Fern extracts potentiate fluconazole activity and inhibit morphological changes in Candida species

Objective: To investigate the antifungal activity of the fern species Lygodium venustum(L. venustum) and Pityrogramma calomelanos(P. calomelanos) against Candida albicans and Candida tropicalis ***: The microdilution method was used to evaluate the antifungal activity, as well as the modulating effects of ethanolic extracts of these plants in combination with fluconazole. The minimum inhibitory concentration(MIC), minimum fungicide concentration and morphological changes were also ***: The extract obtained from L. venustum presented a MIC > 8 192 mg/m L, while the extract obtained from and P. calomelanos presented a MIC = 8 192 mg/m L, indicating that they present weak antifungal activity. However, combination of the extracts with Fluconazole potentiated the antifungal activity of this drug. At different experimental conditions, such as concentration of the extract and type of strain, the extracts inhibited hyphae and pseudohyphae formation, indicating that these fern species can affect the morphology of the ***: The extracts obtained from the fern species L. venustum and P. calomelanos dose not present significant antifungal activity. However, P. calomelanos potentiates the activity of fluconazole and both extracts inhibits the morphological changes in Candida species, indicating that they have potential pharmacological activity as modulators of fungal biology. Therefore, novel studies are required to characterize the interference of these extracts in the virulence and pathogenicity of Candida species as well as the potential of fern species to treat fungal infections.

Acute fatty liver of pregnancy is a rare disease that affects women in the third trimester of pregnancy. Although infrequent, the disease can cause maternal mortality. The diagnosis is not always clear until the pregnancy is terminated, and significant complications, such as acute pancreatitis, can occur. Pancreatic involvement typically only occurs in severe cases after the development of hepatic and renal impairment. To date, little knowledge is available regarding how the disease causes pancreatitis. Treatment involves supportive measures and pregnancy interruption. In this report, we describe a case of a previously healthy 26-year-old woman at a gestational age of 27 wk and 6 d who was admitted with severe abdominal pain and vomiting. This case illustrates the clinical and laboratory overlap between acute fatty liver of pregnancy and pancreatitis, highlighting the difficulties in differentiating each disease. Furthermore, the hypothesis for this overlapping is presented, and the therapeutic options are discussed.

Knowledge about Diabetes Mellitus and Self-Care Activities before and after an Educational Program: A Pilot Study

background: Health education has proven to be an effective strategy to enable people with diabetes mellitus to manage this condition. However, few studies in brazilian population samples have been conducted to evaluate the effects of educational programs on diabetes knowledge and self-care. Aim: To evaluate the knowledge about diabetes mellitus and the performance of self-care activities, before and after participation in an educational program. Methods: This is an intervention study, with a quantitative approach, in a single comparison group, for the analysis of “before and after” results related to an educational program focused on self-care and concurrent physical training. The study interventions consisted of 42 exercise sessions, as well as individual educational meetings, according to the needs of each participant and through nursing consultations, using educational material prepared from the literature. The sample was initially composed of 33 adults with diabetes mellitus, but 18 completed the study. For the assessment of knowledge and self-care activities, the revised brazilian versions of the Diabetes Knowledge Scale and the Diabetes Self-Care Activity Questionnaire were used, respectively, in the two moments of the study, that is, prior to the first educational meeting and immediately before the first physical training session, and after the last educational meeting, which ran parallel to the 42nd physical training session, making a six-month interval between the two assessment moments for each participant. Results: The analysis of knowledge about diabetes showed significant improvement after the educational program and, as for self-care, there was clinical improvement in all dimensions, but only the dimension “general diet” obtained statistically significant improvement. Conclusion: The educational program has been shown to be beneficial for improving knowledge and self-care of the disease, which reiterates the need to maintain interventions of this natur

Evaluation of Porous Structure of Cement Pastes Made with Residual Rice Husk Ash

In several countries, the residual RHA （rice husk ash） has been produced in rice processing industries or in thermoelectric plants that use rice husk to generate heat and/or electrical energy, usually without burning process control. This causes a reduction in the amorphous silica content of residual RHA, which distinguishes them from the RHA produced according to controlled burning process, which is totally amorphous and considered a highly reactive pozzolan. In this paper, the hydration products and the porous structure of binders paste were studied by replacing, in weight of 5%, 10% and 20% of Portland cement OPC （ordinary Portland cement）, by residual RHAs named A and b, which have high and low content of amorphous silica, respectively, using microstructure evaluation techniques as XRD （X-ray diffraction）, TG （thermogravimetric） tests and MIP （mercury intrusion porosimetry）. A reducing the size of the pores of the pastes was observed according to the increase of content replacement of RHA A and RHA b.

Recent data indicate that IGF1R/IRS signaling is a potential therapeutic target in bCR-AbL1-negative myeloproliferative neoplasms(MPN);in this pathway,IRS2 is involved in the malignant transformation induced by JAK2^(V617F),and upregulation of IGF1R signaling induces the MPN ***157,a synthetic compound designed as an IGF1R-IRS1/2 inhibitor,has been shown to induce antineoplastic effects in solid ***,we aimed to characterize the molecular and cellular effects of NT157 in JAK2^(V617F)positive MPN cell lines(HEL and SET2)and primary patient hematopoietic *** JAK2^(V617F)cell lines,NT157 decreased cell viability,clonogenicity,and cell proliferation,resulting in increases in apoptosis and cell cycle arrest in the G2/M phase(pbited IRS1/2,JAK2/STAT,and NFκb signaling,and it activated the AP-1 complex,downregulated four oncogenes(CCND1,MYb,WT1,and NFKb1),and upregulated three apoptotic-related genes(CDKN1A,FOS,and JUN)(pbited erythropoietin-independent colony formation in cells from polycythemia vera patients(p<0.05).These findings further elucidate the mechanism of NT157 action in a MPN context and suggest that targeting IRS1/2 proteins may represent a promising therapeutic strategy for MPN.

Culture of Scenedesmus acuminatus in Corn Steep Liquor

Microalgae have been evaluated as a source of lipids for biodiesel production. They can be grown on effluent and produce biomass while removing organic and mineral components from the medium. The use of agro-industrial wastes for the production of microalgae is an alternative to reduce the costs related to the composition of the culture medium, thus reducing operational costs considerably. This work was conceived as a research about the cultivation of microalgae Scenedesmus acuminatus on the substrate composed by corn steep liquor. A central composite design （CCD） was carried to study the influence of the effluent and inoculum concentration on the chemical oxygen demand （COD） of the culture. Statistical analysis indicated that the percentage of inoculum and effluent had an influence on the COD. The cultivation, together with the coagulation procedure, was efficient for the removal of organic matter, verified through the COD reduction and changing of pH value. These two parameters reached the appropriate level according to the standard required by current legislation.

Leptin Signaling Modulates Expression of Polycomb and Trithorax Complexes in the brain of Fat Tissue Implanted Polycystic Ovarian Sindrome Mice

The Polycystic Ovary Syndrome (PCOS) is the most common androgenic disorder in women during reproductive life. PCOS may also be accompanied by metabolic syndrome and recent studies point to leptin as playing a role in disrupting infertility and in changing the energy balance in obese mice through its action on the hypothalamus. The aim is to assess the expression of the Polycomb & Trithorax Complexes genes in brain of mice transplanted with fat tissue from normal mice, in order to better understand the neuronal mechanisms underlying the reversion of PCOS. Three b6 V-Lepob/J mouse groups: Normal weight, obese and seven-day-treatment obese had their brain RNA extracted and submitted to an 84 Polycomb & Trithorax Complexes genes PCR Array plate and MetacoreTM pathways localization. Genomic profiles obtained were compared to the ones of the normal-weight-mice group. Differentially expressed genes were 13% and 26% respectively to control and treatment. Major changes were in genes: Snai1/31;Smarca1/?17;Dnmt3b/4.7;Ezh1/ 15. Altered genes were associated to canonical pathways and provided 3 networks related to epigenetics. Underlying neuronal changes caused by leptin in obese mice brain, there is an important role being played by the histone code. Here there is evidence that leptin drives the chromatin packing to a more condensed pattern. Upregulation of methyltransferase genes, like Ezh1, favors this thought. In summary the Polycomb & Trithorax complexes might answer for the silencing of some downregulated genes in the obese mice brain when exposed to leptin.

Effect of Different Sources and Doses of Nitrogen on Vegetative and Productive Performance of Sugarcane under a Dystrophic Red Latosol Condition

The aim of this work was to evaluate the effects of the application of different sources and doses of nitrogen(N)in the growth and productivity of sugarcane in cane *** experiments were conducted in the area of Rio Paraiso II farm,belonging to the Raízen factory,in the city of Jataí,*** soil was classified as dystrophic red *** experimental design used was in randomized block,analysed in factorial scheme of 2×4×4,with three *** factors analysed were two sources of N(urea and ammonium nitrate),four doses of N(0,60,120 and 180 kg/ha)and four evaluation times(210,250,290 and 330 d after planting(DAP)).The parameters analysed were plant height(PH),diameter of stalks(DS),length of internodes(LI)and productivity of stalks(PS).Results observed significant difference in the sources and doses of N for the evaluated parameters in sugarcane *** in the average doses of 110 kg/ha and 100.87 kg/ha provided the greatest PH and LI of sugarcane,*** nitrate at a dose of 180 kg/ha presented the largest DS of sugarcane(cane plant)at 330 *** PS of sugarcane SP80-1816 at 60 kg/ha and 120 kg/ha ammonium nitrate has higher *** the PS,it is recommended the application of doses between 60 kg/ha and 120 kg/ha of N of the source ammonium nitrate in sugar cane(cane plant).
