Diagnostic sensitivity of imaging modalities for hepatocellular carcinoma smaller than 2 cm

AIM:To compare the imaging results with histology and to evaluate the diagnostic sensitivity of imaging modalities for hepatocellular carcinoma(HCC)smaller than 2 ***:Nodules smaller than 2 cm(n=34)revealed by ultrasonography(US)in 29 patients with liver cirrhosis were *** diagnosis of HCC was performed by ultrasonographic guidance:moderately-differentiated HCC(n=24);well-differentiated HCC(n=10).The patterns disclosed by the four imaging modalities defined the conclusive diagnosis of HCC:(1)contrast-enhanced computed tomography(CECT),hypervascularity in the arterial phase and washout in the equilibrium phase;(2)Sonazoid contrast-enhanced US(CEUS),hypervascularity in the early vascular phase and defect in the Kupffer phase;(3)gadolinium-ethoxybenzyl-diethylenetriamine pentaacetic acid(Gd-EOBDTPA)-enhanced magnetic resonance imaging(MRI),hypervascularity in the arterial phase and/or defect in the hepatobiliary phase;and(4)CT arterioportal angiography:hypervascularity by CT during arteriography and/ or perfusion defect by CT during arterial ***:Overall,the sensitivity of diagnosing HCC smaller than 2 cm was 52.9%(18/34)(95%CI:35.170.2)by CECT;67.6%(23/34)(95%CI:49.5-82.6)by Sonazoid CEUS;76.5%(26/34)(95%CI:58.8-89.3) by Gd-EOB-DTPA MRI;and 88.2%(30/34)(95%CI: 72.5-96.7)by CT arterioportal *** diagnostic sensitivity of detecting moderately-differentiated HCC by CECT,Sonazoid CEUS,Gd-EOB-DTPA MRI and CT arterioportal angiography was 62.5%(15/24)(95%CI: 40.6-81.2),79.2%(19/24)(95%CI:57.8-92.9),75.0% (18/24)(95%CI:53.3-90.2)and 95.8%(23/24)(95% CI:78.9-99.9),respectively.A significant difference(P< 0.05)was observed between CECT and CT arterioportal angiography in all *** was no difference between Sonazoid CEUS,Gd-EOB-DTPA MRI,and CT arterioportal *** combined sensitivity of Sonazoid CEUS and Gd-EOB-DTPA MRI was 94.1%(32/34).CONCLUSION:Changing the main diagnostic modality for HCC smaller than 2 cm

Intravenous immunoglobulin suppresses IL-10 production by activated B cells in vitro

A therapeutic preparation of polyclonal human IgG, i.e., intravenous immunoglobulin (IVIg), has been employed to treat several inflammatory and autoimmune disorders. B cells are supposed to be a target of IVIg, but the molecular mechanism is elusive because of the lack of a suitable experimental system. To gain an insight into the beneficial effect of IVIg on B cells, we first established an experimental setting in which IVIg modulates a murine B cell function in vitro, and then aimed at identifying the mechanistic features at the molecular level. Here we show that IVIg down-regulates IL-10 production by CpG-activated B cells in vitro. The responsible component of IVIg was identified as the F(ab’)2 portion, whose polyclonality is mandatory for the suppressive effect. IVIg, bound to the surface of activated B cells, was found to be co-localized with intracellular SHP-1 on confocal laser microscopy, suggesting that B cell-surface immunoreceptor tyrosine-based inhibitory motif-harboring receptors that recruit SHP-1 are target molecules for IVIg in our experimental setting. Overall, we postulate a scenario in which IVIg attenuates B cells by suppressing IL-10 production, a B cell growth factor, and thus down-regulates the production of pathogenic antibodies.

Recently,nanomaterials(NMs) have been used in industry,consequently arriving just around us as many kinds of *** are being used in medical devices as *** the amount of NMs contained in those products may be a little,we may need to pay attention to their safety because it may be different from that of their bulky *** present,not only OECD and ISO but also many other cooperative study groups are trying to establish test methods for NMs’ safety evaluation heading for the international cooperation on development and harmonization of the test *** this presentation,first I will talk about the particle size-dependent toxicity of polystyrene(PS) particles in vitro,and then introduce an in vivo test method performed with colloidal silica,silver and zinc *** particles were tested assuming them to be the aggregates of *** and genotoxicity of eleven size-different PS spheres with defined diameters ranging from 0.1 to 9.2 μm were tested in a Chinese hamster cell line,CHL[1].The 4.45-μm PS particles were most cytotoxic and smaller or larger particles showed less *** was a big toxicity difference between the 4.45-and 5.26-μm particles,although both precipitated and covered cells to the same extent in the *** analysis of particles and chemical analysis of dispersion vehicles did not show any difference between *** particles induced polyploidy in the chromosome aberration test and the intensity was correlated well with their *** images clearly showed the uptake of the 4.45-μm PS particles into CHL *** cytometric analysis seemed to measure the uptake of PS particles *** findings suggest that polyploidy may be induced by cytokinesis block due to particles *** are considering that such a size specific toxicity may be induced in the test with NM *** may need to provide information about aggregate sizes or some parameters indicating dispersion cond

Fibroblast growth factor receptor signaling as therapeutic targets in gastric cancer

Fibroblast growth factor receptors(FGFRs) regulate a variety of cellular functions, from embryogenesis to adult tissue homeostasis. FGFR signaling also plays significant roles in the proliferation, invasion, and survival of several types of tumor cells. FGFR-induced alterations, including gene amplification, chromosomal translocation, and mutations, have been shown to be associated with the tumor initiation and progression of gastric cancer, especially in diffuse-type cancers. Therefore, the FGFR signaling pathway might be one of the therapeutic targets in gastric cancer. This review aims to provide an overview of the role of FGFR signaling in tumorigenesis, tumor progression, proliferation, and chemoresistance. We also discuss the accumulating evidence that demonstrates the effectiveness of using clinical therapeutic agents to inhibit FGFR signaling for the treatment of gastric cancer.

Correlation between mood and heart rate variability indices during daily life

We investigated the correlation between mood and heart rate variability (HRV) indices during daily life. The RR-interval and body acceleration of 40 normal male subjects were recorded using ambulatory device for 48 to 72 hours. Every hour that the subjects were awake they registered their current mood on a Visual Analogue Scale questionnaire. The questionnaire scales eight of the subjects’ current moods. Those are happiness, tension, fatigue, worry, depression, anger, vigor, and confusion. The following four HRV indices were calculated. Those are heart rate, root mean square of successive differences of RR-interval sequence, the normalized high-frequency (0.15 - 0.4 Hz) power of RR-in- terval variability, and mean frequency in the high-frequency band of RR-interval variability. The calculated HRV indices data and the mood data were normalized individually, the data with body acceleration exceeding 30 mG were excluded from the analysis to reduce the effect of exercise, and the differences from the first day (?mood and ?HRV-index) were taken to reduce the effect of circadian rhythm. The most three highly correlated combinations were ?vigor and ?HFnu (R = –0.24, p < 0.0001), ?vigor and ?RMSSD (R = –0.24, p < 0.0001), and ?vigor and ?HR (R = 0.22, p < 0.001). Vigor exhibited the most significant correlations with HRV indices of eight moods.

Down-regulation of pancreatic transcription factors and incretin receptors in type 2 diabetes

Type 2 diabetes is one of the most prevalent and serious metabolic *** diabetic conditions,chronic hyperglycemia and subsequent induction of oxidative stress deteriorate pancreaticβ-cell function,which leads to the aggravation of type 2 *** such phenomena are well known as glucose toxicity,its molecular mechanism remains *** this review article,we describe the possible molecular mechanism forβ-cell dysfunction found in type 2 diabetes,focusing on(1)oxidative stress,(2)pancreatic transcription factors(PDX-1 and MafA)and(3)incretin receptors(GLP-1 and GIP receptors).Under such conditions,nuclear expression levels of PDX-1 and MafA are decreased,which leads to suppression of insulin biosynthesis and *** addition,expression levels of GLP-1 and GIP receptors are decreased,which likely contributes to the impaired incretin effects found in *** together,it is likely that downregulation of pancreatic transcription factors(PDX-1and MafA)and down-regulation of incretin receptors(GLP-1 and GIP receptors)explain,at least in part,the molecular mechanism forβ-cell dysfunction found in type 2 diabetes.

Image Findings Following Vertebroplasty in Osteoporotic Vertebral Compression Fractures: Bone Healing and Sagittal Alignment

Purpose: To clarify the effect of percutaneous vertebroplasty for vertebral compression fracture by assessing the changes of radiographic and CT image findings. Materials and Methods: A retrospective radiological analysis comprising 101 vertebrae of 48 patients who underwent percutaneous vertebroplasty for painful osteoporotic vertebral compression fracture was conducted. Whole spine radiographs and CT images were compared in patients preoperatively and 6 months postoperativey. Sagittal Cobb angles in three regions, sagittal vertical axis, and pelvic tilt were measured using whole spine lateral radiographs. CT findings due to the vertebral compression fracture, its healing process, and complications were evaluated. Results: On radiographs, sagittal alignment had an average gain of no more than 2.5° in each region. Sagittal vertical axis and pelvic tilt did not show significant change. Out of 68 vertebrae that demonstrated cortical disruption on preoperative CT, 37 (54%) demonstrated fusion of disrupted cortex on postoperative CT. Conclusion: No significant difference was observed between preoperative and postoperative spinal sagittal alignment on radiographs. However, CT did reveal healing process through fusion of disrupted cortex, intervertebral bridging, and increased density of cancellous bone. It is suggested that cement in the space of fracture may play a role in both mechanical stability and bone union.

SMALL ORGAN SIZE 1 and SMALL ORGAN SIZE 2/DWARF AND LOW-TILLERING Form aComplex to Integrate Auxin and Brassinosteroid Signaling in Rice

Although auxin and brassinosteroid （BR） synergistically control various plant responses, the molecular mechanism underlying the auxin-BR crosstalk is not wen understood. We previously identified SMOS1, an auxin-regulated APETALA2-type transcription factor, as the causal gene of the small organ size 1 （smosl） mutant that is characterized by a decreased final size of various organs in rice. In this study, we identified another smos mutant, smos2, which shows the phenotype indistinguishable from smosl. SMOS2 was identical to the previously reported DWARF AND LOWoTILLERING （DLT）, which encodes a GRAS protein involved in BR signaling. SMOS1 and SMOS2/DLT physically interact to cooperatively enhance transcriptional transactivation activity in yeast and in rice nuclei. Consistently, the expression of OsPHI-1, a direct target of SMOS1, is upregulated only when SMOS1 and SMOS2/DLT proteins are both present in rice ceils. Taken together, our results suggest that SMOS1 and SMOS2/DLT form a keystone complex on auxin-BR signaling crosstalk in rice.
