Pharmacokinetic Comparison and Bioequivalence Evaluation of Two 20-mg Vonoprazan Fumarate Tablets in Bangladeshi Healthy Male Subjects

Background: Vonoprazan fumarate, a novel potassium-competitive acid blocker, outperforms traditional proton pump inhibitors in acid suppression and can be effectively combined with antibiotics to eradicate Helicobacter pylori. Objective: The study aimed to determine if two Vonoprazan formulations—Vonoprazan Fumarate 20 mg Tablet of Beximco Pharmaceuticals Limited, Bangladesh (test product) and Takecab 20 mg Tablet of Takeda Pharmaceutical Company Limited, Japan (reference product)—met FDA’s bioequivalence requirements by comparing their pharmacokinetic characteristics in healthy Bangladeshi adults. Methods: This was a single-center, randomized, open-label, two-period, two-sequence, laboratory-blind, double-crossover experiment. After 10 hours of fasting, 18 subjects were randomly assigned to receive a single oral dose of either formulation. During each treatment period, blood samples were collected at specific times (pre-dose and up to 48 hours post-dose) to measure plasma Vonoprazan levels using liquid chromatography-tandem mass spectrometry. A non-compartmental model was used to calculate pharmacokinetic parameters using the plasma drug concentration-time profile. A statistical comparison of the pharmacokinetic parameters of the two formulations of the test and reference product was conducted using SAS® statistical software to assess the bioequivalence. Primary pharmacokinetic parameters (Cmax, AUC0-t, and AUC0-∞) and secondary parameters (Tmax, T1/2, Kel, and AUC extrapolation) were calculated for both drug formulations. If the confidence intervals for the natural log-transformed Cmax, AUC0-t, and AUC0-∞ values fell between 80% and 125%, the drug products would be considered bioequivalent. Result: The geometric mean ratio of Vonoprazan between the test and reference groups was found to be 109.04% (99.47% - 119.53%), 101.37% (95.58% - 107.50%), and 101.24% (95.43% - 107.41%), with 90% confidence intervals (CIs) for the Cmax, AUC0–t, and AUC0–∞, and these outcomes

The analytic cosmological solutions of Einstein's field equations for a type of static metric representing plane, spherical and hyperbolic symmetric spaces are presented and their properties are discussed separately. A general type of solution is obtained which represents the plane, spherical and hyperbolic symmetric cosmological models. Its physical properties are also discussed in details.

A Simple,Efficient and Rapid Method for Good Quality DNA Extraction from Rice Grains

An efficient and good DNA extraction protocol should be simple, affordable and yield enough DNA with high quality. Rice(Oryza sativa L.) DNA extraction methods often use seedlings or leaves rather than the grains and tend to be time-consuming, involve multiple steps, and use hazardous chemicals and expensive enzymes. Rice grains offer several benefits over seedlings and leaves as a source of DNA for genetic analysis. However, these benefits are underutilized because the bulk of a rice grain is made up of starch. It is particularly important, but difficult to get rid of the starch while extracting DNA from rice grains. This co-precipitated polysaccharide is a known inhibitor of DNA polymerase activity in polymerase chain reaction(PCR). We describe here a very simple and highly affordable Chelex~?-100 based DNA extraction method from rice grains. It does not require any hazardous chemicals or enzymes. This method reproducibly extracts DNA with good purity indices(A_(260)/A_(230) and A_(260)/A_(280) values), but requires only a few steps.

Identification of visible colored dissolved organic matter in biological and tertiary municipal effluents using multiple approaches including PARAFAC analysis

This study provided insights into the persistent yellowish color in biological and tertiary effluents of municipal wastewater through a multi-characterization approach and fluorescence excitation-emission matrix-parallel factor(EEM-PARAFAC)*** characterization was performed on three to five full-scale municipal wastewater treatment plants(WWTPs),including differential log-transformed absorbance(DLn A)spectroscopy,resin fractionation,size-exclusion chromatography for apparent molecular weight analysis(SECAMW),and X-ray photoelectron spectroscopy(XPS)*** acids(HPOA)were abundant in visible colored dissolved organic matter(DOM).The SEC-AMW result showed that the molecular weight of the colored substances in the secondary effluents is mainly distributed in the range of 2–3 k *** XPS analysis,C-O/C-N and pyrrolic/pyridonic(N-5)were found to be positively correlated with *** component models were built on biological(two components)and tertiary effluent(three components)and the correlation analysis revealed that PARAFAC component 2 in biological effluent(BE-C2)and component 1in tertiary effluent(TE-C1),which were ascribed to Hydrophobic acids and Humic acid-like,were the responsible visible colored DOM components cause yellowish *** addition,component similarity testing found that the identified visible colored DOM PARAFAC BE-C2,and PARAFAC TE-C1 were identical(0.96)in physicochemical properties,with 4%removal efficacy on average,compared with 11%for invisible colored *** implied that tertiary effluents containing colorants(TE-C1)were resistant to degradation/removal using different disinfection and filtration processes in advanced *** sheds light on many physicochemical aspects of PARAFAC-identified visible colored DOM components and provides spectral data to build an online monitoring system.

A novel hole-transport material(HTM)based on an anthradithiophene central bridge named BTPA-7 is *** comparison to spiro-OMeTAD(2,2’,7,7’-tetrakis-(N,N-di-p-methoxyphenylamine)-9,9’-spirobifluorene),the synthetic steps of BTPA-7 are greatly reduced from 6 to 3 and the synthetic cost of BTPA-7 is nearly a half that of ***,BTPA-7 exhibits a relatively lower conductivity but higher hole mobility and higher glass transition temperature(Tg)than *** with the photovolatic performance for spiro-OMeTAD,FA0.85MA0.15PbI3 and MAPbI3 PSC devices based on BTPA-7 exhibit slightly lower PCEs with the values of 17.58%(18.88%for spiro-OMeTAD)and 11.90%(13.25%for spiro-OMeTAD),***,a dramatically higher JSC of PSC based on BTPA-7is achieved,which arises from the higher hole mobility of *** addition,the relatively hydrophobic character of BTPA-7 eventually enhances the PSC device *** cost,higher hole mobility,higher Tg,satisfactory photovoltaic performance,and superior device stability of BTPA-7 can be utilized as a substitute for spiro-OMeTAD in PSCs.

TaARPC5 is required for wheat defense signaling in response to infection by the stripe rust fungus

Numerous studies using a combination of confocal microscopic-and pharmacological-based approaches have demonstrated that the actin cytoskeleton dynamically responds to pathogen ***,we observed that phalloidin treatment induced actin nucleation,resulting in enhanced resistance of wheat against the stripe rust pathogen Puccinia striiformis ***(Pst).To define the mechanism underpinning this process,we characterized a family of conserved actin-binding proteins,the actin related protein(ARP)family,which controls actin ***,we identified and characterized a wheat ARPC gene(Ta ARPC5),which encodes a 136-amino acid protein containing a P16-Arc domain,the smallest subunit of the ARP2/3 *** ARPC5 m RNA accumulation was induced following the infection of plants with the avirulent Pst strain,and following the elicitation with flagellin(e.g.,flg22)as *** localization analysis revealed that Ta ARPC5 is primarily localized to the cortical actin cytoskeleton,and its precise cellular localizations suggest the proximity to processes correlated with the actin-organelle *** treatment with virulent Pst,Ta ARPC5-knockdown plants exhibited a significant reduction in the expression of PTI-specific m ***,we observed enhanced induction of reactive oxygen species(ROS)accumulation and a decrease in Ta CAT1 expression following infection with an incompatible Pst *** with yeast complementation assays,the current study demonstrates a role for Ta ARPC5 in resistance signaling in wheat against Pst infection by regulating the host actin cytoskeleton.
