Familial dysautonomia(FD), a hereditary sensory and autonomic neuropathy, is caused by a mutation in the Elongator complex protein 1(ELP1) gene that leads to a tissue-specific reduction of ELP1 protein. Our work to generate a phenotypic mouse model for FD headed to the discovery that homozygous deletion of the mouse Elp1 gene leads to embryonic lethality prior to mid-gestation. Given that FD is caused by a reduction, not loss, of ELP1, we generated two new mouse models by introducing different copy numbers of the human FD ELP1 transgene into the Elp1 knockout mouse(Elp1) and observed that human ELP1 expression rescues embryonic development in a dose-dependent manner. We then conducted a comprehensive transcriptome analysis in mouse embryos to identify genes and pathways whose expression correlates with the amount of ELP1. We found that ELP1 is essential for the expression of genes responsible for nervous system development. Further, gene length analysis of the differentially expressed genes showed that the loss of Elp1 mainly impacts the expression of long genes and that by gradually restoring Elongator, their expression is progressively rescued. Finally, through evaluation of co-expression modules, we identified gene sets with unique expression patterns that depended on ELP1 expression.

Pouteria campechiana leaf extract and its bioactive compound myricitrin are mosquitocidal against Aedes aegypti and Culex quinquefasciatus

Objective: To test the mosquitocidal potential of leaf extracts of Pouteria campechiana prepared with different solvents and elucidate the structure of an isolated mosquitocidal compound. Methods: The leaf extracts of Pouteria campechiana prepared with three solvents(petroleum benzene, ethyl acetate and acetone) and potential bioactive fractions were tested against various stages of Aedes aegypti and Culex quinquefasciatus by using the WHO protocols, and the chemical profile and its functional groups were identified by GC-MS and Fourier transmissioninfrared spectroscopy(FT-IR). The structure of bioactive compound was characterized by nuclear magnetic resonance(NMR) spectral technique. Results: The preliminary phytochemical results revealed the presence of alkaloids, amino acids, flavonoids, quinones, saponins, steroids, tannins, and terpenoids in the acetone extract. A significant toxic potential was observed in the acetone extract against both Aedes aegypti and Culex quinquefasciatus mosquitoes. The acetone extract exhibits remarkable larvicidal(LC50: 12.232 μg/mL and LC90: 63.970 μg/mL), pupicidal(LC50: 18.949 μg/mL and LC90: 167.669 μg/m L) and adulticidal(LC50: 20.689 μg/mL and LC90: 72.881 μg/mL) effects against Aedes aegypti. Furthermore, the same extract was subjected to isolation of bioactive compound by GCMS and FT-IR analysis. GC-MS results showed the presence of 5 major compounds, and octacosane(18.440%) was detected as the predominant compound. The FT-IR result of acetone extract demonstrated the presence of various functional groups like alkanes/alkynes, ester, aromatic and amides. The NMR spectrum results of isolated compound were well matched to glycoside linked flavonoids. Based on the chromatography and spectral techniques the isolate molecule was identified as myricitrin by FT-IR and nuclear magnetic resonance spectral data. In this work,diamond-like carbon(DLC)thin film coatings are deposited on silicon substrates by using plasma-enhanced chemical vapour deposition(PECVD)*** varying the hydrogen(H2)flow rate,CH4−Argon(Ar)flow rate and deposition temperature(Td)as per a Box-Behnken experimental design(BBD),15 DLC deposition experiments are carried *** Young’s modulus(E)and the coefficient of friction(COF)for the DLCs are *** using a second-order polynomial regression approach,two metamodels are built for E and COF,that establish them as functions of H2 flow rate,CH4-Ar flow rate and Td.A non-dominated sorting genetic algorithm(NSGA-III)is used to obtain a set of Pareto solutions for the multi-objective optimization of E maximization and COF *** to various practical scenarios,evaluation based on distance from average solution(EDAS)approach is used to identify the most feasible solutions out of the Pareto solution *** experiments are conducted which shows the efficacy of the polynomial regression—NSGA-III—EDAS hybrid *** surface morphology of the DLCs deposited as per the optimal predictions is also studied by using atomic force microscopy.

This work introduces the facile hydrothermal synthesis of double perovskite La2CuCoO6.X-ray diffraction pattern confirmed the formation of a monoclinic phase with P121/c1 *** electron microscopy results revealed that the self-assembled porous rods were composed of nanocrystallite *** estimated specific surface area of these mesoporous rods with an average pore diameter of 6 nm was^41 m^2·g^–*** presence of ions with oxidation states of La^3+,Cu^2+,and Co^2+/Co^3+on the surface of the mesoporous La2CuCoO6 rods was confirmed by X-ray photoelectron spectroscopic *** cyclicvoltammetry and chronopotentiometry,the electrode fabricated from the mesoporous La2CuCoO6 rods were found to exhibit pseudocapacitive behavior with a specific capacitance of 259.4 F·g^–1 at a current density of 0.5 A·g^–***^89%retention in specific capacitance was achieved after 1000 charge/discharge cycles at a constant current density of4 A·g^–1.

Image smog restoration using oblique gradient profile prior and energy minimization

Removing the smog from digital images is a challenging pre-processing tool in various imaging ***,many smog removal(i.e.,desmogging)models are proposed so far to remove the effect of smog from *** desmogging models are based upon a physical model,it means it requires efficient estimation of transmission map and atmospheric veil from a single smoggy ***,many prior based restoration models are proposed in the literature to estimate the transmission map and an atmospheric ***,these models utilized computationally extensive minimization of an energy ***,the existing restoration models suffer from various issues such as distortion of texture,edges,and ***,in this paper,a convolutional neural network(CNN)is used to estimate the physical attributes of smoggy *** gradient channel prior(OGCP)is utilized to restore the smoggy ***,a dataset of smoggy and sunny images are ***,we have trained CNN to estimate the smog gradient from smoggy ***,based upon the computed smog gradient,OGCP is utilized to restore the still smoggy *** analyses reveal that the proposed CNN-OGCP based desmogging model outperforms the existing desmogging models in terms of various performance metrics.

Effect of Emilia sonchifolia(Linn.)DC on alcohol- induced oxidative stress in pancreas of male albino rats

Objective:To explore the efficacy of n-hexane extract of Emilia sonchifolia(***) against ethanol induced pancreatic dysfunction in the young Wistar albino ***:The rats were divided into four *** rats in group I received distilled water orally,groupⅡreceived oral administration of 20%(w/v) ethanol dissolved in drinking water,groupⅢreceived oral administration of 20%(w/v) ethanol in distilled water+n-hexane extract of *** (250 mg/kg body weight),and groupⅣreceived oral administration of n-hexane extract of ***(250 mg/kg body weight) *** marker enzymes aspartate aminotransferase (AST),alanine aminotransferase(ALT),pancreatic enzymatic antioxidants superoxide dismutase. lipid peroxidation,catalase,glutathione peroxidase,non-enzymatic antioxidants glutathione and vitamin C were measured and ***:Administration of 20%ethanol lor 16 weeks significantly increased the liver marker enzymes AST,ALT(P<0.05).reduced the pancreatic enzymatic antioxidants superoxide *** peroxidation,catalase,glutathione peroxidase,glutathione and vitamin C(P<0.05).Histopathological examination showed that the ethanol provoked the oxidative stress which was demonstrated as pancreatic necrosis and *** administration of n-hexane extract of ***(250 mg/kg body weight) protected the pancreas against the damage induced by ethanol which was confirmed by the histopathological studies and the normalization of biochemical ***: Thus n-hexane extract of *** shows a promise in therapeutic use in alcohol induced oxidative stress.

Objective:To evaluate the protective effect of ethanol extract of Mollugo nudicaulis(***) against perchloroethylene-induced ***:The hepatoprotective activity of the ethanol extract of ***(200 mg/kg body wt) was studied in percholoroethylene (1 000 mg/kg body wt) induced hepatotoxicity in Wistar albino *** serum levels of AST, ALT,ALP,bilirubin and the liver content of SOD,CAT,GPx,GST,CSH,vitamin C were assessed to evaluate the hepatoprotective and antioxidant activities of the *** activity of the extract was compared with silymarin,a standard reference *** addition,serum urea,uric acid and creatinine levels were measured to evaluate the kidney *** histopathological examination of the liver tissues was observed to support the biochemical ***: The results revealed that the extract significantly(P<0.05) restored the serum levels of AST,ALT, ALP,bilinibin and significantly(P<0.05) increased the antioxidant enzymes SOD,CAT,GPx,GST, GSH,vitamin C in perchloroethylene-induced rats to its *** biochemical observations were suppoited by the histopathological studies of the liver ***:The results led to the conclusion that *** possess hepatoprotective and antioxidant activites against perchloroethylene-induced hepatotoxicity in rats.

Effective field theory approach to lepto-philic self-conjugate dark matter

We study self-conjugate dark matter(DM)particles interacting primarily with Standard Model(SM)leptons in an effective field theoretical *** consider SM gauge-invariant effective contact interactions between Majorana fermion,real scalar and real vector DM with leptons by evaluating the Wilson coefficients appropriate for interaction terms up to dimension 8,and obtain constraints on the parameters of the theory from the observed relic density,indirect detection observations and from the DM-electron scattering cross-sections in direct detection *** energy LEP data has been used to study sensitivity in the pair production of low mass(≤80 GeV)DM *** production of DM particles of mass≥50 GeV in association with mono-photons at the proposed ILC has rich potential to probe such effective operators.

Effect of processing parameters on the electromagnetic radiation emission during plastic deformation and crack propagation in copper-zinc alloys

This paper presents some investigations on the effect of processing parameters on the emission of electromagnetic radiation （EMR） during plastic deformation and crack propagation in copper-zinc alloys. Timing of the EMR emissions, maximum stress during crack instability, stress-intensity factor, elastic strain energy release rate, maximum EMR amplitude, RMS value of EMR amplitude, EMR frequency and electromagnetic energy release rate were analysed for the effect of rolling directions at different percentage of zinc content in Cu-Zn alloy specimens. The same parameters were also analysed for 68-32 Cu-Zn alloy specimens at different annealing temperatures and at different angles 0, to the rolling direction. EMR emissions are observed to be highly anisotropic in nature. At θ=45° to 60°, marked changes in mechanical and electromagnetic parameters were observed. Specimens annealed at 500℃, just above the recrystallization temperature, and at 700℃, when grain-size growth is rapid, EMR responses have been found to have well-defined patterns.
