The CRISPR/Cas9 system has been tailored to a revolutionary genetic tool because of its remarkable simplicity and *** complex genome editing in the mouse since the 1990 s has been dominated by the use of embryonic stem（ES） cells,CRISPR/Cas9 now offers a versatile and fast approach to precisely modify virtually any DNA regions directly in mouse ***,this relative simplicity does not preclude a conscientious preparatory work that is often neglected when initiating a ***,we describe the key steps leading to successful generation of a double knockout（KO） mouse by simultaneously targeting two homolog genes,Tmem176 a and Tmem176 b,which are located in the same genomic ***,we show that similar efficiency can be obtained in a mixed genetic background or directly in the C57BL/6 inbred ***,presented as a detailed case study that should be helpful to the non-specialists,we focus on the genotyping strategy to anticipate the various possibilities.

Progressive familial intrahepatic cholestasis type 3 (PFIC3) is a rare cholestatic liver disease. Such liver disease can get worse by female hormone disorder. Albumin dialysis or Molecular Adsorbent Recirculating System (MARS) has been reported to reverse severe cholestasis-linked pruritus. Here, we report the first use of MARS during a spontaneous pregnancy and its successful outcome in a patient with PFIC3 and intractable pruritus. Albumin dialysis could be considered as a pregnancy-saving procedure in pregnant women with severe cholestasis and refractory pruritus.

In the clinical context of the patients with liver cirrhosis, accurate evaluation of the renal function is potentially crucial. Indeed, it can lead to early diagnosis of both acute kidney injury and chronic kidney disease and to reliable characterization of the renal status of the patient before performing a liver transplantation. Despite some limitations, the assay of serum creatinine (SCr) is universally used to estimate glomerular filtration rate (GFR) because of its wide availability, its simplicity and because it is inexpensive. Nevertheless, several reports show that the value of this assay to estimate GFR is strongly challenged in cirrhotic patients, especially in patients with liver failure and/or severely impaired renal function. This has led to seek new alternatives to estimate more reliably the GFR in these patients. Although the reference methods, based on the utilization of exogenous markers, allow measuring GFR and thereby constitute the “gold standard” to evaluate renal function, they are not feasible in routine clinical practice. Several studies have shown that a cystatin C (CysC) based formula perform better than the SCr-based estimates in cirrhotic patients and the estimation of GFR by these formulas could therefore lead to optimize the management of the patients. A new estimate based on CysC has been recently developed using a large number of patients and the first results regarding the evaluation of its performance are promising, making this new formula the best candidate for a reference estimate of the renal function in cirrhotic patients.

The aim of this paper is to give an overview on models and methods used to solve tactical planning problems. The modeling and the elaboration of the well-know tactical planning problems （master planning ＆ scheduling, material requirement planning and multi-site planning） are discussed. These problems are modeled from two ＂lot sizing＂ models called the Capacitated Lot Sizing Problem （CLSP） and Multi Level Capacitated Lot Sizing Problem （MLCLSP）. From both models, a lot of extensions has been proposed in the literature. The purpose of this paper is twofold： first, classifications of the CLSP and MLCLSP as well as their extensions are given. For each model, the major scientific contributions are mentioned. These classifications made from seventy papers give an overview of ＂lot sizing＂ models dedicated to the MPS, MRP and Multi-site and show the diversity of models. Second, from a classification, an analysis of methods used for each model is given. The instance size, best gap and reference for gap computation are given for each contribution, This work can be used to elaborate an optimization tool for tactical planning problematic such as Advanced Planning System.

Since 1979, China has recorded a remarkable trade performance, which has been driven by international processing and the offshoring strategies of foreign firms. The diversification of Chinese exports and their technological upgrading have been phenomenal However, there is also inertia, illustrated by the persistent dualism of the trade sector, the unrelenting specialization in downmarket products and the deteriorating terms of trade. These weaknesses have helped its partners to adjust to the rise of this new trade power. In the past decade, China＇s economy has faced the adverse effects o fan export-led growth and the global crisis has revealed its vulnerability. China is now forced to rebalance its economy. This will imply major changes in foreign trade, in favor of ordinary trade and away from processing. In the foreseeable future, China is unlikely to become the driver of international demand but will remain the engine of Asian economic integration.

铝是一种在地球上广泛存在的金属材料。由于铝具有非常好的机械性能、导电性以及导热性,并且其密度较低,因而广泛应用于交通运输以及建筑行业。轻质,耐腐蚀,易成型的铝合金材料非常适用于食物包装。世界上,每年要生产出2100亿个饮料罐,其中85%是用铝合金制造的。北美,欧洲和日本在以重熔为主要手段的铝合金回收行业处于领先地位,然而中国,印度和俄罗斯等国回收铝合金比例也在快速增长。事实上,回收一个铝合金易拉罐所消耗的能量相比于生产出同样的数量的易拉罐少20倍,其温室气体的排放量也减少近20倍。此外,铝合金易拉罐是100%可回收材料,并且在回收加工的过程中不会丢失其本身的优异性能。本文是以回收的铝合金易拉罐为原料,研究其在经过高能球磨和热固结后材料的力学性能。这种方法现在在工业中并不常用,但它有可能取代重熔回收。因此,本项目着眼于研究通过固态回收,制备纳米结构铝合金粉末以及铝合金纳米复合材料,并研究粉末固结后材料的力学性能与结构的关系。通过不同的高能球磨参数,制备出铝合金粉末和添加纳米Si C增强颗粒的纳米复合材料粉末。我们总共制备了4种不同的粉末,包括两种不同的组分和两种不同的球磨时间。我们通过粉末的维氏硬度,ICP测试,XRD分析,粒度分析以及SEM分析等方式表征出粉末的力学性能,成分以及结构。提高球磨时间和添加5%纳米Si C颗粒会提高粉末的维氏硬度。通过ICP分析原始易拉罐和球磨后的粉末成分,再对比制成后材料的理论成分,证实了通过混合和球磨两种不同成分的铝合金易拉罐,成功制备出一种新成分铝合金。粉末通过两种不同的方式进行固结和挤压。通过这两种方式制备的材料,其抗拉强度到达了600～750MPa之间。相比于冷压直接挤压的样品,经过SPS的样品尽管材料塑性变形阶段没有增大,但是其弹性变形阶段的延伸率增加了,其总的延伸率数值从1.5%增加到3%。额外添加5%Si C的样品虽然没有增加材料的硬度,但是其增加了材料最终的抗拉强度。此外,铝合金基体内观察到了铁的夹杂,它可能会影响材料的抗拉强度,并对材料的最终断裂产生重大影响。本研究是以回收铝合金易拉罐为原料,研究经过机械合金化制成的粉末,以及通过热固结和挤压后制成材料的机械性能和显微结构。

Since the global financial crisis of 2007/2008, China＇s foreign trade has continued to grow faster than international trade, but its drivers are now different from those prevailing before. The participation of the Chinese economy in the global production chains through processing activities is no longer the main driver of its trade performance. The new driving force of change is ordinary trade, based mainly on local inputs and domestic demand. China, which played a major role in the globalization process as an export base for multinational companies, is now shifting to having a ＂＇normal＇foreign trade system, which is more closely integrated into the domestic economy.

China and India are two demographic giants that have become big developing economic powers. They have maintained their specialization in textiles and developed outward-oriented sectors linked to new technologies, taking advantage of offshoring and outsourcing. Their increasing contribution to international trade is changing the world supply and demand of manufactured goods, primary goods and services. They are new leaders in the international division of labor, but beyond technological catch-up, their challenge is quality upgrading. Both countries are increasingly contributing to global economic growth, but they cannot yet trigger the growth of the rest of the world by themselves.

Aims Both extreme drought and insect herbivores can suppress plant growth in grassland ***,most studies have examined extreme drought and insects in isolation,and there is reason to believe that insects might alter the ability of grasslands to withstand ***,few studies have tested the interactive effects of extreme drought and insect herbivores in grassland *** Here,we tested the drought–herbivore interactions using a manipulative experiment that factorially crossed extreme drought with the exclusion of insect herbivores in a temperate semiarid grassland in Inner *** Findings Our results demonstrated that both extreme drought and insect herbivores separately decreased total plant *** combined,insect herbivores reduced the impact of drought on total cover by increasing the relative abundance of drought-resistant dominant *** results highlight that the negative effect of extreme drought on total plant cover could be alleviated by maintaining robust insect herbivore communities.
