Remnant neuromuscular junctions in denervated muscles contribute to functional recovery in delayed peripheral nerve repair

Schwann cell proliferation in peripheral nerve injury(PNI)enhances axonal regeneration compared to central nerve ***,even in PNI,long-term nerve damage without repair induces degeneration of neuromuscular junctions(NMJs),and muscle atrophy results in irreversible *** peripheral regeneration of motor axons depends on the duration of skeletal muscle *** overcome this difficulty in nerve regeneration,detailed mechanisms should be determined for not only Schwann cells but also NMJ degeneration after PNI and regeneration after nerve ***,we examined motor axon denervation in the tibialis anterior muscle after peroneal nerve transection in thy1-YFP mice and regeneration with nerve reconstruction using *** number of NMJs in the tibialis anterior muscle was maintained up to 4 weeks and then decreased at 6 weeks after *** contrast,the number of Schwann cells showed a stepwise decline and then reached a plateau at 6 weeks after *** regeneration,we reconstructed the degenerated nerve with an allograft at 4 and 6 weeks after injury,and evaluated functional and histological outcomes for 10 to 12 weeks after grafting.A higher number of pretzel-shaped NMJs in the tibialis anterior muscle and better functional recovery were observed in mice with a 4-week delay in surgery than in those with a 6-week *** repair within 4 weeks after PNI is necessary for successful recovery in *** of synaptic acetylcholine receptor degeneration may play a key role in peripheral nerve *** animal experiments were approved by the Institutional Animal Care and Use Committee of Tokyo Medical and Dental University on 5 July 2017,30 March 2018,and 15 May 2019(A2017-311C,A2018-297A,and A2019-248A),respectively.

This paper discusses a distributed decision procedure for determining the electricity price for a real-time electricity market in an energy management system. The price decision algorithm proposed in this paper derives the optimal electricity price while considering the constraints of a linearized AC power grid model. The algorithm is based on the power demand-supply balance and voltage phase differences in a power grid. In order to determine the optimal price that maximizes the social welfare distributively and to improve the convergence speed of the algorithm, the proposed algorithm updates the price through the alternating decision making of market participants. In this paper, we show the convergence of the price derived from our proposed algorithm. Furthermore, numerical simulation results show that the proposed dynamic pricing methodology is effective and that there is an improvement in the convergence speed, as compared with the conventional method.

In vitro manipulation of induced pluripotent stem cells(iPSCs) by environmental factors is of great interest for three-dimensional(3D) tissue/organ induction. The effects of mechanical force depend on many factors, including force and cell type. However,information on such effects in i PSCs is lacking. The aim of this study was to identify a molecular mechanism in i PSCs responding to intermittent compressive force(ICF) by analyzing the global gene expression profile. Embryoid bodies of mouse i PSCs, attached on a tissue culture plate in 3D form, were subjected to ICF in serum-free culture medium for 24 h. Gene ontology analyses for RNA sequencing data demonstrated that genes differentially regulated by ICF were mainly associated with metabolic processes,membrane and protein binding. Topology-based analysis demonstrated that ICF induced genes in cell cycle categories and downregulated genes associated with metabolic processes. The Kyoto Encyclopedia of Genes and Genomes database revealed differentially regulated genes related to the p53 signaling pathway and cell cycle. q PCR analysis demonstrated significant upregulation of Ccnd1, Cdk6 and Ccng1. Flow cytometry showed that ICF induced cell cycle and proliferation, while reducing the number of apoptotic cells. ICF also upregulated transforming growth factor β1(Tgfb1) at both m RNA and protein levels, and pretreatment with a TGF-β inhibitor(SB431542) prior to ICF abolished ICF-induced Ccnd1 and Cdk6 expression. Taken together,these findings show that TGF-β signaling in i PSCs enhances proliferation and decreases apoptosis in response to ICF, that could give rise to an efficient protocol to manipulate i PSCs for organoid fabrication.

The effects of ethephon and abscisic acid(ABA) application on ripening-related genes of pre-harvest ‘Kohi' kiwifruit(Actinidia chinensis) were studied to clarify the possibility of the fruit ripening on the vine. The fruits were treated on the vine at mature stage with 250 μL·L^(-1)ethephon or 100 μmol·L^(-1)ABA, and the expression levels of chlorophyll synthase(Ac CLS), chlorophyllase1(Ac CLH1), polygalacturonase(Ac PG), expansin(Ac EXP),β-amylase(Acβ-AM), sucrose synthase(Ac SUSY), MADS-box SEPALLATA4/RIPENING INHIBITOR-like(Ac SEP4/RIN) and FRUITFUL-like(Ac TDR4/FUL) genes were analyzed. The expression levels of Ac PG, Ac EXP, Acβ-AM, and Ac SUSY increased in the ethephon-treated fruit, but those of Ac CLH1 at 6and 9 days after treatment and Ac CLS decreased. Moreover, the expression levels of Ac SEP4/RIN and Ac TDR4/FUL, the latter of which is associated with ethylene biosynthesis, were higher in the ethephon-treated fruit. The expression level of each gene in ABA-treated fruit was not significantly different from that of the untreated control. The results suggest that ethephon application increases the expression levels of Ac PG,Ac EXP, Acβ-AM, Ac SUSY, Ac SEP4/RIN, and Ac TDR4/FUL in ‘Kohi' kiwifruit on the vine.

We read with great interest the study by Way et al.1 published in this journal,which described and compared the acute changes in central arterial stiffness(AS),wave reflections,and central hemodynamics(CH)in response to a bout of high-intensity interval exercise(HIIE)and moderate-intensity continuous exercise(MICE)in adults with *** authors reported a significant reduction in augmentation index(AIx)corrected by a heart rate(HR)of 75 beats/min(AIx@75)and central systolic blood pressure after HIIE,without significant changes in the pulse wave velocity(PWV).In view of this important research topic and our recent contributions to it,we would like to raise some points of discussion that might help to better elucidate the findings observed in the current study,and also suggest pertinent directions to further studies.

A 76-year-old woman with a 5-mo history of recurrent diarrhea and generalized edema was admitted to our hospital. Colonoscopy revealed edematous mucosa,and histopathological examination was compatible with collagenous colitis. Protein leakage from the colon,particularly in the ascending portion,was identified on 99mTc-human serum albumin scintigraphy. Collagenous colitis associated with protein-losing enteropathy (PLE) without small bowel disease was diagnosed. Prednisolone treatment ameliorated diarrhea and hypoproteinemia. Collagenous colitis should be included in the differential diagnosis of chronic diarrhea with hypoproteinemia for appropriate management.
