Evaluation of thermostable endoglucanase in Paenibacillus lautus strain BHU3 for yield enhancement

In the present study,Paenibacillus lautus strain BHU3 isolated from landfill soil was evaluated for the presence of potential endoglucanases which are the first candidate of cellulase enzyme system to act on ***-silico analysis revealed high potential thermostable endoglucanases which can efficiently interact with *** most potent and thermostable endoglucanase(locus tag ***30_18280)belonged to glycosyl family-5 and had interaction energy of−12.981 kcal/mol for the best docked cluster containing three out of ten docking conformations,and Tm value of 73.3℃.MD simulation of 100 ns proved highly stable binding interactions of CPZ30_18280 endoglucanase with cellulose with root mean square deviation(RMSD)values ranging from 0.15 to 0.30 *** interactions with characteristic active site residues(tyrosine,tryptophan and aspartate)of glycosyl family-5 endoglucanases were ***,to enhance the production of endoglucanases,the fermentation conditions were optimized employing approaches like one factor at a time(OFAT)and response surface methodology(RSM).Maximum activity of endoglucanase was determined at 60℃.The optimized condi-tions for enhanced production of endoglucanase(10.15 U/mL)were pH 6.63,yeast extract conc.3.44 g/L,wheat bran 3.59%,and inoculum size 2.65%.Hence,*** strain BHU3 has enormous potential to synthesize highly efficient thermostable endoglucanases under optimized regime using ***,it could find immense industrial applications including large scale cellulose conversion to bioethanol.

Gas hydrate saturation from NGHP 02 LWD data in the Mahanadi Basin

During the Indian National Gas Hydrate Program(NGHP)Expedition 02,Logging-while-drilling(LWD)logs were acquired at three sites(NGHP-02-11,NGHP-02-12,and NGHP-02-13)across the Mahanadi Basin in area *** applied rock physics theory to available sonic velocity logs to know the distribution of gas hydrate at site NGHP-02-11 and *** physics modeling using sonic velocity at well location shows that gas hydrate is distributed mainly within the depth intervals of 150-265 m and 100 -215 mbsf at site NGHP-02-11 and NGHP-02-13,respectively,with an average saturation of about 4%of the pore space and the maximum concentration of about 40%of the pore space at 250 m depth at site NGHP-02-11,and at site NGHP-02-13 an average saturation of about 2%of the pore space and the maximum concentration of about 20%of the pore space at 246 m depth,as gas hydrate is distributed mainly within 100-246 mbsf at this *** of gas hydrate estimated from the electrical resistivity method using density derived porosity and electrical resistivity logs from Archie's empirical formula shows high saturation compared to that from the sonic ***,estimates of hydrate saturation based on sonic P-wave velocity may differ significantly from that based on resistivity,because gas and hydrate have higher resistivity than conductive pore fluid and sonic P-wave velocity shows strong effect on gas hydrate as a small amount of gas reduces the velocity significantly while increasing velocity due to the presence of *** site NGHP-02-11,gas hydrate saturation is in the range of 15%e30%,in two zones between 150-180 and 245-265 *** NGHP-02-012 shows a gas hydrate saturation of 20%e30%in the zone between 100 and 207 *** NGHP-02-13 shows a gas hydrate saturation up to 30%in the zone between 215 and 246 *** observations from rock physics modeling and Archie’s approximation show the gas hydrate concentrations are relatively low(<4%of the pore space)at the sites of the

Abstract： Estimation of evapotranspiration （ET） for mountain ecosystem is of absolute importance since it serves as an important component in balancing the hydrologic cycle. The present study evaluates the performance of original and location specific calibrated Hargreaves equation （HARG） with the estimates of Food and Agricultural Organization （FAO） Penman Monteith （PM） method for higher altitudes in East Sikkim, India. The results show that the uncalibrated HARG model underestimates ET0 by 0.35 mm day^-1 whereas the results are significantly improved by regional calibration of the model. In addition, this paper also presents the variability in the trajectory associated with the climatic variables with the changing climate in the study site. Non- parametric Mann-Kendall （MK） test was used to investigate and understand the mean monthly trend of eight climatic parameters including reference evapotranspiration （ET0） for the period of 1985 - 2009. Trend of ET0 was estimated for the calculations done by FAO PM equation. The outcomes of the trend analysis show significant increasing （p ≤ 0.05） trend represented by higher Z-values, through MK test, for net radiation （Rn）, maximum temperature （Tmax） and minimum temperature （Train）, especially in the first months of the year. Whereas, significant （0.01 ≥ p ≤0.05） decreasing trend in vapor pressure deficit （VPD） and precipitation （P） is observed throughout the year. Declining trend in sunshine duration, VPD and ET0 is found in spring （March - May） and monsoon （June - November） season. The result displays significant （0.01≤ p ≤0.05） decreasing ET0 trend between （June - December） except in July, exhibiting the positive relation with VPD followed by sunshine duration at the station. Fetal kidney stem cells ameliorate cisplatin induced acute renal failure and promote renal angiogenesis

AIM: To investigate whether fetal kidney stem cells(f KSC) ameliorate cisplatin induced acute renal failure(ARF) in rats and promote renal ***: The f KSC were isolated from rat fetuses of gestation day 16 and expanded in vitro up to 3rd passage. They were characterized for the expression of mesenchymal and renal progenitor markers by flow cytometry and immunocytochemistry, respectively. The in vitro differentiation of f KSC towards epithelial lineage was evaluated by the treatment with specific induction medium and their angiogenic potential by matrigel induced tube formation assay. To study the effect of f KSC in ARF, f KSC labeled with PKH26 were infused in rats with cisplatin induced ARF and, the blood and renal tissues of the rats were collected at different time points. Blood biochemical parameters were studied to evaluate renal function. Renal tissues were evaluated for renal architecture, renal cell proliferation and angiogenesis by immunohistochemistry, renal cell apoptosis by terminal deoxynucleotidyl transferase nickend labeling assay and early expression of angiogenic molecules viz. vascular endothelial growth factor(VEGF), hypoxia-inducible factor(HIF)-1α and endothelial nitric oxide synthase(eN OS) by western ***: The fK SC expressed mesenchymal markers viz. CD29, CD44, CD73, CD90 and CD105 as well asrenal progenitor markers viz. Wt1, Pax2 and Six2. They exhibited a potential to form CD31 and Von Willebrand factor expressing capillary-like structures and could be differentiated into cytokeratin(CK)18 and CK19 positive epithelial cells. Administration of fK SC in rats with ARF as compared to administration of saline alone, resulted in a significant improvement in renal function and histology on day 3(2.33 ± 0.33 vs 3.50 ± 0.34, P < 0.05) and on day 7(0.83 ± 0.16 vs 2.00 ± 0.25, P < 0.05). Understanding arsenic behavior in alluvial aquifers:Evidence from sediment geochemistry,solute chemistry and environmental isotopes

The hydro-geochemistry and isotopic variations in groundwater,coupled with sediment geochemistry,were investigated in the Middle Gangetic Plain,India,to better understand the aquifer dynamics that influence the arsenic(As)evolution and ***-four groundwater samples,thirteen River water samples,and two sediment cores(33 mbgl)were *** samples were analyzed for major ions and trace metals,including As and stable isotopic variability(δ^(2)H,δ^(18)O,andδ^(13)C).The study area was categorized into older and younger alluvium based on existing geomorphological *** alluvium exhibits higher As enrichment in sediment and groundwater,ranging of 2.59–31.52 mg/kg and bdl to 0.62 mg/*** samples were thermodynamically more stable with As(OH)_(3)species ranging from 88.5%to 91.4%and FeOOH from 69%to 81%,*** and mineralogical analysis suggested goethite and siderite act as a source and sink for ***,statistical analysis suggested reductive dissolution as the primary mechanism for As mobilization in the study ***-temporal analysis revealed elevated concentrations of As in the central and northeastern regions of the study *** isotope(δ^(2)H andδ^(18)O)analysis inferred active recharge conditions primarily driven by *** depleted d-excess value and enrichedδ^(18)O in the groundwater of younger alluvium indicate the effect of groundwater recharge with significant evaporation *** recharge potentially decreased the quantity of arsenic in groundwater,whereas evaporation enrichment increased *** infiltration during recharge introduces oxygenated water into the aquifer,leading to changes in the redox conditions and facilitating biogeochemical *** carbon isotope(δ^(13)C)results suggest that high microbial activity in younger alluvium promotes As leaching from sediment into the groundwater.
