A versatile approach for temporary storage and shipping of in vitro cultured cells,cell sheets and tissue engineered constructs-a preliminary report

temporary storage/shipping of cell/tissue engineering products from bench to bedside is a key aspect of re-generative *** current proof-of-concept study presents a multipurpose device for temporary storage/shipping of cell culture dishes containing cell/tissue *** device,made with readily available raw ma-terials,contains three elements viz.a specialized lid,polymeric plates having grooves and a set of nuts and *** part of the performance evaluation,the device was first subjected to a simulated storage/shipping process,wherein the leak-proof and aseptic containment of the contents was ***,the setup was used for temporary storage/shipping of dishes having(a)L929 cell monolayers cultured on treated surfaces,(b)SIrC,HaCat and A549 cell sheets cultured on thermo-responsive surfaces,(c)HOS-cell encapsulated agar gels and(d)HOS-cell seeded silk fibroin *** results showed that the health of cell monolayers/cell sheets/tissue constructs after the process was comparable to that before the *** device was scalable,simple to handle,can be made for a single or multi-use purpose,and can be resizable to load other culture *** design of the storage/shipping device described in this report thus offers versatile features and applications.

A study of in vitro antibacterial activity of lanthanides complexes with a tetradentate Schiff base ligand

Objective:to establish the antibacterial activity of lanthanides complexes with a tetradentate Schiff base ligand ***:(N,N'-bis(l-naphthaldimine)-o-phenylenediamine)was prepared from the condensation of 2—hydroxy-1-naphthaldehyde with o-phenylenediamine in a molar ratio of 2:*** antimicrobial activity of the resultant Ln(Ⅲ)complexes was investigated using agar well diffusion and micro-broth dilution techniques;the latter was used to establish the minimum inhibitory concentrations for each compound ***:Most of Ln(Ⅲ)complexes were found to exhibit antibacterial activities against a number of pathogenic bacteria with MICs ranging between 1.95-250.00μg/*** aureus was the most susceptible bacterial species to[Lal(NO_3)_2(H_2O)](NO_3)complex while Shigella dysenteriae and Escherichia coli required a relatively higher MIC(250μg/mL).trie complexes La(Ⅲ)and pr(Ⅲ)were effective inhibitors against Staphylococcus aureus,whereas Sm(Ⅲ)complex was effective against Serratia *** the other hand,Gd(Ⅲ),La(Ⅲ)and Nd(Ⅲ)were found to be more potent inhibitors against Pseudompnas aeruginosa than two of commonly used *** remaining Ln(Ⅲ)complexes showed no remarkable activity as compared to the two standard drugs ***:tetradentate Schiff base ligand L and its complexes could be a potential antibacterial compounds after further investigation.

Biocompatibility of folate-modified chitosan nanoparticles

Objective:to evaluate the acute toxicity of carboxymethyl chitosan-2,2' ethylenedioxy bisethylamine-folate(CMC-EDBE-FA) and as well as possible effect on microbial growth and in vitro cell ***:CMC-EDBE-FA was prepared on basis of carboxymethyl chitosan tagged with folic acid by covalently linkage through 2,2' elhylenedioxy bis-ethylamine. In vivo acute toxicity,in vitro cyto-toxicity and antimicrobial activity of CMC-EDBE-FA nanoparticle were ***:Vancomycin exhibited the antibacterial activity against vancomycin sensitive Staphylococcus aureus,but CMC-EDBE-FA nanoparticle did not give any antibacterial activity as evidenced by minimal inhibitory concentration(MIC),minimal bactericidal concentration(MBC),disc agar diffusion(DAD) and killing kinetic ***,the CMC-EDBE-FA nanoparticle showed no signs of in vivo acute toxicity up to a dose level of 1000 mg/kg p.o.,and as well as in vitro cyto-toxicity up to 230 μg/***:these findings suggest that CMC-EDBE-FA nanoparticle is expected to be safe for biomedical applications.

to produce peptide antibiotics in large scale is a worthy yet challenging *** this study, the coding sequence of a human peptide antibiotic hPAB-β(derived from human beta defensin 2) was optimized according to the codon usage bias of yeast and the 6×his tagged and untagged hPAB-βgene were cloned into the pPIC9K vector *** multiple target gene integrated *** transformants were screened by G418 selection,and the 6×his tagged hPAB-βsecreting into the liquid culture medium had lower bactericidal activity than that of the untagged one determined by agar diffusion *** that the 6×his tag may affect the biological activity of this small peptide in our experimental *** untagged hPAB-βwas secreted into the culture supernatant at 241.2±29.5 mg/L after 60 hours of methanol induction in a 3.7 L *** hPAB-βwas purified to 95%homogeneity by reverse phase chromatography(Source 30 rPC) and gel filtration(Bio-gel P6) and the final yields of the purified hPAB-βreached 74.9±1.5 mg/*** authority of recombinant hPAB-βwas characterized by western blot using antibodies specifically against the synthetic hPAB-β.the minimum inhibitory concentrations(MICs) of the purified recombinant hPAB-βagainst type or clinical strains of microorganisms were about 31-250μg/ml, and these results showed that the methylotrophic yeast-inducible system is suitable for large-scale production of active antibacterial hPAB-βand the recombinant hPAB-βcould retain its bioactivity similar to that of the synthetic hPAB-β.

目的　探讨用中西药在体外消除肠道杆菌携带的耐药质粒 prSt98的可能性。方法　将原存在于伤寒杆菌中的耐药质粒 prSt98经接合转移分别导入大肠杆菌和鼠伤寒杆菌 ,选用双黄连和氧氟沙星对上述 3种细菌进行亚抑菌浓度的消除试验 ,并对实验前后的受试菌分别进行K -B纸片扩散法和最小抑菌浓度法 (MIC)药敏试验及质粒电泳检测。结果　两种药物在体外未能将受试菌携带的 prSt98消除 ;但该质粒编码的耐药标志减少 ;部分菌株对原有药物的耐药程度明显下降。结论　双黄连和氧氟沙星能降低prSt98宿主菌的耐药性 ,但药物作用后同一质粒在不同宿主菌中耐药性变化的差异 ,显示该质粒表达的多样性和复杂性。
