Association of adiponectin gene polymorphisms with knee osteoarthritis

AIM To investigate the possible relationship of adiponectin(ADIPOQ) gene polymorphisms, plasma adiponectin, and the risk of knee osteoarthritis(OA).METHODS A total of 398 subjects, 202 knee OA patients and 196 healthy individuals, were enrolled in the case-control study. Genotyping at +45T/G(rs2241766) and +276G/T(rs1501299) loci was performed using polymerase chain reaction-restriction fragment length polymorphism. Plasma adiponectin levels were assessed using enzymelinked immunosorbent assay. OA severity was determined using the Kellgren-Lawrence(KL) grading *** No significant associations were observed in the genotype distributions and allele frequencies at two loci of +45T/G and +276G/T polymorphisms in the ADIPOQ betweenknee OA patients and control subjects. There was a significant association between genotype distribution of +276G/T polymorphism and KL grade 2, 3 or 4(P = 0.037, P = 0.046, P = 0.016, respectively). At +45T/G locus, the percentage of GG genotype was notably greater in control subjects(13.40%) compared with OA subjects(1.70%)(P = 0.023). Plasma adiponectin was markedly decreased in OA subjects compared with control subjects(P = 0.03). Likewise, circulating adiponectin in OA subjects was notably lesser than that in control subjects in GG genotype of +45T/G(P = 0.029) and +276G/T polymorphisms(P = 0.012).CONCLUSION Polymorphisms +45T/G and +276G/T of the ADIPOQ gene might not be responsible for OA susceptibility among Thais.

Osteoarthritis(OA)is a degenerative articular disorder manifested by cartilage destruction,subchondral sclerosis,osteophytes,and synovitis,resulting in chronic joint pain and physical disability in the *** purpose of this study was to investigate mitochondrial DNA copy number(mtDNACN)and inflammatory cytokines in primary knee OA patients and healthy volunteers.A total of 204 knee OA patients and 169 age-matched healthy volunteers were *** relative blood leukocyte mtDNACN was assessed by quantitative real-time polymerase chain reaction(qRT-PCR),and ten inflammatory cytokines in their plasma were detected by multiplex *** leukocyte mtDNACN in the OA group was significantly lower than that in the control *** mtDNACN in the control group was negatively correlated with their age(r=−0.380,P<0.0001),whereas mtDNACN in the OA group was positively correlated with their age(r=0.198,P<0.001).Plasma interleukin-4(IL-4)and IL-6 were significantly higher in the knee OA group than in the control *** plasma IL-6 level was positively correlated with blood leukocyte mtDNACN in the OA group(r=0.547,P=0.0014).IL-5 showed as a major factor(coefficient 0.69)in the second dimension of principle components analysis(PCA)-transformed data and was significantly higher in the OA group(P<0.001)as well as negatively correlated with mtDNACN(r=−0.577,P<0.001).These findings suggest that elevation of plasma IL-4 and IL-6 and a relative reduction in mtDNACN might be effective biomarkers for knee ***-5 is a plausible factor responsible for decreasing blood leukocyte mtDNACN in knee OA patients.

The title compound, neogeodin hydrate （C17H14C1208, CAS： 94540-50-8）, was derived from marine fungus Aspergilhts terreus CRIM301. It was unequivocally characterized by IR, NMR spectroscopies, and single-crystal X-ray crystallography and tested for various biological activities. Neogeodin hydrate crystallizes in the triclinic space group P1 with a = 8.1159（5） A, b = 8.2472（4） A, c= 14.1278（7） A, a = 81.448（2）°, β = 84.860（2）°, γ= 70.400（2）°, V = 880.13（8） A3; Z = 2. It comprises a diphenyl ether, asterric acid skeleton and dichloro substituents. The methoxyphenoxy rings of the inversely related molecules form a ribbon-like structure that is stabilized by O-H...O hydrogen bonds through the doubly disordered carboxyl groups and by C-H...O interactions, generating the same R22（8） ring motif. The chlorinated methylbenzoate rings, making mostly a right angle, link the parallel upper and lower ribbons via bifurcated O-H...O and C-H...O hydrogen bonds, yielding endless channels. The channels formed are further sustained by C-H...O and π...π interactions Neogeodin hydrate exhibits inhibition against superoxide anion radical formation in the xanthine/xanthine oxidase （XXO） assay, but has no aromatase inhibitory activity.

A novel isoxazole compound CM2-II-173 inhibits the invasivephenotype of triple-negative breast cancer cells

Invasion and metastasis are important hallmarks of breast cancer and are the leading cause of patient ***-negative breast cancer (TNBC) is an aggressive type of breast cancer characterized by a poor prognosis and a lackof effective targeted therapies. The present study investigated the inhibitory effect of a novel FTY720 derivative on theinvasive phenotype of TNBC cells. Here, we showed that a novel compound with an isoxazole ring, 4-(3-Decylisoxazol-5-yl)-1-hydroxy-2-(hydroxymethyl)butan-2-aminium chloride (CM2-II-173), significantly inhibitedinvasiveness of MDA-MB-231 TNBC cells. Expression of matrix metalloproteinase (MMP)-9 and invasiveness ofMCF10A normal breast cells induced by sphingosine-1-phosphate (S1P) were reduced by CM2-II-173 *** of pMEK1, pAkt, pERK, and p38 MAPK by S1P were inhibited by treatment with *** and anchorage-independent growth of MDA-MB-231 TNBC cells were significantly decreased by CM2-II-173. CM2-II-173 efficiently induced apoptosis in MDA-MB-231 TNBC cells. CM2-II-173 significantly inhibitedinvasive phenotypes of breast, liver, prostate, and ovarian cancer cells. CM2-II-173 exhibited a more potent effect onthe invasiveness of MDA-MB-231 TNBC cells compared to FTY720. Taken together, this study demonstrated thatCM2-II-173 has the potential to be a lead compound that can inhibit cancer progression of not only TNBC cells, butalso of liver, prostate, and ovarian cancer cells.

A comprehensive health education plus monitoring support program for older adults with knee osteoarthritis coexisting with overweight and type 2 diabetes

Objectives:To examine the patient outcomes of a comprehensive health education plus village health volunteer monitoring support program on older adults with knee osteoarthritis who are overweight and have type 2 ***:Quasi-experimental,two-group,pretest-posttest design was *** random geographic cluster sampling,older adults who met the inclusion criteria were assigned to two groups,62 in the intervention group and 71 in the control *** intervention group received the comprehensive health education plus village health volunteer monitoring support *** control group received only the comprehensive health education *** outcomes,including blood glucose level,knee pain and range of motion,body weight,physical ability(Timed Up and Go Test),fatigue,depressive symptoms,quality of sleep,and quality of life,were assessed at baseline,3 months,and 6 months after enrollment and analyzed using descriptive statistics and multivariate analysis of ***:One hundred and ten participants completed the study(55 participants in each group).Most patient outcomes in the two groups showed general improvement with statistical significance(P<0.001):pain decreased,physical ability improved,less depressive symptoms and fatigue,quality of life and sleep *** interaction effect(the group and time)demonstrated statistically significant differences between the intervention and control groups on pain,knee range of motion,and fatigue according to each time follow-up(P<0.001).Conclusions:The comprehensive health education plus village health volunteer monitoring support program promotes good patient outcomes in this *** integrated health education resource and support for older adults with knee osteoarthritis and type 2 diabetes is an effective,non-surgical treatment that highlights professional nursing roles and non-professional rolesdvillage health *** should consider implementing a health education plus monito

Comparison of protein patterns between Plasmodium falciparum mutant clone T9/94-M1-1(b3) induced by pyrimethamine and the original parent clone T9/94

Objective:To compare the protein patterns from the extracts of the mutant clone T9/94-M1-1(b3)induced by pyrimethamine,and the original parent clone T9/94 following separation of parasite extracts by two-dimensional electrophoresis(2-DE).Methods:Proteins were solubilized and separated according to their charges and *** separated protein spots were then detected by silver staining and analyzed for protein density by the powerful image analysis ***:Differentially expressed protein patterns(up—or down-regulation)were separated from the extracts from the two clones.A total of 223 and 134 protein spots were detected from the extracts of T9/94 and T9/94-M1-1(b3)clones,*** reduction in density of protein expression was observed with the extract from the mutant(resistant)clone compared with the parent(sensitive)clone.A total of 25 protein spots showed at least two-fold difference in density,some of which exhibited as high as ten-fold ***:These proteins may be the molecular targets of resistance of Plasmodium falciparum to *** study to identify the chemical structures of these proteins by mass spectrometry is required.

Abiotic and Biotic Factors Controlling Grain Aroma along Value Chain of Fragrant Rice:A Review

The aroma of fragrant rice is one of the grain quality attributes that significantly influenceconsumer preferences and prices in world markets. The volatile compound 2-acetyl-1-pyrroline (2AP) isrecognized as a key component of the aroma in fragrant rice. The variation in grain 2AP content amongvarious fragrant rice varieties is associated with the expression of the badh2 gene, with 19 alleles havingbeen identified so far. The grain 2AP content is strongly influenced by environmental and managementfactors during cultivation as well as post-harvest conditions. This review pinpointed the major abiotic andbiotic factors that control grain 2AP content. Abiotic factors refer to water, temperature, light quality,fertilizer application (both macro- and micro-nutrients), and soil properties, including salinity, while bioticfactors include microorganisms that produce aromatic compounds, thus influencing the grain aroma infragrant rice. Post-harvest management, including storage and drying conditions, can significantly impactthe grain 2AP content, and proper post-harvest conditions can intensify the grain aroma. This reviewsuggests that there are rice varieties that can serve as potential sources of genetic material for breedingrice varieties with high grain aroma content. It offers an overview of recent research on the major factorsaffecting the aroma content in fragrant rice. This knowledge will facilitate further research on theproduction of high-quality rice to meet the demands of farmers and consumers.

Mutated Clones of Caladium Humboldtii ＇Phraya Savet＇ from in vitro Culture and Occurrence of Variants from Somatic Hybridization between Two Caladium Species

Variations in Caladium humboldtii cv. ＇Phraya Savet＇ from in vitro culture were observed. Callus and small shoots were induced from unexpanded leaf segments cultured on modified MS medium supplemented with 2.69 p.M l-Naphthalene acetic acid （NAA） and 17.76 μM N6-Benzyladenine （BA）. Shoots were transferred onto modified MS medium supplemented with 8.88 μM BA for shoot multiplication. Subsequently, roots were induced on MS without growth regulator. The regenerated plantlets were vigorously grown in glasshouse conditions. From 4 morphological groups （leaf color ratio, leaf color, petiole and leaf pattern）, the regenerated ＇Phraya Savet＇ caladium plants were divided into 6 types. The occurrence of variants was 52%. Most of the mutated plants were observed from only leaf color ratio （green ： white） and leaf shape. The most significance for commercial value from mutated clones was round leaf obtaining 20%. Characteristics of new clones from somatic hybridization between C humboldtii cv. ＇Phraya Savet＇ and C. bicolor cv. ＇Suvarnabhum＇ using thin cell layer technique from in vitro calli were investigated. Each thin cell layer of induced callus, about 0.5 - 1 mm thick, from both caladiums was alternately laid on the top of each other for 8 layers. Subsequently, the combination of thin cell layers was cultured and the regenerated plantlets were grown in glasshouse conditions. From 3 morphological groups （leaf pattern, leaf color and petiole）, the regenerated caladium plants were found dissimilarly to both original caladiums at 85 percent with 8 types of different characters. Somatic hybridization between C. humboldtii cv. ＇Phraya Savet＇ and C. bicolor cv. ＇Suvarnabhum＇ gave rise to a number of most hybrids with all conserving C. bicolor characters.

Higher plasma bilirubin predicts veno-occlusive disease in early childhood undergoing hematopoietic stem cell transplantation with cyclosporine

AIM: To analyze the association between plasma bilirubin levels and veno-occlusive disease(VOD) in non-adult patients undergoing hematopoietic stem cell transplantation(HSCT) during cyclosporine ***: A total of 123 patients taking cyclosporinewere evaluated using an electronic medical system at the Seoul National University Children's Hospital from the years 2004 through 2011. Patients were grouped by age and analyzed for incidence and type of adverse drug reactions(ADRs) including VOD. RESULTS: The HSCT patients were divided into three age groups: G#1 ≥ 18; 9 ≤ G#2 ≤ 17; and G#3 ≤ 8 years of age). The majority of transplant donor types were cord blood transplantations. Most prevalent ADRs represented acute graft-vs-host disease(a GVHD) and VOD. Although the incidences of a GVHD did not vary among the groups, the higher frequency ratios of VOD in G#3 suggested that an age of 8 or younger is a risk factor for developing VOD in HSCT patients. After cyclosporine therapy, the trough plasma concentrations of cyclosporine were lower in G#3 than in G#1, indicative of its increased clearance. Moreover, in G#3 only, a maximal total bilirubin level(BILmax) of ≥ 1.4 mg/d L correlated with VOD incidence after cyclosporine therapy. CONCLUSION: HSCT patients 8 years of age or younger are more at risk for developing VOD, diagnosed as hyperbilirubinemia, tender hepatomegaly, and ascites/weight gain after cyclosporine therapy, which may be represented by a criterion of plasma BILmax being ≥ 1.4 mg/d L, suggestive of more sensitive VOD indication in this age group.
