The impact of perioperative blood transfusions on short-term outcomes following hepatectomy

Effects of different doses of cadmium on secondary metabolites and gene expression in Artemisia annua L.

This study aims to elucidate the underlying molecular mechanisms of artemisinin accumulation induced by cadmium （Cd）. The effects of different Cd concentrations （0, 20, 60, and 120 μmol/L） on the biosynthesis ofArtemisia annua L. were examined. Intermediate and end products were quantified by HPLC-ESI- MS/MS analysis. The expression of key biosynthesis enzymes was also determined by qRT-PCR. The results showed that the application of treatment with 60 and 120 μmol/L Cd for 3 days significantly improved the biosynthesis of artemisinic acid, arteannuin b, and artemisinin. The concentrations of artemisinic acid, arteannuin b, and artemisinin in the 120 llmol/L Cd-treated group were 2.26, 102.08, and 33.63 times higher than those in the control group, respectively. The concentrations of arteannuin b and artemisinin in 60 μmol/L Cd-treated leaves were 61.10 and 26.40 times higher than those in the control group, respectively. The relative expression levels of HMGR, FPS, ADS, CYP71AV1, DbR2, ALDH1, and DXR were up-regulated in the 120 μmol/L Cd-treated group because of increased contents of artemisinic metabolites after 3 days of treatment. Hence, appropriate doses of Cd can increase the concentrations of artemisinic metabolites at a certain time point by up-regulating the relative expression levels of key enzyme genes involved in artemisinin biosynthesis.

Prokaryotic type II adaptive immune systems have been developed into the versatile CRISPR technology, which has been widely applied in site- specific genome editing and has revolutionized biomedical research due to its superior efficiency and flexibility. Recent studies have greatly diversified CRISPR technologies by coupling it with various DNA repair mechanisms and targeting strategies. These new advances have significantly expanded the generation of genetically modified animal models, either by including species in which targeted genetic modification could not be achieved previously, or through introducing complex genetic modifications that take multiple steps and cost years to achieve using traditional methods. Herein, we review the recent developments and applications of CRISPR-based technology in generating various animal models, and discuss the everlasting impact of this new progress on biomedical research.

STING regulates bCR signaling in normal and malignant b cells

STING is an endoplasmic reticulum(ER)-resident protein critical for sensing cytoplasmic DNA and promoting the production of type Ⅰ interferons;however,the role of STING in b cell receptor(bCR)signaling remains *** generated STING V154M knock-in mice and showed that b cells carrying constitutively activated STING specifically degraded membrane-bound IgM,Ig a and Igβ via SEL1L/HRD1-mediated ER-associated degradation(ERAD).b cells with activated STING were thus less capable of responding to bCR activation by phosphorylating Igα and Syk than those without activated *** immunized with T-independent antigens,STING V154M mice produced significantly fewer antigen-specific plasma cells and antibodies than immunized wild-type(WT)*** further generated b cell-specific STING^(KO) mice and showed that STING^(KO) b cells indeed responded to activation by transducing stronger bCR signals than their STING-proficient *** b cell-specific STING^(KO) mice were T-independently immunized,they produced significantly more antigen-specific plasma cells and antibodies than immunized STIN^(WT)*** both human and m ouse IGHV-unmutated malignant chronic lymphocytic leukemia(CLL)cells downregulated the expression of STING,we explored whether STING downregulation could contribute to the well-established robust bCR signaling phenotype in malignant CLL *** generated a STING-deficient CLL mouse model and showed that STING-deficient CLL cells were indeed more responsive to bCR activation than their STING-proficient *** results revealed a novel b cell-intrinsic role of STING in negatively regulating bCR signaling in both normal and malignant b cells.

将牛体外受精后发育到7天和8天的胚胎进行核型鉴定,结果表明胚胎的性比例与胚胎的发育速度密切相关。发育7天体外受精胚胎进行核型鉴定的结果如下:处于扩展囊胚阶段的胚胎,雄性比例为100％;处于扩展中囊胚、囊胚和早囊阶段的胚胎,雄性比例分别为89％、75％和40％。将发育8天的囊胚进行核型鉴定,雄性比例下降为20％。体外受精胚胎平均有丝分裂指数为3.5％。这些结果提示:利用胚胎发育速度与性别的关系,可以对体外受精胚胎进行无损伤的性别鉴定。

background:Few studies have assessed the relationship between multimorbidity patterns and mortality risk in the Chinese *** aimed to identify multimorbidity patterns and examined the associations of multimorbidity patterns and the number of chronic diseases with the risk of mortality among Chinese middle-aged and older ***:We used data from the China Kadoorie biobank and included 512,723 participants aged 30 to 79 *** was defined as the presence of two or more of the 15 chronic diseases collected by self-report or physical examination at *** patterns were identified using hierarchical cluster *** regression was used to estimate the associations of multimorbidity patterns and the number of chronic diseases with all-cause and cause-specific ***:Overall,15.8%of participants had *** prevalence of multimorbidity increased with age and was higher in urban than rural *** multimorbidity patterns were identified,including cardiometabolic multimorbidity(diabetes,coronary heart disease,stroke,and hypertension),respiratory multimorbidity(tuberculosis,asthma,and chronic obstructive pulmonary disease),gastrointestinal and hepatorenal multimorbidity(gallstone disease,chronic kidney disease,cirrhosis,peptic ulcer,and cancer),and mental and arthritis multimorbidity(neurasthenia,psychiatric disorder,and rheumatoid arthritis).During a median of 10.8 years of follow-up,49,371 deaths *** with participants without multimorbidity,cardiometabolic multimorbidity(hazard ratios[HR]=2.20,95%confidence intervals[CI]:2.14-2.26)and respiratory multimorbidity(HR=2.13,95%CI:1.97-2.31)demonstrated relatively higher risks of mortality,followed by gastrointestinal and hepatorenal multimorbidity(HR=1.33,95%CI:1.22-1.46).The mortality risk increased by 36%(HR=1.36,95%CI:1.35-1.37)with every additional ***:Cardiometabolic multimorbidity and respiratory multimorbidity

本文评价了用合成的聚氨酯泡沫块代替传统的含泥炭基质作桃带节插条、半硬枝插条和硬枝插条的生根基质。8月份采的半硬枝插条在泡沫块中生根率高于两种含泥炭的基质。三种扦插基质间初生根数量和根长度均无差异。18个品种(桃和油桃)的半硬枚插条在泥凝—珍珠岩—蛭石基质中平均生根率为58%,而在泡沫块中则为77%。10月份采集的硬枝插条在泡沫块中或泥炭基质中其生根率或根级均无差异。尽管带节插条生根率低子半硬枝插条和硬枝插条,但在泡沫块和泥炭基质间并无差异。

背景用药过失是导致患者并发症和死亡的重要原因，与患者安全不足相关。然而，目前尚缺乏全科诊所处方的安全评估工具。目的为了识别并更新一套用以评估全科诊所处方安全的指示因子，且对患者有害风险与每个指示因子的相关性进行分析。设计和场所采用兰德公司-美国加利福尼亚大学洛杉矶分校（RAND/ UCLA）专家共识法探索英国全科医学处方的安全指示因子。方法通过系统性回顾及既往共识识别处方指示因子。采用 RAND 法做进一步识别，采用电子德尔菲法评价相关风险。对英国12家全科诊所运用了 RAND 法，11家诊所采用电子德尔菲法进行了风险评估。结果发现有56个处方安全指示因子（专家小组评定中位分数达7~9为共识）。这些指示因子覆盖处方风险含：治疗适应证、药物-药物联合使用的风险及化验室监测不足。＆gt;80%的参试诊所存在这些因子中的23个（41%）高风险或极度风险。结论全科医学专家小组识别出56个指示因子可用于评估全科医学处方安全。其中有23个指示因子与患者的高风险和极度风险关联，应关注研究成果以维护患者的安全。
