L-carnitine improves developmental competence of buffalo oocytes in vitro

Objective:To investigate the effect of L-carnitine on in vitro maturation and subsequent in vitro embryo production of buffalo ***:Cumulus oocyte complexes(COCs)were aspirated from ovaries of slaughtered *** were classified into good and fair qualities based on morphological observation of numbers and integrity of cumulus cells surrounding the *** categories of COCs were placed in in vitro maturation medium with supplementation of different concentrations(0,0.250,0.375 or 0.500 mg/mL)of *** from both qualities were in vitro fertilized and in vitro cultured for 7 days,to examine the developmental ***:Supplementation of L-carnitine to in vitro maturation medium increased the cumulus cell expansion rate of COCs to grade A,and reduced the cumulus cell expansion of COCs to grade B and grade C in both good and fair quality ***,L-carnitine induced the in vitro meiotic progression of buffalo oocytes to metaphaseⅡin both good and fair quality ***,L-carnitine reduced the rate of oocyte degeneration in both good and fair quality oocytes.L-carnitine increased the rate of cleaved formation at day 2 and blastocyst formation at day 7 during in vitro culture in both qualities of ***,a higher rate of blastocyst production was observed in L-carnitine-treated fair quality oocytes,which was higher than the results in the untreated good quality ***:L-carnitine enhances meiotic maturation and subsequent embryo development from both good and fair quality buffalo oocytes.

This paper discusses the impact of e-governance powered by blockchain in the project and program management industry. With the rise in technological innovations, many countries have turned to e-governance for efficiency in service delivery, transparency, and decision-making. E-governance backed by blockchain technology entails improving the public services provision by implementing structures of information and communication technologies. There are many challenges with the traditional project management approach that causes organizations and its stakeholders’ cost and time. Thus, the introduction of blockchain has given many organizations a new approach to adopt in order to eliminate the challenges associated with the typical project management approach. In spite of the cutting-edge technology of blockchain and its broad applications in program management and e-governance, there are still many challenges that restrain its adoption on a broader scale. The research discusses the challenges of the blockchain deployment in the program management field and e-governance in private and government sectors and it highlights the efforts put by both sectors to make use of the technology. Also, the research covers the opportunities and the advantages of a blockchain adoption in various segments. The paper uses various case studies in the UAE, in both private and government sectors, and a qualitative research approach was implemented. The case studies were on government entities such as Smart Dubai and the Ministry of Health Prevention and also private entities like DP World and Emirates NBD. The paper concludes with recommendations and solutions on how to maximize the benefit of blockchain in the program management industry and how it is improving the decision-making process.

The Unconventional Oil and Gas industry has seen growth over the last ten years that has drastically transformed the domestic energy outlook while bringing up increased concerns over climate and environmental issues. The rise of ESG and RSG can be seen as direct answers to these growing issues as communities and operators have both begun to demand better practices to limit the overall effects of UOG production. Few quantifiable metrics exist that holistically try to determine the overall effect UOG production has on local water resources. The FR2 metric/framework developed in this paper attempts to use commonly kept data such as water withdrawn and flowback volumes in conjunction with a new water stress index to quantify the effects operators are having on local water supplies. Testing this framework on a handful of operators from the Marcellus basin using open-source data revealed the value added by these methods as well as their use in a general RSG program.

AIM： To evaluate the qualitative and quantitativechanges in N-linked glycosylation, which occurredin association with diethyl nitrosamine-inducedhepatocellular carcinoma （HCC） in ***： Liver tissues of （1） normal （non-tumorbearing）rats; and （2） tumor-bearing rats; were collectedand were used for histological and GlycanMap？ ***, GlycanMap？ analysis is a high-throughputassay that provides a structural and quantitativereadout of protein-associated glycans using a unique,automated 96-well assay technology coupled tomatrix-assisted laser desorption/ionization time-offlightmass spectrometry and custom *** studies were carried out to ensure thedevelopment of HCC in the tested ***： The N-glycomic analysis revealed 5glycans; Glc1Man9GlcNAc2, Gal2Man3GlcNac4Fuc1Neu1,Man4G l c N a c 2, G a l 2Man3G l c N a c 4Neu3OAc 3, andMan3GlcNac5Fuc1, which showed significant changesin rat HCC tissues when compared with normal livertissues. Four glycans were increased （P 〈 0.05） andGlc1Man9GlcNAc2 was decreased （5.89 ± 0.45 vs 3.54± 0.21, P 〈 0.01） in HCC tissues compared to normal liver tissues. An increase （66.5 ± 1.05 vs 62.7 ± 1.1,P 〈 0.05） in high-mannose structures in HCC rats wasobserved compared to normal rats. Importantly, HCCrats showed an increase （P 〈 0.05） in both tumorassociatedcarbohydrates and in branched glycans. Thechanges in glycans correlated well with glycan flowchanges reported in the glycan biosynthetic pathway,which indicates the importance of enzyme activitiesinvolved in glycan synthesis at different ***： The reported HCC-associated changesin glycan flow and subcellular localization explain theincrease in high mannose glycans and siayl Lewisglycans common in HCC liver tissues.

PIK3R5 is the regulatory subunit of Phosphoinositide 3-kinase γ (PI3Kγ) that is responsible for phosphory-lating membrane lipids to activate the AKT pathway. PIK3R5 binds Gβγ and facilitates the interaction with p110γ catalytic subunit (PIK3CG) during PI3Kγ activation. The identification of PIK3R5 P629S mutation in AOA2 patients indicated a potential defect in the AKT pathway resulting from impaired PIK3R5 interaction with Gβγ and PIK3CG, defective AKT pathway can result in cerebellar cell death causing neurological symptoms. Our in silico macromolecular docking of the wild type and mutant PIK3R5 protein models with ligand revealed an energy requirement to maintain the mutant complexes compared to no energy required to maintain the wild type complexes, in addition, the mutant structures were loose compared to rigid wild type structures, such structural changes may impair the molecular function of the PIK3R5 and hence affect the AKT pathway.

Labroides dimidiatus has been proven to remove ectoparasites and monogeneans from client fishes and studies showed that they were not infected with the parasite. Due to this, there is a possibility that a defense mechanism against pathogen and parasitic invasion exist in the epidermal mucus which serves as a mechanical as well as biochemical barrier. The study was performed to identify the amino acid and fatty acid components using GC （gas chromatography） and HPLC （high performance liquid chromatography） in epidermal mucus ofL. dimidiatus. The present study revealed 16 components of amino acid and 22 types of fatty acid in epidermal mucus ofL. dimidiatzts. Linoleic acid （C18：2n6c） was the most prominent PUFA （polyunsaturated fatty acid） which contributed approximately 11.69% of total fatty acids. The other major fatty acids are palmitic acid （C 16：0）, oleic acid （C 18： l n9c）, linoledaidic acid （C18：2n6t）, arachidic acid （C20：0）, Gamma-Linoleic acid （C18：3n6） and gadoleic acid （C20：1） which contained reasonable amounts of 9.52%, 8.06%, 6.26%, 8.33%, 6.21% and 9.05% of total fatty acids, respectively. This present study also demonstrated the presence of various amino acids in skin extract. Glycine, glutamine, arginine, asparagin and alanine were found at high concentration of 8.09%, 6.95%, 5.73%, 4.74%, 4.58% respectively. The most abundance percentage of linoleic acid （C18：2n6c） was found to be the metabolic precursor of arachidonic acid （AA） which inducing platelet aggregation, facilitate the blood clotting process and adhesion in endothelial cells during wound healing and might be responsible for rapid tissue growth in L. dimidiatus. It can be concluded that the amino acid and fatty acid profile from the epidermal mucus ofL. dimidiatus contains most of the essential components required to play a possible role in its defense mechanism. A greener method has been tested to utilize algal biomass as a feedstock to produce bio-oil in addition to acetone,butanol,and ethanol(ABE)*** hydrolysis treatments were used prior to fermentation including combination of thermal,chemical,and enzymatic,which resulted in maximum sugar release of 27.78 g/***-based terpenes was used instead of common toxic chemicals together with Clostridial fustants to produce bio-alcoholic *** fusion technique were used to produce the novel Clostridia fusants(***+*** and ***+***).Fused strains were then subjected to UV radiation for strain *** fusansts showed clear improvement in thermal stability and resistance to biobutanol *** experiments showed maximum biobutanol final production of 7.98 g/L using CbCt versus 7.39 g/L using *** extraction from virgin algae was tested using a green,bio-based approach using ter-penes with ultrasonication and green Bligh and Dyer method,*** preliminary study on algal biomass,the combinations of ultrasonication followed by the green Bligh and Dyer have resulted in oil yield of 46.27%(d-limonene)and 39.85%(p-cymene).Oil extraction from an algae sample following fermentation using the combined extraction method resulted in significantly higher oil yield of 65.04%.

Determinants of Risky Sexual Behaviour among Inmates in Katsina Correctional Service: A Mixed Methods Study

Background: Incarceration does not vitiate sexual desire. Therefore, sexual activities occur behind bars, regardless of the stringent rules in correctional institutions. However, little has been documented about risky sexual behaviors, determinants, and experiences among inmates of correctional institutions in Katsina State, Nigeria. The study determined the sexual practices, prevalence, and determinants of risky sexual behaviors among prison inmates in Katsina. Methodology: A descriptive cross-sectional design with concurrent mixed methods of data collection was used to interview 216 inmates using a semi-structured questionnaire supplemented with 22 in-depth interviews. Result: Most (94%) respondents were male, almost half (44.4%) had secondary education, more than half (55.1%) were engaged in business or trading before incarceration, and the majority (75.9%) were awaiting trial. Nearly all inmates (98.6%) reported having sexual desire. Drug use (80.1%) and unprotected sex (79.2%) were quite common but sexual violence was low (7.4%). The number of sexual partners of respondents remained a significant predictor of sexual activity. Inmates who had one (1) or no sexual partner were 64% less likely to engage in risky sexual activity within the confines of the prison (AOR = 0.36, 95% CI = 0.20 - 0.63, p = 0.01).
