decreasing a-synuclein aggregation by methanolic extract of Centella asiatica in zebrafish Parkinson's model

Objective: To observe the effects of Centella asiatica(C. asiatica) methanolic extract on a-synuclein aggregation and its expression in rotenone-exposed zebra ***: Zebra fish(Danio rerio) were exposed to 5 m g/L rotenone for 28 days and coincubated with 2.5, 5.0 and 10.0 m g/mL of C. asiatica methanolic extract. The medium was changed every 48 h for maintain the concentration of rotenone and extract. After 28 days zebra fish were sacrificed on the ice block and protein was isolated from zebra fish brain for ELISA of dopamine and Western blotting of a-synuclein. Immunohistochemistry was conducted to observe the a-synuclein expressions from histopathological preparation of zebra fish brain. The head were soaked in 10% formaline for less than 24 h and embedded onto paraffin block, then sliced for immunohistochemistry using anti a-synuclein antibody. We also measured zebra fish motility for 5 min in each ***: C. asiatica has important bioactive compounds such as asiaticoside that has antiin flammatory and antioxidant properties. It may inhibit cascade reaction due to oxidative stress induced by rotenone. decreasing reactive oxygen species proposed probability of radical attack to a-synuclein protein that caused aggregation and increase of its *** motility of zebra fish was also maintained in C. asiatica groups due to the increasing dopamine level in rotenone-induced zebra fish. High level of reactive oxygen species inactivated enzyme for dopamine synthesis such as tyrosine hydroxylase, and oxidized dopamine itself. Oxidized dopamine increased a-synuclein aggregation. Thus, the dopamine level decreased in rotenone-induced zebra fish, but C. asiatica increased dopamine ***: C. asiatica has a potential to be developed as an anti-Parkinson's disease treatment due to its capability for minimized the sign of Parkinson's such as a-synuclein aggregation and expression, increasing motility and dopamine as well.

The application of machine learning models to predict material properties is determined by the availability of high-quality *** present an expert-curated dataset of lithium ion conductors and associated lithium ion conductivities measured by *** *** dataset has 820 entries collected from 214 sources;entries contain a chemical composition,an expert-assigned structural label,and ionic conductivity at a specific temperature(from 5 to 873°C).There are 403 unique chemical compositions with an associated ionic conductivity near room temperature(15–35°C).The materials contained in this dataset are placed in the context of compounds reported in the Inorganic Crystal Structure Database with unsupervised machine learning and the Element Movers *** dataset is used to train a CrabNet-based classifier to estimate whether a chemical composition has high or low ionic *** classifier is a practical tool to aid experimentalists in prioritizing candidates for further investigation as lithium ion conductors.

Establishment of Human Neuroblastoma Cell-Line (SK-N-SH) as an In Vitro Model of Morphine Addiction

Morphine is a schedule II-controlled substance that used to allow the diminution of intra-operative, post-operative orchronic pain. However, its usage is limited due to addiction and overdose liabilities. Morphine was observed to cause tolerance,dependence and withdrawal in human. Justification: to date lack of scientific evidence of morphine addiction was carried out byusing specific single human neuroblastoma cell-line (SK-N-SH). Therefore, this study was performed to establish the morphineaddiction model in this cell line. The cells were exposed to morphine for 24 hrs before treatment with methadone, as ananti-withdrawal drug for subsequence 24 hours. The cytosolic fraction of the cell was used in different objectives including receptoraffinity, withdrawal properties, endocytic machinery, desensitisation or internalisation and cellular adaptation. The result shows thatmorphine and methadone bind to the μ-opioid receptor. The morphine-treated cells were observed to increase the expression ofaddiction markers, have a low rate of the endocytic machinery, cause desensitisation of receptor and reduce cellular *** changes by morphine were normalised by the treatment of methadone. As a whole, it is postulated that neuroblastoma cell line,SK-N-SH, can be used as an in-vitro model to demonstrate morphine addiction before animal and human testing.

目的图像分割对于定量分析肺功能降低、无肺功能区域的范围和程度是必需的。该研究的目的是建立和验证一种肺通气和灌注SPECT图像自动分割方法,并比较自动、手动SPECT图像分割结果与相关CT参考容积的差别。方法 77例患者(69例阻塞性肺病患者、8例无明显通气、灌注功能受损患者)先行低剂量CT检查,随后行通气/灌注(V/P) SPECT显像。使用57例患者的低剂量CT图像构建2个用于SPECT图像自动分割的活动形态肺模型(右肺和左肺)。在20例患者的SPECT图像中验证相关分割算法,并将其结果与相应的CT图像分割结果进行比较,同时比较SPECT图像自动分割与手动分割的差别。结果左、右肺自动分割与手动分割的Dice系数(一种相似性度量系数)分别为(0.82±0.05)%和(0.83±0.04)%。SPECT图像自动分割的结果与相关CT参考容积比较,左肺(R=0.69, P<0.001)与右肺(R=0.53, P=0.02)差异有统计学意义。SPECT图像手动分割与相关CT参考容积比较的统计结果与自动分割相似,左肺[偏差(-10±491) ml; R=0.60,P=0.005]、右肺[偏差(36±524) ml; R=0.62, P=0.004]。结论 SPECT图像自动分割与手动分割的结果基本一致。SPECT图像手动分割结果与相关CT参考容积间有较大的容积差别,表明功能性SPECT图像的分割是一项具有挑战性的工作。目前的算法只是实现大范围自动定量测定分析的第一步。
