This study presents the development and evaluation of a poly(3-hydroxybutyrate-co-4-hydroxybutyrate)(P34HB)ultrafine fiber slow-release system for in vivo osteogenic induction of human umbilical cord mesenchymal stem cells(HUCMSCs).Utilizing dual-nozzle and cell electrospinning techniques,the system encapsulates L-ascorbic acid-2-phosphate magnesium(AsP),βB-glycerophosphate sodium and dexamethasone(DEX)within the'fbers,ensuring sustained osteogenic *** scaffold's morphology,characterization,hydrophilicity,mechanical properties and cellular behavior were *** subcutaneous implantation in rabbits was con-ducted to observe its ectopic osteogenic induction *** fabricated P34HB ultrafine fiber slow-release *** ization *** distribution of HUCMSCs and inducing components within the scaffold,with no chemical reactions affecting the active *** vitro tests showcased a prolonged release of DEX and ASP,while biocompatibility assays highlighted the scaffold's suitability for cellular *** red,type I collagen,and osteopontin(OPN)staining verifed the scaffold's potent osteogenic induction effect on ***,immediate implantation into New Zealand White rabbits led to significant new bone formation within 8 *** findings underscore the system's potential for immediate in vivo implantation without prior in vitro induction,marking a promising advancement in bone tissue engineering.

Molecular mechanism of Xuebijing in treating pyogenic liver abscess complicated with sepsis

BACKgroUND:Xuebijing(XBJ)can alleviate the inflammatory response,improve organ function,and shorten the intensive care unit(ICU)stay in patients with pyogenic liver abscess(PLA)complicated with sepsis,but the molecular mechanisms have not been *** study aimed to explore the molecular mechanism of XBJ in treating PLA complicated with sepsis using a network pharmacology ***:The active ingredients and targets of XBJ were retrieved from the ETCM *** targets related to PLA and sepsis were retrieved from the GeneCards,PharmGKB,DisGenet,Online Mendelian Inheritance in Man(OMIM),Therapeutic Targets Database(TTD),and DrugBank *** targets of PLA complicated with sepsis were mapped to the targets of XBJ to identify potential treatment ***-protein interaction networks were analyzed using the STrING *** treatment targets were imported into the Metascape platform for Gene Ontology(GO)functional enrichment and Kyoto encyclopedia of Genes and Genomes(KEGG)pathway enrichment *** docking was performed to validate the interactions between active ingredients and core ***:XBJ was found to have 54 potential treatment targets for PLA complicated with ***-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor(TNF)were identifi ed as core *** enrichment analysis revealed important pathways,including the interleukin-17(IL-17)signaling pathway,the TNF signaling pathway,the nuclear factor-kappa B(NF-κB)signaling pathway,and the Toll-like receptor(TLr)signaling *** docking experiments indicated stable binding between XBJ active ingredients and core ***:XBJ may exert therapeutic eff ects on PLA complicated with sepsis by modulating signaling pathways,such as the IL-17,TNF,NF-κB,and TLr pathways,and targeting IL-1β,IL-6,and TNF.

Diabetic osteoporosis(DOP)is a significant complication that poses continuous threat to the bone health of patients with diabetes;however,currently,there are no effective treatment *** patients with diabetes,the increased levels of ferroptosis affect the osteogenic commitment and differentiation of bone mesenchymal stem cells(BMSCs),leading to significant skeletal *** address this issue,we aimed to target ferroptosis and propose a novel therapeutic approach for the treatment of *** synthesized ferroptosis-suppressing nanoparticles,which could deliver curcumin,a natural compound,to the bone marrow using tetrahedral framework nucleic acid(tFNA).This delivery system demonstrated excellent curcumin bioavailability and stability,as well as synergistic properties with *** in vitro and in vivo experiments revealed that nanoparticles could enhance mitochondrial function by activating the nuclear factor E2-related factor 2(NrF2)/glutathione peroxidase 4(GPX4)pathway,inhibiting ferroptosis,promoting the osteogenic differentiation of BMSCs in the diabetic microenvironment,reducing trabecular loss,and increasing bone *** findings suggest that curcumin-containing DNA tetrahedron-based ferroptosissuppressing nanoparticles have a promising potential for the treatment of DOP and other ferroptosis-related diseases.

The medium-temperature dependence of jet transport coefficient in high-energy nucleus-nucleus collisions

The medium-temperature T dependence of the jet transport coefficient̂q was studied via the nuclear modification factor rAA(p_(T))and elliptical flow parameter v_(2)(p_(T))for large transverse momentum p_(T) hadrons in high-energy nucleus-nucleus *** a next-to-leading-order perturbative QCD parton model for hard scatterings with modified fragmentation functions due to jet quenching controlled by q,we check the suppression and azimuthal anisotropy for large p_(T) hadrons,and extract q by global fits to rAA(pT)and v_(2)(pT)data in A+A collisions at rHIC and LHC,*** numerical results from the best fits show that q∕T^(3) goes down with local medium-temperature T in the parton jet *** with the case of a constant q∕T^(3),the going-down T dependence of q∕T^(3) makes a hard parton jet to lose more energy near T_(c) and therefore strengthens the azimuthal anisotropy for large pT *** a result,v_(2)(p_(T))for large pT hadrons was enhanced by approximately 10%to better fit the data at rHIC/*** the first-order phase transition from QGP to the hadron phase and the additional energy loss in the hadron phase,v_(2)(p_(T))is again enhanced by 5-10%at rHIC/LHC.

Troy: Efficient Service Deployment for Windows Systems

The modern university computer lab and kindergarden through 12th grade classrooms require a centralized solution to efficiently manage a large number of desktops. The existing solutions either bring virtualization overhead in runtime or requires loading a large image over 30 GB leading to an unacceptable network latency. In this work, we propose Troy which takes advantage of the differencing virtual hard disk techniques in Windows *** such, Troy only loads the modifications made on one machine to all other machines. Troy consists of two modules that are responsible to generate an initial image and merge a differencing image with its parent image, respectively. Specifically, we identify the key fields in the virtual hard disk image that links the differencing image and the parent image and find the modified blocks in the differencing images that should be used to replace the blocks in the parent image. We further design a lazy copy solution to reduce the I/O burden in image merging. We have implemented Troy on bare metal machines. The evaluation results show that the performance of Troy is comparable to the native implementation in Windows, without requiring the Windows environment.

The OsCLV2s-OsCrN1 co-receptor regulates grain shape in rice

The highly conserved CLV–WUS negative feedback pathway plays a decisive role in regulating stem cell maintenance in shoot and floral meristems in higher plants,including Arabidopsis,rice,maize,and ***,we find significant natural variations in the OsCLV2c,OsCLV2d,and OsCrN1 loci in a genome-wide association study of grain shape in ***2a,OsCLV2c,OsCLV2d,and OsCrN1 negatively regulate grain length–width ratio and show distinctive geographical distribution,indica–japonica differentiation,and artificial selection ***,OsCLV2a and OsCrN1 interact biochemically and genetically,suggesting that the two components function in a complex to regulate grain shape of ***,the genetic contributions of the haplotypes combining OsCLV2a,OsCLV2c,and OsCrN1 are significantly higher than those of each single gene alone in controlling key yield *** findings identify two groups of receptor-like kinases that may function as distinct co-receptors to control grain size in rice,thereby revealing a previously unrecognized role of the CLV class genes in regulating seed development and proposing a framework to understand the molecular mechanisms of the CLV–WUS pathway in rice and other crops.

Actin fibers are an important part of the cytoskeleton, providing vital support for the plasma membrane. This function is driven by its ATPase (ATP: adenosine triphosphate) activity, i.e., ATP+H2O -> ADP+Pi. This seemingly simple reaction has attracted much attention because the hydrolysis of ATP provides energy to support life processes. However, the reaction mechanism of ATP hydrolysis in actin is not clear. In order to gain deep insights into the functions of actin, it is essential to elucidate the reaction mechanism of the actin ATP hydrolysis. In this paper, we have studied the reaction mechanism of the ATP hydrolysis in actin by the combined quantum mechanical and molecular mechanics (QM/MM) calculations. Our results show that 1) bond cleavage of the P gamma-OS of ATP and bond formation between oxygen of the lytic water and P gamma atoms take place simultaneously, and this is the rate-limiting step of the hydrolysis; 2) the proton on the lytic water transfers to the phosphate to form H2P gamma O4-via one bridge water. The energy barrier of the complete reaction is 17.6 kcal/mol (1 kcal=4.184 kJ), which is in high agreement with the experimental value.

Experimental and numerical simulation study on the erosion behavior of the elbow of gathering pipeline in shale gas field

During the production period of shale gas, proppant particles and rock debris are produced together,which will seriously erode the elbows of gathering pipelines. In response to this problem, this paper takes the elbow of the gathering pipeline in the Changning Shale Gas Field as an example to test the erosion rate and material removal mechanism of the test piece at different angles of the elbow through experiments and compares the four erosion models with the experimental results. Through analysis, it is found that the best prediction model for quartz sand-carbon steel erosion is the Oka model. Based on the Oka model, FLUenT software was used to simulate and analyze the law of erosion of the elbow of the gas gathering pipeline under different gas flow velocities, gas gathering pressure, particle size, length of L1,and bending directions of the elbow. And a spiral pipeline structure is proposed to reduce the erosion rate of the elbow under the same working conditions. The results show that this structure can reduce erosion by 34%.
