resulted in a significant decrease in testosterone serum levels in groups 3 （77.3%; P 〈 0.01）, 4 （77.3%; P 〈 0.01） and 5 （90.9%; P 〈 0.01）, associated with a significant increase in LH serum levels （P 〈 0.01）. Testicular histology showed a dose-dependent increase in the percentage of empty seminiferous tubules. Moreover, testicular function was affected; a significant decrease in phosphatase acid activity （P 〈 0.01） and testosterone （P 〈 0.05） contents were observed. Conclusion： Crude garlic consumption during 1 month reduced testosterone secretion and altered spermatogenesis at 10%, 15% and 30% doses.

Elaboration and characterization of alumina - fluorapatite composites

Alumina and fluorapatite powder were mixed in a wet medium in order to elaborate biphasic ceramics composites. The effect of fluorapatite addition (26.5 wt%) in the densification and the mechanical properties of the alumina matrix were measured. The phase developments have been systematically analysed by scanning electronic microscopy, X-ray diffraction, Infrared spectroscopy and 31P and 27Al magic angle scanning nuclear magnetic resonance. The Brazilian test was used to measure the mechanical resistance of alumina - 26.5 wt% fluorapatite composites. The densification and strength rupture of composites increase versus sintering temperature and holding time. At 1600°C, the composites densities reached 85% and the rupture strength was about 22 MPa. Also, the composites sintering at 1500°C for 5 hours provides samples with similar density and having higher mechanical resistance, above 26 MPa. For longer holding times, the mechanical properties were hindered by the exaggerated grain growth and the formation of intragranular porosity. From 1400°C, the characterization of the alumina - 26.5 wt% fluorapatite composites indicates the formation of calcium aluminates.

Protective Effect of <i>Lepedium sativum</i>against 2, 4-Dichlorophenoxyacetic Acid Hepatotoxicity: An Electron Microscopic Study

Medicinal plants have been used to treat many diseases with fewer side effects compared to clinical medicines. The need for treatment by medicinal plants has been increasing in response to the accumulation of environmental pollutants. The herbicide 2, 4-Dichlorophenoxyacetic acid has been considered as fast spreadable environmental pollutant, which may cause many disorders in animals and possibly in humans. The aim of this study is to assess the protective and/or curative effect of Lepidium sativum against hepatotoxicity induced by 2, 4-D herbicide in rats. 25 Albino Wistar rats were grouped into 4 groups: a control group, the curative group treated for 4 weeks with LS water extract after a single dose 2, 4-D LD50 administration, protective group which was treated for 4 weeks with LS water extract prior to the 2, 4-D LD50 administration, and LS only group that was given the LS water extract as a positive control. Using transmission electron microscope, the histological features showed that curative group hepatocytes still suffered from focal necrosis and vacuolation of hepatocytes as a result of 2, 4-D toxic effect whereas in the protective group hepatocytes were less accentuated expressed as less frequent necrotic area, and still large mitochondria were present with numerous undissolved lipid droplets and a small amount of rough endoplasmic reticulum. For the LS only group, nearly normal cells with centric nucleus, normal small mitochondria were at a normal nucleus-cytoplasmic ratio, but a few lipid droplets were noticed. Ultrastructural and Ultrahistochemical Studies on Rat Hepatocytes under the Effect of Lead Acetate Administration

The present study was carried out to investigate the effect of lead acetate on the ultrastructure of albino rat hepatocytes with special reference to its effect on the mitochondrial and lysosomal activity. Lead acetate was given orally to albino rats at a dose of 1% lead acetate / 100 g body weight, three times / week for one month （Gila）, two months （Glib）, three months （GIIc）. Liver total protein, body / liver weight ratio and cytochrome P-450 value were calculated. Three parts of the liver samples were incubated in media containing adenosine triphosphate, p-nitrophenyl phosphate and 2% glutlraldehyde and prepared for electron microscopy to visualize adenosine triphosphatase, acid phosphatase and the fine structures ofhepatocytes respectively. The main changes of the fine structures was found in the nucleus, as irregularity of the nuclear membrane, clumped heterochromatin and sun radiation of lead inclusion bodies. The cytoplasm was characterized by shortened, dilated rough endoplasmic reticulum, destroyed and hazy mitochondria with increased number of lysosomes with storage secretion. Ultrastructure findings showed hepatocytes damage due to increased hydrolysis enzymes and decreased cytotoxic enzymes （cytochrome P-450） as well as oxidative enzymes that proved the toxic effect of lead according to the duration of exposure.

MED-BS Clustering Algorithm for the Small-Scale Wireless Sensor Networks

With the spectacular progress of technology, we have witnessed the appearance of wireless sensor networks (WSNs) in several fields. In a hospital for example, each patient will be provided with one or more wireless sensors that gather his physiological data and send them towards a base station to treat them on behalf of the clinicians. The WSNs can be integrated on a building surface to supervise the state of the structure at the time of a destroying event such as an earthquake or an explosion. In this paper, we presented a Mobility-Energy-Degree-Distance to the Base Station (MED-BS) Clustering Algorithm for the small-scale wireless Sensor Networks. A node with lower mobility, higher residual energy, higher degree and closer to the base station is more likely elected as a clusterhead. The members of each cluster communicate directly with their ClusterHeads (CHs) and each ClusterHead aggregates the received messages and transmits them directly to the base station. The principal goal of our algorithm is to reduce the energy consumption and to balance the energy load among all nodes. In order to ensure the reliability of MED-BS, we compared it with the LEACH (Low Energy Adaptive Clustering Hierarchy) clustering algorithm. Simulation results prove that MED-BS improves the energy consumption efficiency and constructs a stable structure which can support new sensors without returning to the clusters reconstruction phase.

Desmoid Tumor of the Breast as a Manifestation of Gardner’s Syndrome in an Elderly Woman: A Case Report and Review of Literature

Desmoid tumor of the breast is an extremely rare entity. Histologically, it corresponds to a benign fibroblastic proliferation of a local evolution, with a high recurrence potential. We report the first case of elderly woman presented with desmoid-tumor of the breast as a manifestation of Gardner’s syndrome. A brief literature review was provided. We report the case of a 72-year-old woman who presented with five months history of painful and an exophytic mass of her left breast. She is known to have familial polyposis and had a total colectomy with Gardner’s syndrome. She had a history of osteomas of the maxilla. On clinical examination, there was an exophytic painless mass, on the upper medial quadrant of the breast. She had also lipoma in the left leg and pigmented skin lesions in legs. Her mammograms showed a suspicious stellar image. A computed tomography scan showed an ovoid lesion. Excision of the lesion was performed. Histopathology confirms a desmoid-tumor of the breast. Evolution was marked by lesion recurrence. In conclusion, desmoid tumor of the breast is rare, non-metastatic but locally aggressive. Clinical expression is often nonspecific. Treatment remains controversial;surgical excision is the treatment of choice.

Nurses: Workers Particularly Concerned by Contact Dermatitis in Public Hospital

Background: Contact dermatitis is common disease and represents a significant problem in healthcare sector, mainly among nurses. Many studies reported the prevalence of contact dermatitis from different parts of the world. Nevertheless, data about its frequency in Tunisia especially in public hospital seems to be insufficient. This study aims to assess the prevalence of contact dermatitis among nurses working in public hospital and identify risk factors. Patient and Methods: A cross sectional study was conducted among a representative sample of 1278 health professionals working in a public hospital matched by professional status and department. The survey was based on a questionnaire, a specialized examination and patch tests. Results: The prevalence of occupational contact dermatitis was 22% (17.5 - 27.2). The worker profile at risk of contact dermatitis was a female nurse aged 37 years working in a surgical department during 12.7 ± 9.36 years. Hands were damaged in 92.4% of cases and rhythmicity with occupational exposure was reported by 86% of affected individuals. Patch tests using European Standard Battery were performed among 33 workers and revealed a sensitization to an allergen among 26 workers. Patch test using rubber battery (if suspicion of allergic contact dermatitis to gloves) was performed among 29 agents and positive among 12. The analytical study revealed that history of atopy, job tenure, the mean daily number of hand washing, the mean daily number of worn gloves and mean duration of glove wearing were significantly higher in the affected population. Conclusion: Contact dermatitis affects particularly nurses in public hospitals. Its prevention requires a diagnostic approach based on a detailed professional investigation and patch tests.
