Feasibility and safety of autologous bone marrow mononuclear cell transplantation in patients with advanced chronic liver disease

AIM： To evaluate the safety and feasibility of bone marrow cell （BMc） transplantation in patients with chronic liver disease on the waiting list for liver transplantation. METHODS： Ten patients （eight males） with chronic liver disease were enrolled to receive infusion of autologous bone marrow-derived cells. Seven patients were classified as child-Pugh B and three as child-Pugh c. Baseline assessment included complete clinical and laboratory evaluation and abdominal MRI. Approximately 50 mL of bone marrow aspirate was prepared by centrifugation in a ficoll-hypaque gradient. At least of 100 millions of mononuclear-enriched BMcs were infused into the hepatic artery using the routine technique for arterial chemoembolization for liver tumors. Patients were followed up for adverse events up to 4 mo. RESULTS： The median age of the patients was 52 years （range 24-70 years）. All patients were discharged 48 h after BMc infusion. Two patients complained ofmild pain at the bone marrow needle puncture site. No other complications or specific side effects related to the procedure were observed. Bilirubin levels were lower at 1 （2.19 ± 0.9） and 4 mo （2.10 ± 1.0） after cell transplantation that baseline levels （238 ± 1.2）. Albumin levels 4 mo after BMc infusion （3.73 ± 0.5） were higher than baseline levels （3.47 ± 0.5）. International normalized ratio （INR） decreased from 1.48 （SD = 0.23） to 1.43 （SD = 0.23） one month after cell transplantation. cONcLUSION： BMc infusion into hepatic artery of patients with advanced chronic liver disease is safe and feasible. In addition, a decrease in mean serum bilirubin and INR levels and an increase in albumin levels are observed. Our data warrant further studies in order to evaluate the effect of BMc transplantation in patients with advanced chronic liver disease.

With the expansion of eucalyptus crops to areas with severe water limitations,physiological studies involving eucalyptus clones to identify those that are tolerant to water stress become *** objective of this study was to assess morphological and physiological responses by eucalyptus clones subjected to drought stress and *** experiment consisted of three eucalyptus clones:Vc865,I224 and I144 and two water regimes:control and water stress followed by rehydration,with six *** water potential,gas exchange,maximum quantum efficiency of photo systemⅡand plant height and stem diameter were evaluated under drought stress and ***6 d of rehydration,the number of leaves,leaf area and dry mass of root,leaf,stem and their total were *** clones showed intense reduction of gas exchange during the drought stress period,and only Vc865 and 1144 showed rapid recovery with 3 d of *** 1224 showed greater reduction in height,stem diameter,number of leaves,water potential at midday(Ψ_(w)_(Midday)),and maximum quantum efficiency of photosystemⅡ(F_(v)/F_(m)).clones Vc865and T144 showed lower reductions inΨ_(wMidday)and F_(v)/F_(m) under ***865 had lower reductions in leaf number,leaf area and higher leaf dry mass,while clone I144 had higher height and lower reduction in root dry mass *** these clones showed higher water use efficiency with 3d of *** different phenotypic plasticities gave the clones Vc865 and 1144 efficient mechanisms of acclimatization to stress and more drought tolerance,enhancing their greater capacity for recovery after stress,which allowed lower dry mass *** 1224,however,was more susceptible to drought stress,undergoing greater physiological damage with only partial recovery during rehydration.

Invasive species are a major threat to island biodiversity,and their eradications have substantially contributed to the conservation of island ***,the consequences of eradications on the trophic ecology of native taxa are largely ***,we used the eradication of invasive black rats Rattus rattus and European rabbits Oryctolagus cuniculus from the Berlenga Island,in the western coast of Portugal,as a whole-ecosystem experiment to investigate the effects of the eradication of invasive mammals on the trophic niche and body dimensions of the island-restricted Berlenga wall lizard Podarcis carbonelli berlengensis over a 2-year *** results suggest an expansion of the isotopic niche and an intensification of the sexual dimorphism of the lizard following mammal ***,we found considerable variability in isotopic niche across the island and detected evidence of sex-specific and season-modulated nutritional requirements of this threatened *** findings support that the eradication of 2 of the planets most problematic invasive vertebrates led to changes in the lizard trophic niche and sexual dimorphism in just 2 *** suggests that the ecological pressuresfor example,prey availability and habitat structureto which lizards are exposed have substantially changed *** study emphasizes the scientific value of island eradications as experiments to address a wide range of ecological questions and adds to the increasing body of evidence supporting substantial conservation gains associated with these restoration interventions.

AIM To evaluate the sex-specific effects of a hydroalcoholic extract from Eugenia punicifolia(HEEP) leaves on gastric ulcer *** In this rat study involving males, intact(cycling) females, and ovariectomized females, gastric ulcers were induced using acetic acid. A vehicle, lansoprazole, or HEEP was administered for 14 d after ulcer induction. Body weight was monitored throughout the treatment period. At the end of treatment, the rats were euthanized and the following in vivo and in vitro investigations were performed: macroscopic examination of the lesion area and organ weights, biochemical analysis, zymography, and evaluation of protein expression levels. Additionally, the concentration-dependent effect of HEEP was evaluated in terms of subacute toxicity and *** compared to the vehicle, HEEP demonstrated a great healing capacity by substantially reducing the ulcerative lesion area in males(52.44%), intact females(85.22%), and ovariectomized females(65.47%), confirming that HEEP accelerates the healing of acetic acidinduced gastric lesions and suggesting that this effect is modulated by female sex hormones. The antiulcer effect of HEEP was mediated by prostaglandin E2 only in male rats. Overall, the beneficial effect of HEEP was the highest in intact females. Notably, HEEP promoted the expression of vascular endothelial growth factor(intact vs ovariectomized females) and decreased the expression of caspase-8 and Bcl-2(intact female vs male or ovariectomized female). Additionally, HEEP enhanced fibroblast proliferation and migration into a wounded area in vitro, confirming its healing effect. Finally, no sign of subacute toxicity or cytotoxicity of HEEP was *** In gastric ulcers, HEEP-induced healing(modulated by female sex hormones; in males, mediated by prostaglandin) involves extracellular matrix remodeling, with gastric mucosa cell proliferation and migration.

Hesperidin Reduces cisplatin-lnduced DNA Damage in Bone Marrow cells of Mice

Hesperidin is a bioflavonoid abundantly found in citrus fruits and displays chemoprotective effects against DNA （deoxyribonucleic acid） damage, however there are few reports about hesperidin effects against cisplatin-DNA damage induction. The aim of this work was to evaluate hesperidin antimutagenicity against cisptatin-DNA damage. （1） The antimutagenicity of hesperidin was assayed by bone marrow of mice in vivo using the micronucleus test. Hesperidin pre-treatment protocol reduced the frequency of MNPcE （micronucleated polychromatic erythrocytes） and the dose of 100 mg·kg-1 was highest efficiency, with 65.24% of damage reduction. In the simultaneous treatment protocol, the dose of 200 mg·kg-1 exhibited a more effective reduction of MNPcE, with 94.01% of damage reduction. （2） Hesperidin was also effective in reducing the MNPcE frequency in the post-treatment protocol for all doses, with 77.48%, 82.13% and 90.08% of damage reduction at the doses of 100, 200 and 400 mg·kg-1, respectively. From the study, it can be concluded that hesperidin was able to promote the reduction of micronuclei frequency and DNA damage induced by cisplatin. Hesperidin is a powerful antioxidant compound and its chemoprotective effects on DNA may occur due to its association with the antioxidant cell system which is responsible for eliminate free radicals generated by chemical harmful to DNA.

dengue in renal transplant recipients: clinical course and impact on renal function

AIM To present clinical characteristics from renal transplant recipients with dengue fever and its impact on graft *** We retrospectively evaluated 11 renal transplant recipients(RTR) with dengue infection confirmed by laboratory test, between January 2007 and July 2012, transplanted in the Renal Transplant center of Walter cantídio University Hospital from Federal University of ceará. RESULTS Positive dengue serology(IgM) was found in all patients. The mean time between transplant and dengue infection was 43 mo. Fever was presented in all patients. Nine patients presented with classical dengue and two(18%) with dengue hemorrhagic fever. All cases had satisfactory evolution with complete recovery of the symptoms. The time for symptom resolution varied from 2 to 20 d, with an average of 9 d. An increase of creatinine after the infection was observed in three(27.2%) patients with no clinically impact on the kidney graft function. cONcLUSION RTR with dengue infection seems to have a clinical presentation and evolution similar to those seen in the general population, with no long-term damage to patient and to the graft.
