Concordance in the Interpretation of Cervical Cytology for the Early diagnosis of Cervical Cancer

Objective: To identify the diagnostic concordance in the interpretation of cervical smears. Material and Methods: Cross-sectional study from October 2011 to January 2013. 50 samples were read by every 4 cyto-technologists who are the total staffs that read and interpret cytological specimens from a public health institution of the city of San Luis Potosí, México. The cytological diagnosis was confirmed by a certified pathologist. Checklist was developed to determine the quality of the interpretation composed of three sections: adequate sample, inadequate sample and cytological diagnosis. The diagnostic report was made by Bethesda System 2001 classification. diagnostic concordance was measured through the Kappa coefficient. To establish the differences in cytological diagnosis between each cyto-technologist, chi square test was applied. Results: The concordance is acceptable in the classification of samples as inadequate (k = 0.66). The negative diagnosis was a significant concordance between each cytotechnologist and pathologist;however, no case had very good concordance. In the epithelial abnormalities, only the cytotechnologist 2 had significant concordance with the pathologist, however, its concordance is low. In the case of glandular abnormalities, only cytotechnologist 1 had no significant concordance with the pathologist. The percentage of true negatives was 30%, true positives 20%, false negatives 50% and false positives 0 %. There are significant differences between the readings of cyto-technologist 1 and 2 (p < 0.008), between 1 to 4 (p < 0.001), between 2 and 3 (p < 0.05), and finally between 3 and 4 (p < 0.003). Conclusions: The concordance between cyto-technologists and pathologists is below the required minimum level set by national and international standards.

OBJECTIVE： To determine the role of a pharmacokinetic interaction in the protective effect of curcumin against the gastric damage induced by indomethacin administration as such or as its prodrug acemetacin. METHODS： Wistar rats orally received single dose of indomethacin （30 mg/kg） with and without curcumin （30 mg/kg）; gastric injury was evaluated by determining the total damaged area. Additional groups of rats received an oral single dose of indomethacin （30 mg/kg） or its prodrug acemetacin （34.86 mg/kg） in the presence or absence of curcumin （30 mg/kg）. Indomethacin and acemetacin concentrations in plasma from blood draws were determined by high-performance liquid chromatography Plasma concentration-against-time curves were constructed, and bioavailability parameters, maximal concentration （Cmax） and area under the curve to the last sampling time （AUC0_t） were estimated. RESULTS： Concomitant administration of indomethacin and curcumin resulted in a significantly reduced gastric damage compared to indomethacin alone. However, co-administration of curcumin did not produce any significant alteration in the bioavailability parameters of indomethacin and acemetacin after administration of either the active compound or the prodrug. CONCLUSION： Curcumin exhibits a protective effect against indomethacin-induced gastric damage, but does not produce a reduction of the bioavailability of this nonsteroidal anti-inflammatory drug, indomethacin. Data thus suggest that a pharmacokinetic mechanism of action is not involved in curcumin gastroprotection.

Satisfaction of the Service Provider with the Organization of the Timely Cervical Cancer Detection Program

Cervical cancer is a priority health problem in Mexico. The objective of this work was to know the degree of satisfaction of the service provider with the organization of the program of Timely Detection of Cervical Cancer, specifically in taking of the cervical cytology. The study design was observational, transversal and prospective conducted from January to May 2015 in Jurisdiction 1 of the Health Services of San Luis Potosí, Mexico, in 20 health centers. Participants were 87 service providers whose function is to take cervical cytology (Pap smears). A 32-question questionnaire was applied, with Likert scale with 5 levels of response: 1 (dissatisfied), 2 (little satisfied), 3 (indifferent), 4 (satisfied) and 5 (very satisfied). The score range was 32 points (minimum job satisfaction) up to 160 points (Maximum job satisfaction). We analyzed 2 dimensions: 1) intrinsic factors that have to do with motivation and professional recognition, and 2) extrinsic factors related to infrastructure, materials, remuneration and labor policies. Each factor consisted of four categories. Descriptive statistics and linear correlation of Pearson were applied for the analysis of data. Participants signed an informed consent letter. Of 4 categories, in the intrinsic dimension, the one with the highest percentage of satisfaction was “delegation of activities” with 28.8%. In the extrinsic dimension, the category with the highest percentage of satisfaction was “infrastructure and materials” with 49.4%;while satisfaction increase in intrinsic dimensions also does in extrinsic dimensions, such correlations are significant (p < 0.05), except between delegation of activities and remuneration (r = 0.074 and p = 0.497). The higher hierarchical level, the men, the bachelor academic level and older people showed the highest level of satisfaction.

Human-induced pluripotent stem cells(iPSCs)are an accessible source of adult-derived,patient-specific pluripotent stem cells for use in basic research,drug discovery,disease modeling,and stem cell *** the accessibility of methods to obtain iPSCs regardless of the cell source can enhance their clinical ***,our purpose is to report a simple protocol to obtain iPS-like cells from urine-derived renal epithelial cells(RECs)using different extracellular matrices and transfection *** this study,we began by culturing urine-derived cells from healthy donors to establish a primary culture of renal epithelial cells,followed by their ***,we generated iPS-like cells by transfecting renal epithelial cells(RECs)with vectors expressing Oct4,Sox2,L-Myc,Lin-28,and Klf4,and we compared the efficacy of different extracellular matrices and transfection *** resultant iPS-like cells showed a human embryonic stem cell-like morphology and expressed the specific pluripotency markers Oct3/4,Nanog,Lin28,and *** concluded that Lipofectamine Stem Cell transfection reagent is more effective than FuGENE in obtaining iPS-like cells under the conditions ***,the three matrices are similar in their efficiency of obtaining iPS-like *** report provides an experimental protocol for obtaining and generating iPS-like cells from urine samples for further cell therapy research on different human diseases.

The physicochemical properties and catalytic activity of pure and sulfated titanium oxide (TiO2 and ) is described in this work. Titanium hydroxide synthesized by the sol-gel method was impregnated with a 1 N H2SO4 solution, varying amount of sulfate ions () in the range from 10 to 20 wt%. Pure and modified hydroxides were calcined at 500°C for 3 h and then characterized by TGA-DTG, XRD, BET, FT-IR, potentiometric titration with n-butylamine and 2-propanol dehydration. Catalytic activity of materials was tested in the n-hexane isomerization at 350°C. The results showed that TiO2 and mainly developed anatase phase. All have acceptable specific surface area (95-105 m2/g). Potentiometric titration with n-butylamine revealed that showed higher acidity (430-530 mV) than compared to pure TiO2 (﹣15 mV), indicating that this oxide only has weak acidity. The results showed good relationship between acidity determined by potentiometric titration with n-butylamine and the catalytic activity evaluated by 2-propanol dehydration and n-hexane isomerization. Titanium oxide with 20 wt% ofions was the material that demonstrated the highest catalytic activity for both reactions.
