Lumine.ce.t organic radicals have.garne.e. incre.sing atte.tion owing to the.r ve.satile.applications in se.sing, imaging, and organic light-e.itting diode.(OLe.s), attribute. to the.r unique.e.ission prope.tie. originating from the.double. spin ***, the.natural narrow bandgap of organic fre. radicals typically limits the.r e.ission to the.long-wave.e.gth *** lumine.ce.t organic radicals with short-wave.e.gth e.ission re.ains a significant challe.ge. He.e.n, a se.ie. of carbon-ce.te.e. radicals with short-wave.e.gth e.ission(383–476 nm) by combining N-he.e.ocyclic carbe.e. with various polycyclic aromatic hydrocarbons(PAHs)(2-naphthyl, 2a~Ⅰ and 2b~Ⅰ;2-phe.anthryl, 2a~Ⅱand 2b~Ⅱ;2-anthryl, 2a~Ⅲand 2b~Ⅲ;3-phe.anthryl, 2a~Ⅳand 2b~Ⅳ). The.re.ical calculations re.e.l that the.introduction of PAHs significantly incre.se. the.Δe.(D2-D1) in 2a^(Ⅰ–Ⅲ)and 2b^(Ⅰ–Ⅲ)compare. to that in phe.yl-de.ive. radical conge.e.s. Conse.ue.tly, the.inte.nal transition from D2 to D1 is impe.e., le.ding to a high yie.d of D2 e.ission and a suppre.se. Kasha's rule. the.e.y ove.coming the.limitations impose. by the.r narrow bandgap. Syndactyly type.V (SDTY5) is an autosomal dominant e.tre.ity malformation characte.ize. by fusion of the.fourth and fifthme.acarpals. In the.pre.ious publication, we first ide.tifie. a he.e.ozygous misse.se.mutation Q50R in home.box domain (HD) ofHOXD13 in a large.Chine.e.family with SDTY5. In orde. to substantiate.the.pathoge.icity of the.variant and e.ucidate.the.unde.lyingpathoge.ic me.hanism causing limb malformation, transcription-activator-like.e.fe.tor nucle.se. (TALe.) was e.ploye. toge.e.ate.a Hoxd13Q50R mutant mouse. The.mutant mice.e.hibite. obvious limb malformations including slight brachydactyly andpartial syndactyly be.wee. digits 2-4 in the.he.e.ozygote., and se.e.e.syndactyly, brachydactyly and polydactyly in *** on BMP2 and SHH/GRe.1/Ae.-FGF e.ithe.ial me.e.chymal (e.m) fe.dback, a crucial signal pathway for limbde.e.opme.t, we found the.e.topically e.pre.se. Shh, Gre.1 and Fgf8 and down-re.ulate. Bmp2 in the.e.bryonic limb bud ate.0.5 to e.2.5. A transcriptome.se.ue.cing analysis was conducte. on limb buds (LBs) at e.1.5, re.e.ling 31 ge.e. that e.hibite.notable.disparitie. in mRNA le.e. be.wee. the.Hoxd13Q50R homozygote. and the.wild-type. The.e.ge.e. are.known to be.involve.in various proce.se. such as limb de.e.opme.t, ce.l prolife.ation, migration, and apoptosis. Our findings indicate.that the.e.topice.pre.sion of Shh and Fgf8, in conjunction with the.down-re.ulation of Bmp2, re.ults in a failure.of patte.ning along both the.nte.ior-poste.ior and proximal-distal axe., as well as a de.re.se.in inte.digital programme. ce.l de.th (PCD). This cascade.ltimate.y le.ds to the.de.e.opme.t of syndactyly and brachydactyly in he.e.ozygous mice. and se.e.e.limb malformations inhomozygous mice. The.e.findings sugge.t that abnormal e.pre.sion of SHH, FGF8, and BMP2 induce. by HOXD13Q50R may be.e.ponsible.for the.manife.tation of human SDTY5.
