Stem cell secretome restore the adipo-osteo differentiation imbalance in diabetic dental pulp-derived mesenchymal stem cells

Background:Mesenchymal stem cells(MSCs)from type 2 diabetes mellitus(T2DM)individuals exhibit increased adipogenesis and decreased *** investigated the potential of adipose tissue-derived MSCs(ADMSCs)secretome obtained from healthy individuals in restoring the tumor necrosis factor-α(TNF-α)mediated imbalance in the adipo/osteogenic differentiation in the dental pulp-derived MSCs obtained from T2DM individuals(dDPMSCs).Methods:dDPMSCs were differentiated into adipocytes and osteocytes using a standard cocktail in the presence of(a)induction cocktail,(b)induction cocktail+TNF-α,and(c)induction cocktail+TNF-α+ADMSCs-secretome(50%)for 15 and 21 days *** adipocytes and osteocytes were stained by oil red O and alizarin red and analyzed by using ImageJ *** expression of the key genes involved was analyzed by using reverse-transcription polymerase chain reaction(RT-PCR).Results:Treatment of TNF-αaugmented the adipogenesis(9571±765 vs.19,815±1585 pixel,p<0.01)and decreased the osteogenesis(15,603±1248 vs.11,894±951 pixel,p<0.05)of dDPMSCs as evidenced by the oil red O and alizarin red staining ***,dDPMSCs differentiated along with TNF-αand 50%ADMSCs secretome exhibited enhanced osteogenesis(11,894±951 vs.41,808±3344 pixel,p<0.01)and decreased adipogenesis(19,815±1585 vs.4480±358 pixel,p<0.01).Additionally,dDPMSCs differentiated along with ADMSCs secretome exhibited decreased expression of PPARg(p<0.01),C/EBPa(p<0.05),and FAS(p<0.01)whereas mRNA expression of Runx2(p<0.05),Osterix(p<0.01),and OCN(p<0.05)was upregulated as revealed by the RT-PCR ***:ADMSCs secretome from healthy individuals restore the TNF-αinfluenced differentiation fate of dDPMSCs and therefore can be explored for T2DM clinical management in the future.

Background: Pharmacological factors used to induce insulin resistance (IR) inin vitro models may not mimic the fullin vivo features of type 2 diabetes mellitus (T2DM). This study aimed to examine the ability of diabetic serum (DS) to induce IR and investigate whether adipose-derived mesenchymal stem cell conditioned medium (ADMSC-CM) reverses DS-induced ***: DS was obtained from newly diagnosed T2DM patients. IR was induced in differentiated 3T3-L1 cells by employing dexamethasone, tumor necrosis factor alpha (TNF-α), palmitate and DS. Glucose uptake (2-[N-[7-nitrobenz-2-oxa-1,3-diazol-4-yl] amino]-2-deoxyglucose(2-NBDG) uptake assay), intracellular levels of reactive oxygen species (ROS), and superoxide radicals (O2-) (fluorescence microscopy and fluorometry) were analyzed in control and experimental samples. mRNA expression of key genes involved in glucose transport and inflammation were analyzed by using reverse transcription polymerase chain reaction (RT-PCR). Pro-inflammatory cytokines and phospho-insulin receptor substrate (IRS) (Ser-307) protein expression were analyzed by fluorescence activated cell sorter analysis. Statistical significance was determined by using one-way ANOVA followed by Tukey’s multiple comparison ***: ADMSC-CM significantly increased the DS-mediated decrease in 2-NBDG uptake (11.01 ± 0.50vs. 7.20 ± 0.30,P < 0.01) and reduced DS-driven ROS (fluorescence count, 6.35 ± 0.46vs. 9.80 ± 0.10,P < 0.01) and O2- (fluorescence count, 3.00 ± 0.10vs. 4.60 ± 0.09,P < 0.01) production. Further, the ADMSC-CM restored DS-induced down regulation GLUT4 (1.52- fold,P < 0.05) as well as the up-regulation of PPARγ (0.35-fold,P < 0.01), and IKKβ (0.37-fold,P < 0.01) mRNA, and phospho-IRS (Ser-307) protein expression compared to the baseline (median fluorescence intensity, 88,192 ± 2720vs. 65,450 ± 3111,P < 0.01). Alkali activated slag-fly ash concrete incorporating precious slag as fine aggregate for rigid pavements

This study aims to develop the alkali activated slag-fly ash concrete(AASFC) using precious slag(PS) ball as the fine aggregate. The slag-fly ash proportion, sodium silicate/sodium hydroxide(SS/SH) ratio and sodium hydroxide concentration are the prime variables. The fresh concrete properties like workability, setting time and the hardened concrete properties of AASFC developed under ambient curing were analyzed and similar way by replacing with fly ash(FA). The sodium hydroxide concentration of 8 M, 10 M and 12 M, SS/SH ratio of 1.0, 1.5 and 2.0, alkaline liquid to binder ratio of 0.35 and variation in slag-fly ash proportion as major inputs for mix design. The test results showed that, the fresh state of AASFC blend has a shorter setting time of 12-16 min, because of calcium mineral from the FA, the workability and setting time of AASFC mix was enhanced by the addition of the trisodium phosphate as a retarder. At the higher SS/SH extent, the workability and setting time exaggerated, whereas the increased FA content and sodium hydroxide concentration had a greater influence on the fresh state of concrete. Also, the sodium hydroxide concentration, SS/SH ratio and slag-fly ash proportion are the most inducing parameters on the workability, setting time and strength parameters of AASFC.

Quantification by allometric equations of carbon sequestered by Tectona grandis in different agroforestry systems

Non destructive methods for quantification of carbon seques- tration in tropical trees are inadequately developed. We described a stan- dardized method for estimating carbon stock in teak （Tectona grandis Linn. F.）. We developed linear allometric equations using girth at breast height （GBH）, height and age to quantify above ground biomass （AGB）. We used AGB to estimate carbon stock for teak trees of different age groups （1.5, 3.5, 7.5, 13.5, 18.5 and 23.5 years）. The regression equation with GBH, y = 3.174x - 21.27, r2=0.898 （p 〈0.01）, was found precise and convenient due to the difficulty in determination of height and age in dense natural forests of teak. Characterization of saline soil for the halophytes of largest inland saline wetland of India using geospatial technology

Pattern of spatial variability in granulometric and mineralogical characteristics across an ecologically sensitive sub-tropical wetland(Bihar, India)

Floodplain wetlands are dynamic systems undergoing frequent hydro-sedimentological changes. Tropical/subtropical wetlands have even more pronounced climatic and biological influences on the sediment dynamics. In the current study, a protected subtropical wetland in Bihar(India) has been assessed for its sedimentary and mineralogical status. Sediment characteristics and associated granulometric parameters are discussed to highlight the wetland’s hydrogeomorphic character, energy dimension, and the depositional processes taking place. Statistical grain size parameters like graphic mean, graphic skewness,graphic kurtosis, and their interrelations studies have helped in understanding the sources of deposited materials. Multivariate statistics have identified, sand fractions, measures of central tendencies(mean,median, and mode), percent organic matter(%OM) and percent carbonates(%carbonates) as primary factors explaining the maximum variance in the data. The non-parametric Kruskal-Wallis test has identified %sand, %clay, electrical conductivity(EC), and %carbonates as the major factors responsible for significant spatial variability. The grain texture analysis shows that the majority of samples fall in the brackets of clayey silt; silty sand; and sand, silt, and clay. Common mineral components associated with these are quartz, feldspar, clay minerals, etc. The range for the mean grain size lies between 3Φ to 9Φ(Φ is diameter of the sediments) where most of the grain size distributions are symmetrically to fine symmetrically skewed, indicating intermediate energy to adjust to environmental conditions. The granulometric and physicochemical characteristics of the sediment samples also indicate the directional distribution pattern of sediment along the hydrological pathway. Future studies concentrating on the fluvial discharges of water and sediment will be highly insightful in reconstructing the past net sediment accumulation processes in the Baraila Wetland.

Chemical pre-treatments improve seed germination and seedling growth in Semecarpus kurzii:an ethnomedicinally important plant

Semecarpus kurzii *** an important ethnomedicinal plant used for curing tumours,inflammation,fever and pain,and is also a source of iso ricinoleic *** the present investigation,effect of chemical pretreatments on germination and seedling growth revealed significant *** pre-treatments could improve germination and treatment with KNO_3 （9.9 mM）was significantly superior（94%） to the untreated control（76%）.Mean daily germination varied among the treatments（0.90–4.14 seeds per day）,when compared with 2.27 seeds per day in *** germination time was significantly reduced from 22.14 days（control） to 16.40 days（19.8 mM KNO_3） and 17.16 days（1.45 mM GA_3）.Treatment with KNO_3（19.8 mM） and GA_3（1.45 and 2.90 mM）significantly reduced the time for 50% *** with higher concentrations of GA_3 and KNO_3 exhibited significantly faster germination initiation（11.0 and 12.0 days,respectively） over the control（17.0 days）.Seedling vigour index was not improved by *** treatments did not improve shoot growth,collar thickness,or number of seedling leaves; however,root growth was significantly improved over *** with GA_3（2.90 mM） promoted leaf elongation but reduced leaf width *** with KNO_3 and GA_3 could be recommended to obtain healthy seedlings for establishing new plantations of this important medicinal plant species.

Basin Geomorphology and Drainage Morphometry Parameters Used as Indicators for Groundwater Prospect: Insight from Geographical Information System(GIS) Technique

Influence of structural and lithological controls of various drainage patterns and their stream orientations（for 2nd, 3rd and 4th order steams） were identified to evaluate the direction and controlling factors of drainage network. To investigate the prospect of groundwater, hydrogeomorphological features of river basin viz. Mulki-Pavanje were identified and mapped. To evaluate the characteristics of the basin, different morphometric parameters（linear, areal and relief） were computed in sub-basin wise（SB-I to-VII）. The linear parameters suggest drainage network is controlled by geomorphology. The form factor（Ff）, elongation ratio（Re） and circularity ratio（Rc） suggest that the basin is in an elongated shape. The drainage density（Dd） indicates resistant/permeable strata under medium-dense vegetation with moderate relief. The areal parameters of the sub-basins（except SBI and Ⅲ） indicates moderate ground-slopes associated with moderately permeable rocks, which promote moderate run-off and infiltration. Drainage texture（T） of the whole basin indicates coarse texture while the SB-I, and Ⅲ showing an intermediate texture. The relief parameters namely ruggedness number（Rn） infers low basin relief and poor drainage density. To identify the most deficit/surplus zones of groundwater suitable weightages were assigned to the hydrogeomorphological units and morphometric parameters. The study reveal that the basin manifest that SB-Ⅲ shall be most deficit zone of groundwater, whereas SB-VII, VI and V are found to show increase in groundwater potentiality. Groundwater prospect area in this basin is estimated to be 7% area under poor, 44% area under moderate and 49% area under good to excellent. This paper demonstrated the potential application of geographical information system（GIS） techniques to evaluate the groundwater prospect in absence of traditional groundwater monitoring data.
