Quantitative analysis of RNA levels from single hepatocytes in vivo: combined use of real-time RT-PCR and laser microdissection

AIM: The manner in which a cell responds to and influences its environment is ultimately determined by the genes that are expressed. To better understand cellular functions, the isolation of single cells and subsequent quantification of the expressed genes is essential. METHODS: Normal liver tissue was obtained from operation, snap-frozen in liquid nitrogen and sectioned in crystat. Individual hepatocytes were microdissected. RNA was extracted, then reverse transcribed and amplified using real-time quantitative polymerase chain reaction (PCR). RESULTS: Single hepatocytes were dissected by laser beam and catapulted to the microcentrifuge cap which was put above the slide. In this way, cells were collected, RNA was extracted, reverse transcribed to cDNA and used for analysis of RNA expression by real-time quantitative PCR. The amplification results showed that quantitation of the RNA inside the cell was compatible with the number of cells. CONCLUSION: The expression of RNA in single cells can be quantitated successfully by using laser microdissection and real-time PCR. These techniques provide an opportunity to monitor in vivo gene expression levels in single hepatocytes.

This study was conducted to identify the number and density of active licensed acupuncturists(LAcs),as well as the number of accredited schools in acupuncture and Oriental medicine(AOM),as of January 1,2023,in the United States(U.S.).The number of active LAcs as of January 1,2023 was 34,524,potentially33,364 after removing license duplication in multiple states,among which the largest three states were California(with 7317 LAcs[21.19%of the total]),New York(5024[14.55%])and Florida(2644[7.66%]).The total number of LAcs decreased by 8.87%from 2018,and fell short of our projected number of LAcs in 2023 by 9037,or 20.75%.The overall LAc density in the *** as the number of LAcs per 100,000 populationDwas 10.36,less than in *** were 56 active,accredited AOM schools which offered a total of 147 programs(121 at the level necessary for licensing[entry-level],12 for an advanced practicing degree[advanced-level],and 14 for certifications).Broken down further,offerings included 50 master's degrees in acupuncture,40 master's degrees in Oriental medicine,31 entry-level doctorate degrees(10 in acupuncture and 21 in acupuncture plus Chinese herbal medicine),and 12advanced-level doctorate degrees in *** certification programs included one in East-Asian Medical Bodywork and 13 in *** these schools in 2023,institutions in the West and East Coast states comprised 67.86%(decreased from 77.42%in 2018)of the national ***,Florida and Illinois represented 39.29%.There were 48 jurisdictions with acupuncture practice laws in *** data suggests that the acupuncture profession in the *** been significantly impacted during the coronavirus disease 2019 pandemic.

Node synchronization is essential for the stability of the Bitcoin network. Critics have raised doubts about the ability of a new node to quickly and efficiently synchronize with the Bitcoin network and alleviate the storage pressure from existing full nodes to stockpile new data. Basic pruning and other techniques have been explored to address these concerns but have been insufficient to reduce node synchronization delay and effectively suppress the growth of synchronized data. In this study, we propose SnapshotPrune, a novel pruning and synchronization protocol that achieves fast node bootstrapping in the Bitcoin blockchain. Real Bitcoin historical data are leveraged to measure the synchronization time and monitor the network traffic during node bootstrapping. The protocol requires data downloads that are 99.70% less than Bitcoin Core, 81% less than CoinPrune, and 60% less than SnapshotSave, thereby saving 97.23% of download time. Findings show that the proposed design enhances the storage efficiency and reduces the node synchronization delay compared with existing techniques. We hypothesize that the efficiency of this protocol increases with the block height.

The SWEET(sugar will eventually be exported transporter)family proteins are a recently identiﬁed class of sugar transporters that are essential for various physiological *** the functions of the SWEET proteins have been identiﬁed in a number of species,to date,there have been no reports of the functions of the SWEET genes in woodland strawberries(Fragaria vesca).In this study,we identiﬁed 15 genes that were highly homolo-gous to the *** AtSWEET genes and designated them as FvSWEET1–*** then conducted a structural and evolutionary analysis of these 15 FvSWEET *** phylogenetic analysis enabled us to categor-ize the predicted 15 SWEET proteins into four distinct *** observed slight variations in the exon‒intron structures of these genes,while the motifs and domain structures remained highly ***,the developmental and biological stress expression proﬁles of the 15 FvSWEET genes were extracted and ***,WGCNA coexpression network analysis was run to search for possible interacting genes of FvSWEET *** results showed that the FvSWEET10 genes interacted with 20 other genes,playing roles in response to bacterial and fungal *** outcomes of this study provide insights into the further study of FvSWEET genes and may also aid in the functional characterization of the FvSWEET genes in woodland strawberries.

Gestational Diabetes Mellitus (GDM) is a significant health concern affecting pregnant women worldwide. It is characterized by elevated blood sugar levels during pregnancy and poses risks to both maternal and fetal health. Maternal complications of GDM include an increased risk of developing type 2 diabetes later in life, as well as hypertension and preeclampsia during pregnancy. Fetal complications may include macrosomia (large birth weight), birth injuries, and an increased risk of developing metabolic disorders later in life. Understanding the demographics, risk factors, and biomarkers associated with GDM is crucial for effective management and prevention strategies. This research aims to address these aspects comprehensively through the analysis of a dataset comprising 600 pregnant women. By exploring the demographics of the dataset and employing data modeling techniques, the study seeks to identify key risk factors associated with GDM. Moreover, by analyzing various biomarkers, the research aims to gain insights into the physiological mechanisms underlying GDM and its implications for maternal and fetal health. The significance of this research lies in its potential to inform clinical practice and public health policies related to GDM. By identifying demographic patterns and risk factors, healthcare providers can better tailor screening and intervention strategies for pregnant women at risk of GDM. Additionally, insights into biomarkers associated with GDM may contribute to the development of novel diagnostic tools and therapeutic approaches. Ultimately, by enhancing our understanding of GDM, this research aims to improve maternal and fetal outcomes and reduce the burden of this condition on healthcare systems and society. Siliceous diatoms are one of the most prominent actors in the oceans, and they account for approximately 40% of the primary production and particulate organic carbon export flux. It is believed that changes in carbon flux caused by variations in diatom distribution can lead to significant climate shifts. Although the fundamental pathways of diatom-driven carbon sequestration have long been established, there are no reports of CaCO3precipitation induced by marine diatom species. This manuscript introduces novel details regarding the enhancement of aragonite precipitation during photosynthesis in Skeletonema costatum in both artificial and natural seawater. Through direct measurements of cell surfaces via a pH microelectrode and zeta potential analyzer, it was determined that the diatom-mediated promotion of CaCO3precipitation is achieved through the creation of specific microenvironments with concentrated [CO32–] and [Ca2+] and/or the dehydrating effect of adsorbed Ca2+.Based on this mechanism, it is highly plausible that diatom-mediated calcification could occur in the oceans, an assertion that was supported by the significant deviation of total alkalinity（TA） from the conservative TA-salinity mixing line during a Skeletonema costatum bloom in the East China Sea and other similar occurrences. The newly discovered calcification pathway establishes a link between particulate inorganic and organic carbon flux and thus helps in the reassessment of marine carbon export fluxes and CO2sequestration efficiency. This discovery may have important ramifications for assessing marine carbon cycling and predicting the potential effects of future ocean acidification.
