Pharmacokinetics and Bioequivalence of Dienogest in Healthy Bangladeshi Female Volunteers: An Open-Label, Single-Dose, Randomized, Two-Way Crossover Study

Background: Dienogest is a potential treatment for pelvic pain associated with endometriosis, a condition of significant concern in gynaecology. The current study was conducted as a crossover-randomized bioequivalence assessment of two oral Dienogest 2 mg formulations, aiming to provide valuable insights for healthcare professionals and researchers in this field. Objective: The primary aim of this research was to evaluate and compare the pharmacokinetic characteristics of Dienogest 2 mg tablets. Dinogest (Dienogest 2 mg) tablets, manufactured by Nuvista Pharma Limited in Bangladesh, and Visanne (Dienogest 2 mg) tablets, manufactured by Bayer Pharma in Germany, were the test and reference formulations, respectively. Materials and Method: The study was an open-label, balanced, randomized, two treatments, two sequences, two periods, two-way crossover, laboratory blind, single oral dose bioequivalence study conducted in healthy adult females under fasting conditions. The study was carried out on 13 healthy, non-pregnant female subjects, and all the subjects completed both study periods with a 15-day washout in between. Randomization was used to assign the test and reference formulations to the subjects. Following each oral administration, a series of blood samples were obtained at different time intervals from pre-dose to 72 hours post-dose and analyzed for Dienogest concentrations using a validated bio-analytical method. A standard non-compartmental model was used to analyze the pharmacokinetic parameters. The primary pharmacokinetic parameters were peak plasma drug concentration (Cmax), the area under the plasma concentration-time curve from time zero to time t (AUC0–t), and AUC from t = 0 to infinity (AUC0–∞). Colorectal cancer(CRC)is a predominant life-threatening cancer,with liver and peritoneal metastases as the primary causes of *** inflammation,a known CRC risk factor,nurtures a local inflammatory environment enriched with tumor cells,endothelial cells,immune cells,cancer-associated fibroblasts,immunosuppressive cells,and secretory growth *** complex interactions of aberrantly expressed cytokines,chemokines,growth factors,and matrix-remodeling enzymes promote CRC pathogenesis and evoke systemic responses that affect disease *** evidence suggests that these cytokines and chemokines play a role in the progression of CRC through immunosuppression and modulation of the tumor microenvironment,which is partly achieved by the recruitment of immunosuppressive *** cells impart features such as cancer stem cell-like properties,drug resistance,invasion,and formation of the premetastatic niche in distant organs,promoting metastasis and aggressive CRC growth.A deeper understanding of the cytokineand chemokine-mediated signaling networks that link tumor progression and metastasis will provide insights into the mechanistic details of disease aggressiveness and facilitate the development of novel therapeutics for ***,we summarized the current knowledge of cytokine-and chemokine-mediated crosstalk in the inflammatory tumor microenvironment,which drives immunosuppression,resistance to therapeutics,and metastasis during CRC *** also outlined the potential of this crosstalk as a novel therapeutic target for *** major cytokine/chemokine pathways involved in cancer immunotherapy are also discussed in this review.

Study of markers of atherosclerosis including insulin resistance in patients with chronic renal failure

Objectives: The present study was designed to assess the markers of atherosclerosis including Insulin resistance (IR) in na?ve patients with chronic renal failure (CRF). Methods: Eighty consecutive na?ve patients with CRF were taken up for study. They were divided into non-diabetic group, Group A (N = 50) and diabetic group, Group B (N = 30). Twenty healthy individuals were taken as controls, Group C (N = 20). Patients undergoing renal replacement therapy, having chronic liver disease and with concomitant acute or chronic infection were excluded from the study. Routine hemogram, fasting plasma glucose, fasting serum insulin ,lipid profile, renal function tests, C-reactive protein (CRP), serum uric acid levels, ultrasound of abdomen and ultrasound B scan for carotid intima medial thickness (CIMT) were done for all patients and control. The data collected were analyzed to inter-correlate the parameters using SPSS 16. Results: On comparing parameters between the three groups, values of HOMA-IR, CRP, uric acid, VLDL and CIMT were significantly higher in Groups A and B than Group C whereas values of HOMA-B, HDL and LDL were lower in both groups A and Bas compared to group C. HOMA-IR had significant negative correlation with creatinine clearance (Crcl) (r = -0.449, p = 0.01) in Group A and (r = -0.483, p = 0.007) in Group B. HOMA-IR had significant positive correlation with CIMT (r = 0.413, p = 0.03) in Group A and (r = 0.581, p = 0.001) in Group B. Crcl had significant negative correlation with CIMT(r = -0.375, p = 0.007) in Group A and (r = -0.705, p = 0.001) in Group B. Crclnegatively correlated with C-reactive protein (r = -0.460, p = 0.001) in Group A and (r = 0.431, p = 0.01) in Group B. HOMA-B positively correlated with Crcl (r = 0.667, p = 0.001) and also with CIMT (r = -0.531, p = 0.003) among Group B individuals. Water electrolysis is considered as the most capable and old technology for hydrogen fuel preparation. Electrolysis needs external electrical energy through electrodes to split water into hydrogen and oxygen. An efficient electrolysis requires suitable electrodes to minimize potential drop. In this study Aluminium and Copper Coated Aluminium were used as different combination of Anodes and Cathodes to find out more efficient electrodes combination. NaCl solution in rain water was taken as electrolyte. Rain water was taken to avoid ionic impedance of tap water and expenses of distilled water. In this study, the most efficient electrode combination was Copper Coated Aluminium (anode)-Aluminium (cathode) and gave the highest efficiency of hydrogen production to about 11% at normal temperature for very low concentration of NaCl (0.051 M) which increased with temperature, up to 29% upon rising of temp to 60°C. It was showed that higher concentration of electrolyte would surge the efficiency significantly. If the supplied heat could be provided from any waste heat sources then this study would be more efficient. Effect of Fiber Loading on the Dynamic Mechanical Properties of Jute Fiber Reinforced Polypropylene Composites

Jute fiber (woven fabric, 1 × 1 plain weave) reinforced polypropylene matrix composites were prepared by compression molding with various fiber loading such as 30%, 40%, 46%, 50%, 55% by weight. The mechanical properties such as tensile strength (TS), bending strength (BS), tensile modulus (TM), bending modulus (BM) and impact strength (IS) of the composite were assessed and analyzed. The highest value of TS, BS, TM, BM and IS were 68.1 MPa, 94.1 MPa, 2936 MPa, 4831 MPa and 14.5 kJ/m2 respectively with 50% fiber loading by weight. It was found that the mechanical properties of the composites were increased with the increase in jute fiber content up to 50% by weight;however, further increase in fiber loading the value decreased. On the basis of fiber content, 50% fiber reinforced composites had the optimum set of mechanical properties. Initially the water absorption rate was higher and then it became slower and static with time. Chemical ageing test with various chemical media such as H2O2, NaOH, HCl and NaCl were performed up to 168 hours. After first 24 hours the composite samples showed gradual weight gain (%) and then the weight gain was become slow and steady in the chemical solution.
