Juvenile ferric iron prevents microbiota dysbiosis and colitis in adult rodents

AIM: To assess whether juvenile chronic ferric iron ingestion limit colitis and dysbiosis at adulthood in rats and mice. METHoDS: Two sets of experiments were designed. In the first set, recently weaned mice were either orally administered ferrous (Fe2+) iron salt or ferric (Fe3+) microencapsulated iron for 6 wk. The last week of experiments trinitrobenzene sulfonic acid (TNBS) colitis was induced. In the second set, juvenile rats received the microencapsulated ferric iron for 6 wk and were also submitted to TNBS colitis during the last week of experiments. In both sets of experiments, animals were sacrificed 7 d after TNBS instillation. severity of the inflammation was assessed by scoring macroscopic lesions and quantifying colonic myeloperoxidase (MPo) activity. Alteration of the microflora profile was estimated usingquantitative polymerase chain reaction (qPCR) by measuring the evolution of total caecal microflora, Bacteroidetes, Firmicutes and enterobacteria. RESULTS: neither ferrous nor ferric iron daily exposures at the juvenile period result in any effect in control animals at adulthood although ferrous iron repeated administration in infancy limited weight gain. Ferrous iron was unable to limit the experimental colitis (1.71 ± 0.27 MPo U/mg proteinvs 2.47 ± 0.22 MPo U/mg protein in colitic mice). In contrast, ferric iron significantly prevented the increase of MPo activity (1.64 ± 0.14 MPo U/mg protein) in TNBS-induced colitis. Moreover, this positive effect was observed at both the doses of ferric iron used (75 and 150 mg/kg per day po - 6 wk). In the study we also compared, in both rats and mice, the consequences of chronic repeated low level exposure to ferric iron (75 mg/kg per day po - 6 wk) on TNBS-induced colitis and its related dysbiosis. We confirmed that ferric iron limited the TNBS-induced increase of MPo activity in both the rodent species. Viewed as hidden hunger, anemia is an indicator of poor nutrition and health status, especially for African pre-school children. several actions are being carried out to combat anemia in Africa, without success. The fundamental step to improve this situation is to determine 1) mothers’ knowledge regarding anemia (symptoms/signs and etiologies), and 2) their attitudes towards anemia. The present effort was to determine 1) and 2) on mothers (npan> = 152) with children (1 - pan style="font-family:Verdana;">5 years) through questionnaire. Data showed that the less educated the mother was, the more she gave the wrong definition of anemia. When these mothers have anemic children, 129 go to the hospital, 34 give grenadine (sweet soda), 14 go to the traditional healer, and 6 give the various decoctions. our results showed that less-educated mothers had wrong anemia management, such as attachment to a visual logic that assimilates blood to food with red coloring. The actions must directly involve these mothers and the traditional healers to improve this situation.

Background:The Global Physical Activity Questionnaire(GpaQ)has been used to measure physical activity(pa)and sedentary time in France,but no study has assessed its psychometric *** study aimed to compare the reliability as well as criterion and concurrent validity of the French version of the GpaQ with the French International Physical Activity Questionnaire long form(IpaQ-LF)and use of an accelerometer in a general adult ***:We included 92 participants(students or staff)from the Medicine Campus at the University of Lorraine,Nancy(north-eastern France).The French GpaQ was completed twice,7 days apart,to study test-retest *** IpaQ-LF was used to assess concurrent validity of the GpaQ,and participants wore an accelerometer(ActiGraph GT3X+)for 7 days to study criterion *** as well as concurrent and criterion validity of the GpaQ were tested by the intraclass correlation coefficient(ICC),Spearman correlation coefficient for quantitative variables,and Kappa and Phi coefficients for qualitative *** concurrent and criterion validity of GpaQ were assessed by Bland-Altman ***:The GpaQ showed poor to good reliability(ICC=0.37-0.94;Kappa=0.50-0.62)and concurrent validity(Spearman r=0.41-0.86),but only poor criterion validity(Spearman r=-0.22-0.42).Limits of agreement for the GpaQ and accelerometer were wide,with differences between 286.5 min/week and 601.3 min/***:The French version of the GpaQ provides limited but acceptable reliability and validity for the measurement of pa and sedentary *** may be used for assessing pa and sedentary time in a French adult population.

AIM： To determine factors associated with fibrosis progression in hepatitis C virus （HCV）-infected patients without significant initial pathological lesions. METHoDS： seventy six untreated HCV-infected patients with initially normal liver as defined by a Knodell score ≤ 3, with 2 liver biopsies and detectable HCVRNA were included. Markers of fibrosis progression were assessed. RESULTS： Median duration of infection and time between paired biopsies was 13 （95% CI： 1-28） and 4 （95% CI： 2-16） years respectively. Alaninetransaminase （ALT） activity was normal in 43.4% of cases. 50% demonstrated progression of the necroinflammation and 34% of fibrosis after a median time evolution of 4 years （95% CI： 2-16）. The median difference in the necro-inflammation and fibrosis score between biopsies was low, 1.5 and 0.0 respectively. Univariate analysis showed there was no difference between fibrosis activity or evolution according to genotype or viral load. A higher fibrosis progression （P = 0.03） was observed in patients with body mass index （BMI）〉 25. Fibrosis progression correlated with the time interval between biopsies （P = 0.01）. A significant progression of activity （1.7 vs 0.4, P 〈 0.05） or fibrosis （0.9 vs 0.0, P 〈 0.01） was observed in patients with elevated ALT. There was a significant correlation between activity progression and fibrosis progression（P = 0.003）. Multivariate analysis demonstrated that fibrosis progression was associated with elevated ALT, BMI 〉 25 and the time interval between 2 biopsies. CoNCLUSIoN： There is no fibrosis progression in 66% of patients without significant initial histopathological lesion. Fibrosis progression is associated with elevated ALT and BMI 〉 25.
