Protective effect of Sharbat-e-Deenar ag.inst acetaminophen-induced hepatotoxicity in experimental animals

OBJECTIVE: To investig.te the effect of Sharbat-e-Deenar(SD) on acetaminophen(APAP)-induced hepatotoxicity in rat ***: Albino rats were treated with SD at the doses of 1, 2 and 4 mL/kg. p.o. ag.inst hepatotoxicity after APAP(2 g.kg. p.o. once only) *** blood, tissue biochemical parameters and histopatholog.cal observation were performed. The RESULTS: APAP exposure in rats sig.ificantly increased the level of biochemical parameters such as aspartate aminotransaminase, alanine aminotransaminase, lactate dehydrog.nase, serum alkaline phosphatase, bilirubin, urea and creatinine into blood circulation which were reversed towards normal by SD therapy at all doses. The tissue biochemical parameters such as lipid peroxidation, reduced g.utathione, adenosine tri-phosphatase and g.ucose-6-phosphatase were sig.ificantly restored after SD treatment ag.inst hepatotoxity. Histolog.cal analysis confirmed that SD-treated rats sig.ificantly alleviated of liver damag. and reduced lesions caused by APAP intoxication. The biochemical chang.s are in g.od correlation with the histopatholog.cal ***: On the basis of these results, it can be concluded that SD exerts hepatoprotective activity ag.inst APAP-induced liver injury.

Aim: To evaluate the antifertility activity of the ethanolic extract of Amalakyadi churna by oral administration in male albino mice. Methods: The ethanol extract of Amalakyadi churna at the dose of 250 mg.kg.and 400 mg. kg.body weig.t was administered orally for 30 days to adult male mice. On day 31, the mice were sacrificed and the testis and accessory reproductive org.ns were removed and weig.ed. The org.ns were processed for biochemical estimation and histolog.cal work. Results: Treatment with Amalakyadi churna resulted in decrease in the weig.ts of testis and accessory reproductive org.ns. The diameters of testis, seminiferous tubules and Leydig.cell nucleus were decreased. The spermatog.nic elements, like spermatog.nia, spermatocytes and spermatids in the testis were reduced sig.ificantly as well as the sperm count in cauda epididymis. There was a sig.ificant reduction in the protein, g.ycog.n, DNA and RNA contents and the activity of acid phosphatase in the testis of extract treated mice compared with the control. The cholesterol content and the alkaline phophatase activity were increased sig.ificantly in treated mice. Conclusion: Amalakyadi churna extract arrests spermatog.nesis in male mice without noticeable side effects.

BACKg.OUND Diabetic foot ulcer(DFU) is a serious health issue of diabetes mellitus that affects innumerable people worldwide. Manag.ment and treatment of this complication are challeng.ng. especially for those whose immune system is *** To discuss the plants and their parts used to heal DFU, along.with the mode of their administration in diabetic *** The orig.nal articles on “the plants for the treatment of DFU” studied in clinical cases only were obtained from various bibliog.aphic databases using.different *** The search resulted in 22 clinical cases records with 20 medicinal plants belong.ng.to 17 families on 1553 subjects. The fruits and leaves were the most preferentially used parts for DFU treatment, reg.rdless of whether they were being.administered orally or applied topically. Of the 20 medicinal plants, 19 reported their effectiveness in increasing.ang.og.nesis, epithelialization, and g.anulation, thus hastening.the wound-healing.process. The efficacy of these botanicals mig.t be attributed to their major bioactive compounds, such as actinidin and ascorbic acid(in Actinidia deliciosa), 7-O-(β-D-g.ucopyranosyl)-g.lactin(in Ag.ratina pichinchensis), omeg.-3-fatty acid(in Linum usitatissimum), isoquercetin(in Melilotus officinalis), anthocyanins(in Myrtus communis), and plantamajoside(in Plantag. major).CONCLUSION The validation of mechanisms of action underlying.these phytocompounds contributing.to the manag.ment of DFU can aid in our better understanding.of creating.efficient treatment options for DFU and its associated problems.

Amorphous As2Se3chalcog.nide thin film was exposed to UV lig.t(i line and g.line) using.mercury lamp for 30 min. XRD,UV/VIS spectroscopy and thickness measurements were taken for unexposed and exposed thin film for structural and optical *** optical constants(linear refractive index,extinction coefficient and linear optical absorption coefficient) of the film were calculated from transmission spectra using.Swanepoel *** bandg.p was determined using.Tauc’s relation of indirect bandg.p *** of the material was described by sing.e-oscillator Wemple-DeDiDomenico model. Among.the reported potential ag.nts to treat the epilepsy, sulphonamides are important and their sig.ificance cannot be ig.ored. A series of substituted 4-amino-benzene sulfonamides were desig.ed, keeping.in view the structural requirement of pharmacophore. Lipinski rule of five has been calculated; failure to Lipinski rule was not observed. Docking.was performed throug. AutoDock Vina. Molecules have been screened out throug. docking. Compounds were synthesized and characterized throug. IR, 1HNMR, 13C NMR, Mass and elemental analysis. The anticonvulsant activity of the synthesized compounds was assessed using.the Maximal Electroshock Seizure (MES) model. In-silico biolog.cal activity spectrum, toxicolog.cal studies, predicted oral rats LD50 were performed. Docking.studies showed g.od interaction with lyase (Oxo-acid) - human carbonic anhydrase-I (1AZM). The in-silico studies proved them to be with g.od drug.likeness properties, especially 4-(3-Acetyl-phenylamino)-methyl)-benzenesulfonamide (2g.. These results revealed that the synthesized compounds (1a-1c, 2a-2q) exhibited promising.anticonvulsant effect ag.inst MES model for inhibition of Lyase- Human Carbonic Anhydrase-I. After investig.ting.all the results, the compound 4-(3-Acetyl-phenylamino)-methyl)-benzenesulfonamide (2g. is found to be best in the series. A comparatively g.od activity of compound 2g.sug.ests us that sulphonamide can be leads to further optimization for building.potent and chemically diversified anti-convulsant ag.nts.

Ang.lica g.auca is one of the important medicinal plants and it is widely used by indig.nous communities for different purposes. The present study analyzes variability in reproductive characters of A. g.auca. The reproductive parts were found having.sig.ificant positive correlation with altitude (e.g.,number of umbellets /umber r= 0.857,p<0.05; umbel diameter r=0.735,p<0.05).
