Plasmonic polydopamine-modified TiO_(2)nanotube substrates for surface-assisted laser desorption/ionization mass spectrometry imaging

Mass spectrometry imaging(MSI)has made the spatio-chemical characterization of a broad range of small-molecule metabolites within biological tissues ***,available matrix-assisted laser desorption/ionization mass spectrometry(MALDI-MS)suffers from severe background interferences in low-mass ranges and inhomogeneous matrix ***,surface-assisted LDI-MS(SALDI-MS)has been an attractive alternative for high-sensitivity detection and imaging of small *** this study,we construct a new composite substrate,hydrophobic polydopamine(hPDA)-modified TiO_(2)nanotube(TDNT)coated with plasmonic gold nanoparticle(AuNP-hPDA-TDNT),as a dual-polarity SALDI substrate using an easy and cost-effective fabrication *** from the synergistic effects of TDNT semiconductor and plasmonic PDA modification,this SALDI substrate exhibits superior performance for dual-polarity detection of a vast diversity of small *** reduced background interferences,lower detection limits,and spot-to-spot repeatability can be achieved using AuNP-hPDA-TDNT *** to its unique imprinting performance,various metabolites and lipids can be visualized within jatropha integerrima petals,ginkgo leaves,strawberry fruits,and latent *** valuably,the universality of this matrix-free substrate is demonstrated for mapping spatial distribution of lipids within mouse brain tissue *** together,this AuNP-hPDA-TDNT material is expected to be a promising SALDI substrate in various fields,especially in nanomaterial development and life sciences.

Hidden Genetic Regulation of Human Complex Traits via Brain Isoforms

Alternative splicing exists in most multi-exonic genes,and exploring these complex alternative splicing events and their resultant isoform expressions is ***,it has become conventional that RNA sequencing results have often been summarized into gene-level expression counts mainly due to the multiple ambiguous mapping of reads at highly similar ***-level quantification and interpretation are often overlooked,and biological interpretations are often deduced based on combined transcript information at the gene ***,for the most variable tissue of alternative splicing,the brain,we estimate isoform expressions in 1,191 samples collected by the Genotype-Tissue Expression(GTEx)Consortium using a powerful method that we previously *** perform genome-wide association scans on the isoform ratios per gene and identify isoform-ratio quantitative trait loci(irQTL),which could not be detected by studying gene-level expressions *** analyzing the genetic architecture of the irQTL,we show that isoform ratios regulate edu-cational attainment via multiple tissues including the frontal cortex(BA9),cortex,cervical spinal cord,and *** tissues are also associated with different neuro-related traits,including Alzheimer’s or dementia,mood swings,sleep duration,alcohol intake,intelligence,anxiety or depression,*** randomization(MR)analysis revealed 1,139 pairs of isoforms and neuro-related traits with plausible causal relationships,showing much stronger causal effects than on general diseases measured in the UK Biobank(UKB).Our results highlight essential transcript-level biomarkers in the human brain for neuro-related complex traits and diseases,which could be missed by merely investigating overall gene expressions.

Effects of Different Woodland Types on Soil Properties and Soil Nematode Community Structure

Upland red soil has strong acidity and low fertility, different woodland types produce varying impacts on soil properties and nematode communities. To explore the response of soil, 2 different types of woodland were selected to investigate the changes of soil properties and nematode communities compared to wasteland. The results showed that the content of soil organic carbon and total nitrogen in chestnut forest soil were the highest, 16.41 g/kg and 0.46 g/kg respectively; while the highest contents of soil microbial biomass carbon, nitrogen and phosphorus appeared in wasteland soil, and basal respiration of wasteland soil was 28.36 mg CO_2/kg·d, significantly higher than that of the other 2 soils(p <0.05). The nematode abundance also reached the peak in wasteland soil. Three types of soil exhibited a completely different nematode community structure: the dominant trophic group in wasteland soil was herbivores,while omnivores/predators and bacterivores in forest from farmland and chestnut forest soil, *** influence of different types of woodland on ecological indices of nematode communities was not consistent. Degradation trends of soil properties and nematode communities were presented in the 2woodland types compared with wasteland soil. These results are instructive for the local woodland planning.

Dear Editor, Brassica rapa is an important crop and a model plant for studying Brassica genome evolution and gene function. The silique of B. rapa derived from the gynoecium nor- mally consists of two carpels that are separated by a false septum, and thus has two Iocules （bilocular）. Multilocular （more than two carpels） lines of B. rapa have been discov- ered in nature and are considered advantageous because the multilocular type could potentially increase the seed yield by producing more seeds per silique than the biloc- ular type. However, the molecular basis of the important seed vield-related trait is ooorlv understood.

Flexible bidirectional pulse charging regulation achieving long-life lithium-ion batteries

Typical application scenarios,such as vehicle to grid(V2G)and frequency regulation,have imposed significant long-life demands on lithium-ion ***,we propose an advanced battery life-extension method employing bidirectional pulse charging(BPC)*** traditional constant current charging methods,BPC strategy not only achieves comparable charging speeds but also facilitates V2G frequency regulation *** significantly enhances battery cycle ampere-hour throughput and demonstrates remarkable life extension *** this interesting conclusion,adopting model identification and postmortem characterization to reveal the life regulation mechanism of BPC:it mitigates battery capacity loss attributed to loss of lithium-ion inventory(LLI)in graphite anodes by intermittently regulating the overall battery voltage and anode potential using a negative charging ***,from the perspective of internal side reaction,the life extension mechanism is further revealed as inhibition of solid electrolyte interphase(SEI)and lithium dendrite growth by regulating voltage with a bidirectional pulse current,and a semi-empirical life degradation model combining SEI and lithium dendrite growth is developed for BPC scenarios health management,the model parameters are identified by genetic algorithm with the life simulation exhibiting an accuracy exceeding 99%.This finding indicates that under typical rate conditions,adaptable BPC strategies can extend the service life of LFP battery by approximately 123%.Consequently,the developed advanced BPC strategy offers innovative perspectives and insights for the development of long-life battery applications in the future.

Evaluation of the Therapeutic Significance of Arsenite and Thalidomide in Patients with Myelodysplastic Syndrome

Objective:To analyze the therapeutic significance of arsenite and thalidomide in patients with myelodysplastic ***:From August 2021 to August 2022,80 patients with myelodysplastic syndrome were selected and randomly divided into two *** the control group,the drug used was thalidomide tablets,whereas in the observation group,the drug used was arsenite and *** treatment effect was observed and ***:Before treatment,there was no statistical significance in the blood and serological indices between the two *** treatment,the observation group showed better blood and serological indices than the control group(P<0.05).The clinical efficacy of the observation group was 77.5%,while that of the control group was 50.0%.The observation group had significantly better treatment effect(P<0.05).The incidence of adverse effects in the observation group and the control group was 5.0%and 20.0%,*** observation group had significantly fewer adverse effects(P<0.05).Conclusion:In the treatment of patients with myelodysplastic syndrome,the use of arsenite on the basis of thalidomide can effectively improve the treatment effect and optimize the levels of various blood and serological indices,with fewer adverse effects and a relatively high safety profile.

A ROS-Responsive Aspirin Polymeric Prodrug for Modulation of Tumor Microenvironment and Cancer Immunotherapy

Tumor-promoting inflammation is accompanied by cancer initiation,progression,and ***-2(COX-2)and its downstream product,prostaglandin E2(PGE2),play critical roles in tumor-promoting *** studies have revealed the potential of COX-2 inhibition in improving cancer response to chemotherapy,as well as ***,a nonsteroidal anti-inflammatory drug,has been reported as a COX-2 ***,as a small molecule drug with a carboxyl group,there is still the lack of effective methods of preparing polymer–aspirin conjugates with tumor stimuli-responsive release ***,we synthesized a reactive oxygen species(ROS)-responsive aspirin polymeric prodrug(P3C-Asp)via Passerini three-component reaction between aspirin,4-formylbenzeneboronic acid pinacol ester,and 5-isocyanopent-1-yne,followed by copper(I)-catalyzed alkyne-azide cycloaddition“click”reaction of the aspirin prodrug with dextran(DEX).The P3C-Asp could release aspirin and salicylic acid in response to tumor-specific *** the murine colorectal cancer model,P3C-Asp suppressed tumor growth effectively without significant side effects and eradicated tumors when combined with the immune checkpoint inhibitor,anti-PD-1 antibody(aPD-1).Further analysis revealed that the suppression was attributable to changes in the immune microenvironment,including reduced PGE2 content,as well as increased infiltration of CD8+T cells and M1 *** results mentioned above proved that targeting COX-2 pathway with a proper polymeric prodrug might be a useful strategy for cancer immunotherapy.
