Effect of recombinant and native buffalo OVGP1 on sperm functions and in vitro embryo development: a comparative study

Background： An oviduct-specific glycoprotein, OVGP1, is synthesized and secreted by non-ciliated epithelial cells of the mammalian oviduct which provides an essential milieu for reproductive functions. The present study reports the effects of recombinant buffalo OVGP1 that lacks post-translational modifications, and native Buffalo OVGP1 isolated from oviductal tissue, on frozen-thawed sperm functions and in vitro embryo ***： The proportion of viable sperms was greater（P 〈 0.05） in the recombinant OVGP1-treated group compared to the native OVGP1-treated group at 2 h, 3 h, and 4 h of incubation. The proportion of motile sperms at3 h and 4 h of incubation; and membrane-intact sperms at 4 h was greater（P 〈 0.05） in the native OVGP1-treated group compared to the control and recombinant OVGP1-treated groups. The proportion of capacitated and acrosome-reacted sperms was greater（P 〈 0.05） in the native OVGP1-treated group compared to the recombinant OVGP1 group at 4 h. The rates of cleavage of embryos and their development to the blastocyst stage were greater（P 〈 0.05） in the presence of either native or recombinant OVGP1 in comparison to control at 10 μg/mL concentration as compared to 5 or 20 μg/***： The study suggests that both native and recombinant OVGP1 impart a positive effect on various sperm features and in vitro embryo development. However, native OVGP1 was found to have a more pronounced effect in comparison to recombinant non-glycosylated OVGP1 on various sperm functions except viability. Hence,our current findings infer that glycosylation of OVGP1 might be essential in sustaining the sperm functions but not the in vitro embryo development.

Low sperm numbers in artificial insemination (AI)-doses are being used widely to make the best use of high genetic value bulls as well as sex-sorted semen. Sperm concentration needed for AI to obtain reasonable fertility, taking genetic value of bull and numerous others components into consideration is one of the essential constituents for successful AI breeding program. However, low sperm concentrations in AI-doses lead to reducing post-thaw viability. The reduction in viability of low sperm doses may be affected by fresh semen volume, sperm number and seminal plasma level at final dilution. Reduction in quality and fertility of low sperm doses is one of the limitations for their use in successful AI programme. Sperm number per AI required to achieve optimum fertility is one of the main crucial things to AI industry, and numerous efforts have been made in this regard. Due to great variability among bulls, sperm number per AI could be a limiting factor in achieving acceptable fertility values. Fertility of low sperm doses may vary among bulls, and non-return rates (NRRs) with low sperm doses may be determined by fertility level of bull. On the basis of individual bulls, sperm numbers in AI doses needed to be adjusted to reduce the variations in NRRs among bulls. Utilizing high fertile bulls for low sperm doses with acceptable non-return rates (NRRs) may be a way to cover a large number of bovines under AI in countries like India. Deposition site within the uterine horn may alter non return rates following inseminations with low sperm doses. Following deep-uterine inseminations, acceptable pregnancies may be achieved with low sperm doses and even if ovulation side is unknown.

Post-urethroplasty complications in hypospadias repair: a systematic review and meta-analysis comparing, polydioxanone and polyglactin sutures

Background Polyglactin(PG)and polydioxanone(PDS)sutures are extensively used based on the surgeon's *** development of post-reconstruction urethrocutaneous fistula(UCF)is variably attributed to the choice of suture material for *** meta-an alysiscompares complications of hypospadia srepair using PGand PDS *** The systematic review and meta-analysis were performed as per the Preferred Reporting Items for Systematic Reviews and Meta-Analyses *** authors conducted thorough searches in databases including MEDLINE,EMBASE,CENTRAL,Scopus,Google Scholar,and clinical trial *** measures included UCF,meatal stenosis,wound infection,urethral stricture,glans dehiscence,and overall *** analysis was used with fixed or random-effect models to find the pooled risk ratio and I2 *** The criteria for inclusion were met by five comparative studies with the inclusion of 1244 children *** analysis failed to show a statistically significant difference in the incidence of meatal stenosis,urethral stricture,wound infection,and total complications using PG and PDS ***,it showed a reduction in the incidence of UCF with PDS suture hypospadias repairs(risk ratio=0.66,95%Cl 0.48 to 0.92).Conclusions PDS sutures are associated with decreased incidence of UCF than PG after hypospadias *** incidence of meatal stenosis,urethral stricture,wound infection,and total complications was not affected by the type of suture material used for *** implications This meta-analysis suggests decreased incidence of UCF when PDS sutures are used for hypospadias repair which may impact the choice of suturematerialforrepair.

Spatiotemporal regulation of gene expression by polycomb repressive complex 2(PRc2)is critical for animal and plant *** Arabidopsis fertilization independent seed(FIS)-PRC2 complex functions specifically during plant reproduction from gametogenesis to seed *** a double fertilization event,triploid endosperm proliferates early,followed by the growth of a diploid embryo,which replaces the endosperm in Arabidopsis and many *** genes critical for endosperm proliferation such as IkkU2 and MINI3 are activated after *** we report that two MADS-box AGAMOUS-LIkE(AGL)proteins associate with the key endosperm proliferation loci and recruit the FIS-PRC2 repressive complex at 4-5 days after pollination(DAP).Interestingly,AGL9 and AGL15 only accumulate toward the end of endosperm proliferation at 4-5 DAP and promote the deposition of H3k27me3 marks at key endosperm proliferation *** of AGL9 and AGL15 or overexpression of AGL9 or AGL15 significantly influence endosperm proliferation and ***-wide analysis with cleavage Under Targets and tagmentation(CUT&Tag)sequencing and RNA sequencing revealed the landscape of endosperm H3k27me3 marks and gene expression profiles in Col-0 and agl9 ***&Tag qPCR also demonstrated the occupancy of the two MADS-box proteins and FIS-PRC2 on a few representative target *** studies suggest that MADS-box proteins could potentially recruit PRc2 to regulate many other developmental processes in plants or even in fungi and animals.

Objective:To evaluate the antimalarial activity of noscapine against Plasmodium falciparum 3D7 strain(Pf3D7),its clinical isolate(Pf140/SS),and Plasmodium berghei ANkA(PbA).Methods:Using ring-stage survival assay,phenotypic assessments,and SYBR-green-based fluorescence assay,the antimalarial activities of noscapine were assessed compared with dihydroartemisinin(DHA)in in vivo and in vitro *** addition,hemolysis and cytotoxicity tests were carried out to evaluate its ***-PCR assay was also conducted to determine the effect of noscapine on papain-like cysteine protease Plasmodium falciparum falcipain-2(PfFP-2).Results:The antimalarial efficacy of noscapine against Pf3D7 and Pf140/SS was comparable to DHA,with IC50 values of(7.68±0.88)and(5.57±0.74)nM/mL,respectively,and>95%inhibition of PbA infected *** also showed a safe profile,as evidenced by low hemolysis and cytotoxicity even at high ***,PfFP-2 expression was significantly inhibited in both noscapine-treated Pf3D7 and Pf140/SS(P<0.01).Conclusions:Noscapine has antimalarial properties comparable to standard antimalarial DHA with better safety profiles,which may be further explored as a therapeutic candidate for the treatment of malaria.

Effect of promoters on dynamics of gas-solid fluidized bed——Statistical and ANN approaches

In this study, a bubbling fluidized bed column, 99 mm in inside diameter and 960 mm in height, was used to investigate the effect of rod and disc promoters on fluctuation and expansion ratios. Factorial design （statistical approach） and artificial neural network （ANN） models were developed to predict the fluctuation and expansion ratios in this gas-solid fluidized bed with varying gas flow rates, bed heights, particle sizes and densities. The fluctuation and expansion predicted using these statistical and ANN models, for beds with and without promoters, were found to agree well with corresponding experiments. The statistical model was found to be superior to the ANN model due to its ability to take into account both individual and interactive effects. The rod promoters were found to be more effective in reducing bed fluctuation, and in increasing bed expansion at high gas mass velocities.

Synthesis and Characterization of Bio-based Epoxy Blends from Renewable Resource Based Epoxidized Soybean Oil as Reactive Diluent

A novel bioresin, epoxidized soybean oil was synthesized by in situ method and was characterized employing FTIR and NMR. The bioresin was blended with epoxy（DGEBA） at different ratios as reactive diluents for improved processibility and toughened nature. The composition with 20 wt% bioresin exhibited improved impact strength to the tune of 60% as compared to virgin epoxy. Fracture toughness parameters critical stress intensity factor（kIC） and critical strain energy release rate（GIC） were evaluated using single edge notch bending test and demonstrated superior enhancement in toughness. Dynamic mechanical, thermal, thermo mechanical and fracture morphological analyses have been studied for bio-based epoxy blends. Curing kinetics has been evaluated through DSC analysis to investigate the effect of bioresin on cross-linking reaction of neat epoxy with triethylenetetramine as curing agent.

C-H bond can undergo vicarious Michael addition reaction （VMA） with doubleactivated double bond in the absence of strong base and catalyst under mild conditions. Intramolecular H-bonding, electron-withdrawing inductive effect, and steric hindrance at aposition of nucleophile facilitates C-H addition over N-H addition. By using VMA, high branching multiplicity, novel branching pattern, controllable density and distribution of functional groups can be envisioned for novel dendrimer synthesis.

This paper projects an isolated hybrid model of Offshore wind-diesel-tidal turbine and discusses the stability and reactive power management issue of the whole *** hybrid system often loses its stability as it becomes prone to uncertain load and input parameters and therefore the necessity of Reactive power management becomes *** overall stability of the hybrid offshore wind-diesel-tidal turbine is made possible by the management of reactive power in the hybrid system through the application of FACTS *** therefore the dynamic hybrid model of the DFIG and DDPMSG based offshore wind-diesel-tidal turbine is analysed for stability with different input parameters like wind and tidal *** detailed modelling and simulation,a small signal model of the whole hybrid system is designed and reactive power management of the system is achieved by the incorporation of a STATCOM *** improvement of stability and reactive power compensation of the hybrid system,GA and PSO optimised STATCOM controller is used.
