Paradoxical expression pattern of the epithelial mesenchymal transition-related biomarkers CD44, SLUG, N-cadherin and VSIG1/Glycoprotein A34 in gastrointestinal stromal tumors

AIM To evaluate the immunohistochemical(IhC) expression of five biomarkers, commonly involved in epithelial mesenchymal/mesenchymal epithelial transition(EMT/MET), in gastrointestinal stromal tumors(GISTs). METhODS In 80 consecutive GISTs the IhC examinations were performed using the EMT-related antibodies E-cadherin,N-cadherin, SLUG, V-set and immunoglobulin domain containing 1(VSIG1) and CD44. RESULTS The positivity rate was 88.75% for SLUG, 83.75% for VSIG1, 36.25% for CD44 and 10% for N-cadherin. No correlation was noted between the examined markers and clinicopathological parameters. Nuclear positivity for SLUG and VSIG1 was observed in all cases with distant metastasis. The extra-gastrointestinal stromal tumors(e-GISTs) expressed nuclear positivity for VSIG1 and SLUG, with infrequent positivity for N-cadherin and CD44. The low overall survival was mainly dependent on VSIG1 negativity(P = 0.01) and nuclear positivity for SLUG and/or CD44. CONCLUSION GIST aggressivity may be induced by nuclear upregulation of SLUG and loss or cytoplasm-to-nuclear translocation of VSIG1. SLUG and VSIG1 may act as activated nuclear transcription factors. The CD44, but not N-cadherin, might also have an independent prognostic value in these tumors. The role of the EMT/MET-related transcription factors in the evolution of GISTs, should be revisited with a larger dataset. This is the first study exploring the IhC pattern of VSIG1 in GISTs.

Epithelial-to-mesenchymal transition(EMT) represents conversion of an epithelial cell in an elongated cell with mesenchymal phenotype, which can occur in physiologic and pathologic processes such as embryogenesis(type 1 EMT), wound healing and/or fibrosis(type 2 EMT) and malignant tumors(type 3 EMT). The proliferation rate, metastasizing and recurrence capacity, as also the individualized response at chemotherapics, in both epithelial and mesenchymal malignant tumors is known to be influenced by reversible switch between EMT and mesenchymal-to-epithelial transition(MET). Although much research work has already been done in these fields, the specific molecular pathways of EMT, relating to the tumor type and tumor localization, are yet to be elucidated. In this paper, based on the literature and personal experience of the authors, an update in the field of EMT vs MET in epithelial and mesenchymal tumors is presented. The authors tried to present the latest data about the particularities of these processes, and also of the so-called endothelialto-mesenchymal transition, based on tumor location. The EMT-angiogenesis link is discussed as a possible valuable parameter for clinical follow-up and targeted therapeutic oncologic management. The paper begins with presentation of the basic aspects of EMT, its classification and assessment possibilities, and concludes with prognostic and therapeutic perspectives. The particularities of EMT and MET in gastric and colorectal carcinomas, pancreatic cancer, hepatocellular and cholangiocarcinomas, and lung, breast and prostate cancers, respectively in sarcomas and gastrointestinal stromal tumors are presented in detail.

Red mud, the by-product of aluminum production, has been regarded as a problematic residue all over the world. Its storage involves risks as evidenced by the Ajka red mud spill,an accident in hungary where the slurry broke free, flooding the surrounding areas. As an immediate remediation measure more than 5 cm thick red mud layer was removed from the flooded soil surface. The removed red mud and soil mixture（RMSM） was transferred into the reservoirs for storage. In this paper the application of RMSM is evaluated in a field study aiming at re-utilizing waste, decreasing cost of waste disposal and providing a value-added product. The purpose was to investigate the applicability of RMSM as surface layer component of landfill cover systems. The field study was carried out in two steps： in lysimeters and in field plots. The RMSM was mixed at ratios ranging between 0 and 50% w/w with low quality subsoil（LQS） originally used as surface layer of an interim landfill cover. The characteristics of the LQS ＋ RMSM mixtures compared to the subsoil（LQS） and the RMSM were determined by physical–chemical, biological and ecotoxicological methods. The addition of RMSM to the subsoil（LQS） at up to 20% did not result any ecotoxic effect, but it increased the water holding capacity. In addition, the microbial substrate utilization became about triple of subsoil（LQS） after 10 months. According to our results the RMSM mixed into subsoil（LQS） at20% w/w dose may be applied as surface layer of landfill cover systems.

Chemical investigation of Beouverio felina,a marine-derived entomopathogenic fungus,obtained two new destruxin hexadepsipeptides(1 and 2)containing an/.-Ala^(5)residue instead of the usual A/-Me-L-Ala^(5).Interestingly,the presence of the/.-Ala^(5)residue enabled two slowly-exchanging solution conformers with cis/trons(ILE^(3))-(A/-Me-Val^(4))amide bond for 1 and 2 with 1.5:1 and 1:1 cis/trans ratios,respectively,while only the conformer with c/s amide bond was found in the *** solution and solid-state conformers were studied by NMR,X-ray diffraction and solution and solid-state ECD measurements,as well as by TDDFT-ECD ***(conformational)parameter-activity correlations suggested that the extended destruxin geometry with the preferred c/s(ILE^(3))-(A/-Me-Val^(4))bond probably resulted in improved bioactivity and reduced toxicity for the destruxin *** was the first report on the interconversion phenomenon of cis/trans amide bonds of destruxin class,which could give insights in the development of bioactive destruxin agents with lower toxicity.

In this paper, the author shall deal with the hungarian variants of dualistic world creation myths. The aim of this paper is to shed some light on the nature of the connection between the Central-East European hungarian and South-East European Romanian and Bulgarian myth variants. The hungarian pieces are the westernmost variants of the dualistic creation myth-group. Geographically, they stand close to the Romanian variants, but the serious motivational differences between the Romanian and hungarian variants show that the descent of the two oral narrative groups are distinct, Both the hungarian and the Romanian variants show similarities with the Bulgarian world creation myths, however, geographically they do not relate tightly. This is why a possible explanation of these correlations is, that the hungarian and the Bulgarian myths have common Central Eurasian, South-Eastern Siberian origins, upon which starting from the sixth to the seventh centuries the super strata of various gnostic influences have settled.

Numerous legends are known in Eurasia, in which one or more hunters are trying to get a stag, and then the hunters will find a new homeland, or establish a new cathedral at the location where the deer appeared earlier. According to the structure of the legend, the Eastern legend type is more ancient. In its most archaic version the sole hero spots a doe, who later turns out to be the daughter of a king, later marrying the hero and then they found a new homeland where his people may reproduce. This totemic tradition of the origins can he seen in the hungarian myths of origin, as well as in the oral ritual texts related to wedding ceremonies which connect Central Eurasia with Europe through the hun origin myths. The doe appearing in the Eastern Legends is the symbol of the Sun and the ancestress of the culture hero＇s nation. And this symbolic role descends to the cultural heritage of the Christian era later in the person of Virgin Mary. The aim of the paper is to find whether there are any correspondences between the two branches and if there are, the sort is to be determined; furthermore, the purposes of the hungarian legends are to be sought out about the Miracle Stag in possible relation to the deer legends on the two continents linked together by the Eurasian steppes.

The cis-trans Isomerisation of homologous 2-hydroxycycloalkanecarboxylic Acids under Basic Conditions

The cis→trans isomerisation of homologous 2-hydroxycycloalkanecarboxylic acids in strongly basic aqueous solution was studied starting from the cis isomers. It was found that the cyclopentane, cyclohexane and cycloheptane homologues afforded synthetically useful amounts of the trans acids and the procedure resulted in relatively small quantities of the corresponding olefinic acids. In contrast, the isomerisation of the cis-2-hydroxycyclooctanecarboxylic acid produced roughly equal amounts of the cis and trans isomers and the 1-cyclooctenecarboxylic acid at equilibrium. Molecular modelling with the PM3 semiempirical method of the reactants, products and the intermediates applying explicit water molecules as reaction medium gave a fair estimate for the rate sequence of the idealised （dehydration-free） isomerisation reactions in aqueous base solution.
