Comprehensive Summary The experimental investigation of rare-earth metal-metal bonds remains a challenge in the study of chemical ***,we report the synthesis and characterization of a novel heteronuclear di-metallofullerene,ScY@C_(3v)(8)-C_(82),which contains a mixed rare-earth metal-metal ***@C_(3v)(8)-C_(82)was successfully synthesized by arc-discharging method and characterized by mass spectrometry,UV-vis-NIR spectroscopy and single-crystal X-ray diffraction crystallography,which unambiguously determined its molecular *** calculations were also performed to study the optimized positions of Sc-Y metallic dimer and the electronic *** combined experimental and theoretical results confirmed that both Sc and Y atoms transfer two electrons to the C_(3v)(8)-C_(82)cage,i.e.,(ScY)4+@(C_(3v)(8)-C_(82))4-.In particular,a covalent Sc-Yσ2 bond,which has never been reported before,is proven to be formed inside C_(3v)(8)-C_(82)fullerene *** work presents a novel di-metallofullerene containing mixed rare-earth metal-metal bond and expands the understanding of metal-metal bonding of rare earth elements.

Firmness is one of the most important fruit quality traits in *** postharvest shelf life of this soft fruit is highly limited by the loss of firmness,where cell wall disassembly plays an important *** studies demonstrated that the polygalacturonase FaPG1 has a key role in remodelling pectins during strawberry *** this study,FaPG1 knockout strawberry plants have been generated using the CRISPR/cas9 system delivered via Agrobacterium *** independent lines,cv.“Chandler”,were obtained,and all of them were successfully edited as determined by PCR amplification and T7 endonuclease *** targeted mutagenesis insertion and deletion rates were analyzed using targeted deep *** percentage of edited sequences varied from 47%up to almost 100%,being higher than 95%for seven of the selected *** analyses showed that 7 out of the eight lines analyzed produced fruits significantly firmer than the control,ranging from 33 to 70%increase in *** was a positive relationship between the degree of FaPG1 editing and the rise in fruit *** changes were observed in other fruit quality traits,such as colour,soluble solids,titratable acidity or anthocyanin *** fruits showed a reduced softening rate during postharvest,displayed a reduced transpirational water loss,and were less damaged by Botrytis cinerea *** analysis of four potential off-target sites revealed no mutation *** conclusion,editing the FaPG1 gene using the CRISPR/cas9 system is an efficient method for improving strawberry fruit firmness and shelf life.

The impact of forest microhabitats on physiochemical properties of the soil and that of microbial communities on tropical soils remain poorly *** elucidate the effect of tropical forest stand on leaf litter and soil microbial communities,we studied enzyme activities,microbial biomass,and diversity in three distinct microhabitats in terms of plant richness,diameter at breast height(DBH),and physiochemical properties of soil and litter,each associated with a different Vanilla *** the soil,positive correlations were found between electrical conductivity(EC)and total organic carbon(TOC)with phosphatase activity,and between nitrogen(N)and water-soluble carbon(WSC)content with urease activity(UA).In the litter,the water content was positively correlated with bacterial and fungal biomass,and N and WSC contents were positively correlated with fungal *** correlations were found between plant richness and UA in the soil,plant richness and fungal biomass in the soil and litter,and DBH and fungal biomass in the *** sequencing revealed differences between microhabitats in the relative abundance of some fungal and bacterial taxa and in the bacterial community composition of both litter and *** richness and diversity were different between microhabitats,and,in litter samples,they were negatively correlated with DBH and plant richness,*** contrast,none of the soil and litter physiochemical properties were significantly correlated with microbial *** results show that significant shifts in enzyme activity,microbial biomass,and diversity in the microhabitats were driven by key abiotic and biotic factors depending on the soil or litter sample type.

AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-cas9 system and analyze phenotypic modifications as well as gene expression *** CRISPR-cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, PDE4 D, ABL1, ABL2, LPAR1, SMAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCLUSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

Among all-solid-state batteries, rechargeable Al-ion batteries have attracted most attention because they involve threeelectron-redox reactions with high theoretic specific capacity. However, the solid Al-ion conductor electrolytes are less studied. Here, the microscopic path of Al3+-ion conduction of NASICON-type(Al0.2Zr0.8)20/19Nb(PO4)3oxide is identified by temperature-dependent neutron powder diffraction and aberration-corrected scanning transmission electron microscopy experiments.(Al0.2Zr0.8)20/19Nb(PO4)3shows a rhombohedral structure consisting of a framework of(Zr,Nb)O6octahedra sharing corners with(PO4) tetrahedra; the Al occupy trigonal antiprisms exhibiting extremely large displacement factors. This suggests a strong displacement of Al ions along the c axis of the unit cell as they diffuse across the structure by a vacancy mechanism. Negative thermal expansion behavior is also identified along a and b axes, due to folding of the framework as temperature increases.
