In this study, we investigate the potential for oral Lactobacilli (Lb) to afford innate protection against nasopharyngeal coloniser Neisseria meningitidis serogroup b (Nmb), which causes the bulk of UK meningococcal disease. Oral isolates of L. plantarum, L. salivarious, L. casei, L. rhamnosus, L. gasseri and gut probiotic L. rhamnosus GG were assessed for their ability to suppress nasopharyngeal epithelial inflammatory responses to pathogenic Nmb. The specificity of attenuation was examined using TLR 2 ligand, Pam3Cys, and early response cytokine IL1β;and the mechanism of attenuation was explored using heat-killed organisms and conditioned medium. Pro-inflammatory IL-6 and TNFα cytokine secretion was quantified by ELISA and associated cell death was quantified by PI staining and LDH release. Nmb adhesion, invasion and metabolism were determined using standard gentamicin protection with viable counts, and bioluminescence, respectively. L. plantarum and L. salivarious suppressed IL-6 and TNFα secretions from Nmb-infected epithelial cells. Lb did not need to be alive and could suppress using secretions, which were independent of TLR2 or IL1β receptor signalling. L. plantarum, in particular, reduced Nmb-induced necrotic cell death of epithelial monolayers. Like L. salivarious, it significantly inhibited Nmb adhesion but uniquely L. plantarum abolished Nmb invasion. Using bioluminescence as a reporter of pathogen metabolism, L. plantarum and its secretions were found to inhibit Nmb metabolism during cell invasion assays. We conclude that oral L. plantarum and its secretions could be used to help reduce the burden of meningococcal disease by removing the intracellular nasopharyngeal reservoir of Nmb.

Water resources face many pressures and stresses around the *** the vulnerability of water resources is,therefore,increasingly an essential practice in water resources *** studies of water resources vulnerability have been carried out around the world during the last three ***,the main objective of this study was to analyze water resources vulnerability assessment tools and identify the methodological trend of water resources vulnerability assessment at the local level in existing *** achieve this objective,the Preferred Reporting Items for Systematic Reviews and Meta-Analyses(PRISMA)method has been applied to include or exclude *** on water vulnerability numbering 374 were preselected in the ScienceDirect and the Google Scholar *** 32 out of the 374 articles met the inclusion criteria defined for the *** results show that China(21.9%),U.S.A.(9.4%)and Canada(6.3%)are the most active countries publishing water resources vulnerability *** 191 vulnerability indicators from the 32 articles were classified into five categories of factors:physical factors(33.5%),socio-economic factors(28.3%),environmental or eco-environmental factors(25.1%),institutions and governance factors(7.3%)and infrastructure factors(5.8%).This study shows that almost half of the studies(47%)considered all five categories of factors,9.4%considered four categories of factors,6.3%considered three categories of factors,28%used two types of factors and 9.4%considered only one *** have used several methods to assess water resources vulnerability:overlay and index methods(59.4%),process-based models(12.5%),statistical methods(9.4%),overlay and index methods plus process-based model(9.4%),overlay and index methods plus statistical methods(6.3%)and statistical methods plus process-based models(3.1%).Geographic information system(GIS)is an important tool in assessing the vulnerability of water resou

bACKGROUND Para-aortic lymph nodes(PALN)are found in the aortocaval groove and they are staged as metastatic disease if involved by pancreatic ductal adenocarcinoma(PDAC).The data in the literature is conflicting with some studies having associated PALN involvement with poor prognosis,while others not sharing the same *** resection is not included in the standard lymphadenectomy during pancreatic resections as per the International Study Group for Pancreatic Surgery and there is no consensus on the management of these *** To investigate the prognostic significance of PALN metastases on the oncological outcomes after resection for *** This is a retrospective cohort study of data retrieved from a prospectively maintained database on consecutive patients undergoing pancreatectomies for PDAC where PALN was sampled between 2011 and *** comparison of the data between PALN+and PALN-subgroups,survival analysis with the Kaplan-Meier method and risk analysis with univariable and multivariable time to event Cox regression analysis were performed,specifically assessing oncological outcomes such as median overall survival(OS)and disease-free survival(DFS).RESULTS 81 cases had PALN sampling and 17(21%)were *** N stage was significantly different between PALN+and PALN-patients(P=0.005),while no difference was observed in any of the other *** imaging diagnosed PALN positivity in one *** and DFS were comparable between PALN+and PALN-patients with lymph node positive disease(OS:13.2 mo vs 18.8 mo,P=0.161;DFS:13 mo vs 16.4 mo,P=0.179).No difference in OS or DFS was identified between PALN positive and negative patients when they received chemotherapy either in the neoadjuvant or in the adjuvant setting(OS:23.4 mo vs 20.6 mo,P=0.192;DFS:23.9 mo vs 20.5 mo,P=0.718).On the contrary,when patients did not receive chemotherapy,PALN disease had substantially shorter OS(5.5 mo vs 14.2 mo;P=0.015)and DFS(4.4 mo

bayesian optimization(bO)has been leveraged for guiding autonomous and high-throughput experiments in materials ***,few have evaluated the efficiency of bO across a broad range of experimental materials *** this work,we quantify the performance of bO with a collection of surrogate model and acquisition function pairs across five diverse experimental materials *** defining acceleration and enhancement metrics for materials optimization objectives,we find that surrogate models such as Gaussian Process(GP)with anisotropic kernels and Random Forest(RF)have comparable performance in bO,and both outperform the commonly used GP with isotropic *** with anisotropic kernels has demonstrated the most robustness,yet RF is a close alternative and warrants more consideration because it is free from distribution assumptions,has smaller time complexity,and requires less effort in initial hyperparameter *** also raise awareness about the benefits of using GP with anisotropic kernels in future materials optimization campaigns.

目的　了解健康儿童肺炎链球菌携带株的流行病学特征。方法　测定来自上海 5所幼儿园 (代号分别为JR、PL、SY、WS、ZF) 791名健康儿童鼻咽部分离的 2 2 2株肺炎链球菌健康儿童携带株的耐药谱和血清型 ,并以PFGE、bOXPCR和pbp基因指纹等分子生物学方法分析菌株间亲缘关系。结果　 2 2 2株携带株中有青霉素低度耐药株 32株 ,未发现青霉素高度耐药株。全部菌株包括 2 2种血清型 ,常见为 2 3F(2 5 7% )、6A(13 1% )、19F(10 8% )、6b(8 1% )等 ;青霉素低度耐药株包括 8种血清型 ,主要为 2 3F(5 0 % )、19A和 19F(各占 15 6 % )。 32株青霉素低度耐药菌株可分为 8种bOXPCR谱型和 7种PFGE谱型。PFGE谱型A菌株的耐药谱、血清型以及bOXPCR和pbp基因指纹高度一致。自幼儿园SY的儿童分离的肺炎链球菌青霉素耐药率 31 8% ,远高于平均水平 ,该园 2 1株青霉素耐药菌株中 2 0株属于PFGE谱型A或b。结论　上海 5所幼儿园健康儿童肺炎链球菌携带株青霉素耐药率总体尚较低 ,但个别幼儿园具有较高耐药率。
