Single Molecule Analysis of the Arabidopsis FRA1 Kinesin Shows that It Is a Functional Motor Protein with Unusually High Processivity

The Arabidopsis FRA1 kinesin contributes to the organization of cellulose microfibrils through an unknown mechanism. The cortical localization of this kinesin during interphase raises the possibility that it transports cell wallrelated cargoes along cortical microtubules that either directly or indirectly influence cellulose microfibril patterning. To determine whether FRA1 is an authentic motor protein, we combined bulk biochemical assays and single molecule fluorescence imaging to analyze the motor properties of recombinant, GFP-tagged FRA1 containing the motor and coiled-coil domains （designated as FRAI（707）-GFP）. We found that FRAI（707）-GFP binds to microtubules in an ATP-dependent manner and that its ATPase activity is dramatically stimulated by the presence of microtubules. Using single molecule studies, we found that FRAI（707）-GFP moves processively along microtubule tracks at a velocity of about 0.4 μm s-1. In addition, we found that FRAI（707）-GFP is a microtubule plus-end-directed motor and that it moves along microtubules as a dimer. Interestingly, our single molecule analysis shows that the processivity of FRAI（707）-GFP is at least twice the processivity of conventional kinesin, making FRA1 the most processive kinesin to date. Together, our data show that FRA1 is a bona fide motor protein that has the potential to drive Iong-distance transport of cargo along cortical microtubules.

In this study, we describe the partial genomic organization of ribosomal protein S7 gene isolated from the mosquito Anopheles stephensi. Initially a 558 bp partial cDNA sequence was amplified as precursor mRNA sequence containing 223 bp long intron. 5＇ and 3＇ end sequences were recovered using end specific rapid amplification of cDNA ends （RACE） polymerase chain reaction. The full-length cDNA sequence was 914 nucleotide long with an open reading frame capable of encoding 192 amino acid long protein with calculated molecular mass of 22174 Da and a pI point of 9.94. Protein homology search revealed 〉75% identity to other insect＇s S7 ribosomal proteins. Analysis of sequence alignment revealed several highly conserved domains, one of which is related to nuclear localization signal （NLS） region of human rpS7. Interestingly, intron nucleotide sequence comparison with A. gambiae showed a lesser degree of conservation as compared to coding and untranslated regions. Like this, early studies on the genomic organization and cDNA/ Expressed sequence tag analysis （EST） could help in genome annotation ofA. stephensi, and would be likely to be sequenced in the future.

In this paper,we report synthesis of pure SrMoO_(4),Sm^(3+)(1 at%-5 at%)doped SrMoO_(4)and Bi^(3+)(1 at%-3 at%)co-doped in 4 at%Sm^(3+)doped SrMoO_(4)(SrMoO_(4):4 Sm^(3+))phosphors by solution combustion *** X-ray diffraction(XRD)analysis reveals the tetragonal phase of all samples,also Bi^(3+)co-doping supports crystallite size growth and reduces lattice *** analysis of Sm^(3+)doped SrMoO_(4)ascertains a decrease in band gap and Bi^(3+)co-doping confirms the emergence of an absorbance peak at around 308 nm attributed to Bi^(3+)energy ***(PL)analysis ascertains an increase in emission peaks for Sm^(3+)doped SrMoO_(4)up to 4%concentration,which are attributed to an electron transition from 4 G5/2 to 6 HJ(J=5/2,7/2,9/2,and 11/2)energy levels of Sm^(3+)*** have explained the effects of Bi^(3+)co-doping on the luminescence of Sm^(3+)doped SrMoO_(4).The reduced microstrain and increased crystallinity of the phosphors as a result of Bi^(3+)co-doping and their correlation with the luminescence of Sm^(3+)ions are discussed.

The present study evaluated the potential of white-rot fungal strain Coriolus versicolor to decolorize five structurally different dyes in sequential batch reactors under optimized conditions. The experiments were run continuously for seven cycles of 8 d each. High decolorizing activity was observed even during the repeated reuse of the fungus, especially when the old medium was replaced with fresh medium after every cycle. Biodegradation was the dominating factor as the fungus was able to produce the enzyme laccase mainly, to mineralize synthetic dyes. The nutrients and composition of the medium played important roles in sustaining the decolorisation potential of the fungus. Corncob was found be an easy and cheap substitute for carbon source for the fungus. Glucose consumption by the fungus was in accordance to its decolorisation activity and chemical oxygen demand （COD） reduction.

The necessity for understanding normal human cognitive processes and behavior, and themechanisrns which result in dysfunction in these processes are dependant on utilization of a suitable animal model. In order to develop pharmaceutical agents to alleviate mental disturbances and enable the individual to cope within the norms of society, it is incumbent upon investigators to choose a species in which pharmacokinetic principles are established and resemble those of hurnans. The choice of rats in cognition research studies has specific advantages in that these anirnals possess similar pharrnacodynamic parameters to hurnans. Further advantages include availability, low cost, ease of breeding, maintenance and an extensive literature database which enable comparisons to present findings. However, there are substantial differences in the perforrnance of various rat strains in tasks of learning, memory, attention, and responses to stress or drugs. In addition to rat strain, quantity of thed also exerts profound consequences on animal behavior. The aim of this review is to demonstrate that there are differences in the central nervous systern responsivencess of rat strains to chemicals and these could be related to factors such as source of supplier, type and quantity of feed, or season of the year. It is also evident that the genotype differs amongst strains and this may be responsible for the observed differences in CNS sensitivity to chemicals. Strain differences must be identified and taken into consideration in interpretation of assessrnent of neurobehavioural functions. It is also incumbent upon the investigators to utilize healthy (diet-controlled) animal models.

The current study investigates the behavior of wire electric discharge machining （WEDM） of the super alloy Udimet-L605 by employing sophisticated machine learning approaches. The experimental work was designed on the basis of the Taguchi orthogonal L27 array, consid- ering six explanatory variables and evaluating their influ- ences on the cutting speed, wire wear ratio （WWR）, and dimensional deviation （DD）. A support vector machine （SVM） algorithm using a normalized poly-kernel and a radial-basis flow kernel is recommended for modeling the wire electric discharge machining process. The grey rela- tional analysis （GRA） approach was utilized to obtain the optimal combination of process variables simultaneously, providing the desirable outcome for the cutting speed, WWR, and DD. Scanning electron microscope and energy dispersive X-ray analyses of the samples were performed for the confirmation of the results. An SVM based on the radial-basis kernel model dominated the normalized poly- kernel model. The optimal combination of process vari- ables for a mutually desirable outcome for the cutting speed, WWR, and DD was determined as Ton1, Toffa, Ip1, WT3, SV1, and WF3. The pulse-on time is the significant variable influencing the cutting speed, WWR, and DD. The largest percentage of copper （8.66%） was observed at the highest cutting speed setting 7.05% of copper at the low of the machine compared to cutting speed setting of the machine.

In recent years,social media platforms have gained immense *** a result,there has been a tremendous increase in content on social media *** content can be related to an individual’s sentiments,thoughts,stories,advertisements,and news,among many other content *** the recent increase in online content,the importance of identifying fake and real news has ***,there is a lot of work present to detect fake news,a study on Fuzzy CRNN was not explored into this *** this work,a system is designed to classify fake and real news using fuzzy *** initial feature extraction process is done using a convolutional recurrent neural network(CRNN).After the extraction of features,word indexing is done with high ***,based on the indexing measures,the ranking process identifies whether news is fake or *** fuzzy CRNN model is trained to yield outstanding resultswith 99.99±0.01%*** work utilizes three different datasets(LIAR,LIAR-PLUS,and ISOT)to find the most accurate model.

There are various intense forces causing customers to use evaluated data when using social media platforms and microblogging ***,customers throughout the world share their points of view on all kinds of topics through these *** massive volume of data created by these customers makes it impossible to analyze such data ***,an efficient and intelligent method for evaluating social media data and their divergence needs to be ***,various types of equipment and techniques are available for automatically estimating the classification of *** analysis involves determining people’s emotions using facial *** analysis can be performed for any individual based on specific *** present study describes the analysis of an image dataset using CNNswithPCA intended to detect people’s sentiments(specifically,whether a person is happy or sad).This process is optimized using a genetic algorithm to get better ***,a comparative analysis has been conducted between the different models generated by changing the mutation factor,performing batch normalization,and applying feature reduction using *** steps are carried out across five experiments using *** maximum accuracy obtained is 96.984%,which is associated with the Happy and Sad sentiments.
